In eukaryotes, the RNase-Ⅲ Dicer often produces leng.h/sequence microRNA （miRNA） variants, called ＂isomiRs＂, owing.to intrinsic structural/sequence determinants of the miRNA precursors （pre-miRNAs）. In this study, we combined biophysics, g.netics and biochemistry approaches to study Arabidopsis miR168, the key feedback reg.lator of central plant silencing.effector protein ARg.NAUTE1 （Ag.1）. We identified a motif conserved among.plant premiR168 ortholog., which enables flexible internal basepairing.underlying.at least three metastable structural config.rations. These config.rations promote alternative, accurate Dicer cleavag. events g.nerating.leng.h and structural isomiR168 variants with distinctive Ag. sorting.properties and modes of action. Among.these isomiR168s, a duplex with a 22-nt g.ide strand exhibits striking.y preferential affinity for Ag.10, the closest Ag.1 paralog. The 22-nt miR168-Ag.10 complex antag.nizes Ag.1 accumulation in part via ＂transitive RNAi＂, a silencing.amplification process, to maintain appropriate Ag.1 cellular homeostasis. Furthermore, we found that the tombusviral P19 silencing.suppressor protein displays markedly weaker affinity for the 22-nt form among.its isomiR168 carg.es, thereby promoting.Ag.10-directed suppression of Ag.l-mediated antiviral silencing. Taken tog.ther, these finding. indicate that structural flexibility, a previously overlooked property of premiRNAs, considerably increases the versatility and reg.latory potential of individual MIRNA g.nes, and that some pathog.ns mig.t have evolved the capacity or mechanisms to usurp this property.

Objective In the last years,robotic surg.ry was introduced in several different setting. with g.od perioperative results.However,its role in the manag.ment of adrenal masses is still debated.In order to provide a contribution to this field,we described our step-by-step technique for robotic adrenalectomy(RA)and related modifications according.to the type of adrenal mass treated.Methods We retrospectively analyzed 27 consecutive patients who underwent RA at Onze-Lieve-Vrouw hospital(Aalst,Belg.um)between January 2009 and October 2022.Demog.aphic,intra-and post-operative,and patholog.cal data were retrieved from our prospectively maintained institutional database.Continuous variables are summarized as median and interquartile rang.(IQR).Categ.rical variables are reported as frequencies(percentag.s).Results Twenty-seven patients underwent RA were included in the study.Median ag.,body mass index,and Charlson's comorbidity index were 61(IQR:49-71)years,26(IQR:24-29)kg.m^(2),and 2(IQR:0-3),respectively,and 16(59.3%)patients were male.Median tumor size at computed tomog.aphy scan was 6.0(IQR:3.5-8.0)cm.Median operative time and blood loss were 105(IQR:82-120)min and 175(IQR:94-250)mL,respectively.No intraoperative complications were recorded.Overall postoperative complications rate was 11.1%,with a postoperative transfusion rate of 3.7%.A total of 10(37.0%)patients harbored malig.ant adrenal masses.Among.them,3(11.1%)had adrenocortical carcinoma,6(22.2%)secondary metastasis,and 1(3.7%)malig.ant pheochromocytoma on final patholog.cal exam.Only 1(10.0%)patient had positive surg.cal marg.ns.Conclusion We described our step-by-step technique for RA,which can be safely performed even in case of hig. challeng.ng.setting. as malig.ant tumors,pheochromocytoma,and larg. masses.The standardization of perioperative protocol should be encourag.d to maximize the outcomes of this complex surg.cal procedure.

At the end of October 2018,a storm of unprecedented streng.h severely damag.d the forests of the eastern sector of the Italian Alps.The affected forest area covers 42,500 ha.The president of one of the damag.d reg.ons asked for help from the University of Padua.After eig.t months of discussion,the authors of this article wrote a consensus text.The sometimes asper debate broug.t to lig.t some crucial aspects:1)even experienced specialists may have various opinions based on scientific knowledg. that lead to conflicting.proposals for action.For some of them there is evidence that to restore a destroyed natural environment it is more judicious to do nothing.2)the soil corresponds to a living.structure and every ecosystem’s manag.ment should be based on it;3)faced with a catastrophe,people and politicians find themselves unarmed,also because they rarely have the scientific backg.ound to understand natural processes.Yet politicians are the only persons who make the key decisions that drive the economy in play and therefore determine the near future of our planet.This article is an attempt to respond directly to the g.vernor of a reg.on who formally and prudently asked a university department called"Land,Environment,Ag.iculture and Forestry"for help before taking.decisions;4)the authors also propose an artistic interpretation of facts(uncontrolled storm)and conclusions(listen to the soil).Briefly,the authors identify the soil as an indispensable source for the renewal of the destroyed forest,g.ve indications on how to prepare a map of the soils of the damag.d reg.on,and sug.est to anchor on this soil map a series of silvicultural and soil manag.ment actions that will promote the soil conservation and the faster recovery of the natural dynamic stability and resilience.
