Proteins typically fold in a way to have their hydrophobic residues buried in the core of proteins while the hydrophilic residues exposed on protein surface. The exposure of large hydrophobic patch to solvent is thermodynamically unfavorable, and is not commonly observed. Cell surface receptor urokinase-type plasminogen activator receptor（uPAR） has a large hydrophobic patch in its central ligand binding pocket. Upon the ligand（uPA） binding, this hydrophobic patch on uPAR is sequestered, forming a highly stable and tight-binding complex（Qing Huai, 2006）. The crystallization of uPAR ectodomains in its ligand-free conformation is expected to be difficult, but has been a long sought goal. With the help of an anti-uPAR antibody（8b12）, we recently succeed to crystallize the uPAR:8b12 complex in the absence of any ligands, and determined the crystal structure of uPAR in the ligand-free conformation. Surprisingly, uPAR adopts a conformation similar to its ligand-bound state, exposing a large hydrophobic area to solvent. This observation is different from the previous speculation and biochemical studies（Cai Yuan, 2007 and Haydyn D. T. Mertens, 2012）. uPAR is a glycolipid-anchored cell surface receptor and consists of three Ly6/uPAR/α-neurotoxin-like（LU） domains. Extensive degree of inter-domain flexibility was expected and was indeed observed in solution by biochemical studies including small angle X-ray scattering. However, Our recent uPAR:8b12 structure showed no inter-domain flexibility and the conformation of unliganded uPAR is identical to the ligand-bound conformation. Further functional studies are underway to examine the functional consequences of these structural observations.

通过多年的调查资料、观鸟爱好者所提供的图片和文献整理,我们对多杜鹃寄生系统下的中国寄生性杜鹃及其宿主的多样性进行了系统研究。在中国分布的17种寄生性杜鹃中,迄今已发现11种杜鹃的宿主达55种,均为雀形目鸟类,隶属于15科,其中莺科、鸫科和画眉科分别占到寄生记录总数的 22.6%、20.8% 和 17.0%。大杜鹃(Cuculus canorus)的宿主种类最多(45.5%),是中国最常见、宿主多样性最丰富的寄生性杜鹃。不同种类的寄生性杜鹃的卵色不同,而且杜鹃与其宿主之间均呈现明显的卵色多态性,为典型的卵色协同进化。基于人眼观察,大杜鹃、小杜鹃(C. poliocephalus)和八声杜鹃(Cacomantis merulinus)的卵色均与其宿主高度模拟,而鹰鹃(Cuculus sparverioides)、中杜鹃(C. saturatus)和翠金鹃(Chrysococcyx maculatus)则产非模拟的卵。目前仍有6种寄生性杜鹃的宿主不详,亟待加强研究。

鸟类巢寄生是研究协同进化的模式系统。寄生性繁殖的鸟类进化出非常独特、且相对要厚很多的卵壳。关于厚卵壳的进化,主要有以下假说:1)防止产卵损坏假说,即寄生者在巢上将卵产至宿主的巢中时避免损坏;2)防止宿主啄破假说,即厚卵壳可以有效防止宿主对寄生卵的啄破,使其无法扔掉寄生卵;3)雏鸟生长能量假说,即雏鸟具有排他性,由于其较大的身体肌肉骨骼系统发育需要大量钙质,导致卵壳的钙富集而变得较厚;4)抗菌保护假说,即较厚的卵壳有利于保护寄生者的胚胎免于被宿主巢内的微生物所侵害。本文综述了目前关于寄生性繁殖鸟类的厚卵壳进化机制的研究,认为这些假说之间并非相互排斥,但都缺乏足够的实验证据,尽管防止产卵损坏假说和防止宿主啄破假说得到了较多的支持。对于防止宿主啄破假说,如何去量化宿主对寄生卵的拒绝方式如口吞式扔卵、啄破式扔卵和弃巢等,均直接影响到该假说的检验。对此,我们给出了具体的研究建议。

Design and fabrication of computer-generated holograms for testing optical freeform surfaces

Freeform optical surfaces （foss） will be the best elements in the design of compact optical systems in the future. However, it is extremely difficult to measure freeform surface with sufficient accuracy, which im- pedes the development of the freeform surface. The design and fabrication of computer-generated hologram （CGH） , which has been successfully applied to the tests for aspheric surfaces, cannot be directly adopted to test foss due to their non-rotational asymmetry. A novel ray tracing planning method combined with successively optimizing even and odd power coefficients of phase polynomials in turn is proposed, which can successfully design a non-rotational asymmetry CGH for the tests of foss with an F-O lens. A new eight-step fabrication process is also presented aiming to solve the problem that the linewidth on the same circle of the CGH for testing freeform surface is not uniform. This problem cannot be solved in the original procedure of CGH fabrication. The test results of the step profiler show that the CGH fabricated in the new nrocedure meets the reauirements.

在北极气候影响评估(ACIA)中,一项气候及紫外线b辐射变化对北极陆地生态系统影响的评估着重强调了预期中的变暖的深远意义,尤其是变暖在未来生态系统功能、生物多样性,以及对气候的反馈方面的深远意义。然而,尽管在有些地理区域和有些学科,我们目前对气候和紫外线b辐射驱动下生态过程及生态变化的了解已算充分,但在其他区域及学科,这种了解是微弱的。即使随着北极地区研究成果的积累和新技术的引用,近来我们的预测能力已空前提高,我们目前的了解程度也还受到各种各样的不确定性的限制。这项评估是在每一种都含有不确定性的一系列方式方法,以及常常远非完整的数据集的基础上完成的。不确定性从各种方法和概念框架而来,从不可预测的意外事件、从对模型的缺乏验证、从未来温室气体排放及气候变化的一些特别情景的应用而不是预测而来。此项评估中,以减少不确定性为目的的建议比比皆是,而且涉及所有学科。然而,一再出现的主题是:在北极这样人烟稀少的偏远地区,环境变化及其影响的实验、观察及监测活动达到足够的空间广度和时间长度是极其重要的。

长期以来,就营养物质和碳循环而言,北极生态系统降低了初级生产力;能量,水和温室气体交换的水平已引起了局部和区域性的小幅度降温。大气CO_2中的碳沉积在广袤而寒冷的有机土壤中,冰雪覆盖的低矮植被产生高的反射率,都影响了局部气候。然而,北极生态系统功能的许多方面都对气候变化及其产生的生物多样性影响敏感。当前的北极气候导致了低的有机物质分解速率,因此,尽管有机物和元素输入量较低,但北极生态系统还是趋向于积累有机物和元素,土壤中氮和磷等可利用元素结果成为促进碳固定以及生物量和有机物进一步积累的关键性限制因素。气候变暖可能增加特别是土壤中的碳和元素的周转,起初可能导致元素的丢失,但最后会慢慢的恢复。在北极生态系统中,单个物种和物种多样性已经明显地影响了元素的输入和滞留,另一方面,从长远来看,尽管CO_2和紫外线增加对植物组织化学、分解和氮固定的影响可能变得重要,但对整个生态系统来说, 影响可能很小。碳循环的示踪气体主要形式是CO_2和CH_4,大多数碳以CO_2的形式损失,这些CO_2是由植物和土壤生物产生。来自潮湿苔原生态系统以CH_4形式释放的碳大约是CO_2形式的5%,而且在没有任何其他变化的情况下,对变暖作出响应。冬天过程和植物类型也影响CH_4释放和能量在生物圈和大气之间的交换,因为反射率从冬末到夏天存在很大的变化,在冬末,雪反射了入射的大部分光线,在夏天,生态系统吸收了入射的大部分光线,所以在所有的陆地生态系统中,北极生态系统在能量交换方面表现出巨大的季节性变化。植被深刻地影响北极生态系统水和能量交换。在冰雪覆盖期间,反射率从苔原、森林苔原、落叶林、常绿林依次降低。灌木和树增加了雪的深度,反过来又使冬天的土壤温度增加,因此,由气候变化而引起的未来植被方面的变化很可能深远地改变区域的气候。
