Biomolecular analysis at the single-cell level is increasingly important in the study of cellular heterogeneity and its consequences,particularly in organismic development and complex diseases such as ***-cell molecular analyses have led to the identification of new cell types1 and the discovery of novel targets for diagnosis and *** these analyses are performed predominantly on dissociated single cells,emerging techniques seek understanding of cellular state,cellular function and cell–cell interactions within the native tissue environment by combining optical microscopy and single-cell molecular *** techniques include in situ multiplexed imaging of fluorescently labeled proteins and nucleotides,as well as low-throughput ex vivo methods in which specific cells are isolated for downstream molecular ***,these methods are limited in either the number and type of molecular species they can identify or the number of cells that can be ***-throughput methods are needed for comprehensive profiling of many cells(>1000)to detect rare cell types,discriminate relevant biomarkers from intrinsic population noise,and reduce the time and cost of *** established,highthroughput single-cell analyses are not directly applicable because they require tissue dissociation,leading to a loss of spatial *** current methods exist that can seamlessly connect spatial mapping to single-cell *** this Perspective,we review current methods for spatially resolved single-cell analysis and discuss the prospect of novel multiplexed imaging probes,called laser particles,which allow individual cells to be tagged in tissue and analyzed subsequently using high-throughput,comprehensive single-cell techniques.

The ability to track individual cells in space over time is crucial to analyzing heterogeneous cell ***,microlaser particles have emerged as unique optical probes for massively multiplexed single-cell ***,the microlaser far-field emission is inherently direction-dependent,which causes strong intensity fluctuations when the orientation of the particle varies randomly inside ***,we demonstrate a general solution based on the incorporation of nanoscale light scatterers into *** schemes are developed by introducing either boundary defects or a scattering layer into microdisk *** resulting laser output is omnidirectional,with the minimum-to-maximum ratio of the angle-dependent intensity improving from 0.007(-24 dB)to>0.23(-6dB).After transfer into live cells in vitro,the omnidirectional laser particles within moving cells could be tracked continuously with high signal-to-noise ratios for 2 h,while conventional microlasers exhibited frequent signal loss causing tracking failure.

Protective effect of resveratrol against cadmium-induced toxicity on ovine oocyte in vitro maturation and fertilization

Background:Heavy metal cadmium(Cd)is a widespread environmental contaminant with a potential toxicity that might negatively affect female reproduction and *** has been reported that Cd exposure impaired the quality of oocytes and led to a defective maturation and fertilization,through oxidative stress ***(Res)is a natural polyphenol with strong antioxidant properties that exhibited protective role in preventing oocyte redox homeostasis disruption and quality ***,we explored whether the addition of Res to in vitro maturation(IVM)medium might act as a protection against Cd-induced toxicity on ovine oocyte maturation and ***,we evaluated the effect of supplementing IVM medium with two different Res concentrations(1and 2μmol/L)on nuclear maturation and fertilization of oocytes matured under CdCl2(2μmol/L)***,the concentration of 1μmol/L Res was selected to analyse the effects of this compound on intracellular ROS levels,mitochondrial(mt)distribution and activity,chromatin configuration,cytoskeleton morphology,cortical granules(CGs)distribution and mRNA expression of genes associated with cellular response to oxidative stress(***1,SOD 1,GPX1,GSR,CAT)in Cd-exposed in vitro matured ***:We found that 1μmol/L Res restored the reduced oocyte meiotic competence induced by Cd exposure as well as,Res sustained oocyte ability to be normally fertilized and decreased polyspermic fertilization at both tested ***,we demonstrated that 1μmol/L Res mitigated Cd-induced alterations of oocyte cytoplasmic maturation by reducing reactive oxygen species(ROS)accumulation,preventing mt dysfunction,maintaining the correct meiotic spindle and cortical F-actin assembly and the normal cortical granule distribution as well as up-regulating SIRT1,SOD1 and GPX1 ***:Taken together,our findings highlighted the beneficial influence exerted by Res in preventing Cdinduced disturbance of

Anatomical and functional changes after dexamethasone implant and ranibizumab in diabetic macular edema: a retrospective cohort study

AIM: To investigate the efficacy and safety of ranibizumab(RZB group) and dexamethasone implant(DEX group) intravitreal treatments in patients with treatment-na?ve center involved diabetic macular edema(DME) by means of functional and morphological ***: This retrospective cohort study included 50 eyes of 50 patients with DME treated either with RBZ or DEX. Best-corrected visual acuity(BCVA) and microperimetry were evaluated at baseline and during a 6-month follow-up. In addition, central macular thickness(CMT) by means of structural optical coherence tomography(OCT) and retinal capillary plexus density and choriocapillary density by means of OCT angiography were assessed in all ***: Functional and morphological parameters significantly improved during the study period in both groups. BCVA improved significantly in both groups witha greater increase in the DEX group compared to the RBZ group(P=0.030). Microperimetry significantly differed during follow-up between the two treatments(P=0.031). In both groups CMT significantly decreased(P<0.001) without statistically significant differences between the two groups. A statistically significant increase of deep capillary plexus density was detected in both groups at 30 d after therapy. The retreatment rate was 0.70±0.10 and 0.65±0.10 in the RBZ group and 0.65±0.10 and 0.50±0.11 in DEX group at 120 and 180 d respectively. Two out of 25 patients in DEX group showed intraocular pressure increase requiring hypotonic eye ***: Both treatments are very effective for DME treatment during 6 mo of follow-up with a lower retreatment rate in DEX group.

BACKGROUND Laminopathies are rare diseases,whose cardiac manifestations are heterogeneous and,especially in their initial stage,similar to those of more common conditions,such as ischemic heart *** diagnosis is essential,as these conditions can first manifest themselves with sudden cardiac *** complications usually appear before structural complications;therefore,it is important to take into consideration these rare genetic disorders for the differential diagnosis of brady and tachyarrhythmias,even when left ventricle systolic function is still *** SUMMARY A 60-year-old man,without history of previous disorders,presented in September 2019 to the emergency department because of the onset of syncope associated with *** patient was diagnosed with a high-grade atrioventricular block.A dual chamber pacemaker was implanted,but after the onset of a sustained ventricular tachycardia during physical exertion,a drug eluting stent was implanted on an intermediate stenosis on the left anterior descending artery,which had previously been considered non-haemodynamically *** the follow-up,the treating cardiologist,suspicious of the overall clinical picture,recommended a genetic test for the diagnosis of cardiomyopathies,which tested positive for a pathogenetic mutation of the lamin A/C *** awaiting the result of the genetic test and,later,the pacemaker to be upgraded to a biventricular defibrillator,a remote monitoring device was given to the patient in 2019-related *** case aims to raise awareness of the cardiological manifestations of laminopathies,which can be dangerously misdiagnosed as other, more common conditions.

The number of bariatric operations,as well as the incidence of perioperative complications,has risen sharply in the past ten *** acute portal vein thrombosis is an infrequent and potentially severe postoperative complication that has not yet been reported after biliopancreatic diversion(BPD).Three cases are presented of portal vein thrombosis that occurred following BPD treatment for morbid obesity and type 2 *** thromboses were detectedby abdominal ultrasound and computed tomography with intravenous *** portomesenteric venous thromboses in all three cases presented as unexpected abdominal pain several days after discharge from the *** complications occurred despite adequate perioperative prophylaxis and progressed to bowel gangrene in the diabetic patients *** cases demonstrate the occurrence of this rare type of complication,which may be observed by physicians that do not routinely treat bariatric *** of this surgical complication will allow for early diagnosis and prompt initiation of adequate therapy.

Background:To evaluate the effectiveness and safety of cabazitaxel in castration-resistant prostate cancer patients aged>80 years,we performed a retrospective study on a sample of patients from 11 Italian cancer *** and methods:Fifty-seven patients aged>80 years were treated with cabazitaxel after previous failure with docetaxel;39 completed a comprehensive geriatric assessment questionnaire(34 fit and 5 vulnerable)and 8 patients(14%)had an Eastern Cooperative Oncology Group performance status(PS)>2,while most had a PS of 0-1(86%).Cabazitaxel was administered at a dose of 25 mg/m^(2)in 30(52%)patients and 20 mg/m^(2)or adapted schedules in 27(48%)*** schedules were adopted mainly in patients>85 years(75%),with a PS>2(87.5%),and those classified as vulnerable(100%).Results:The duration of treatment was 4.8 months and was comparable in all subgroups;disease control rate was reported in 36 patients(63%);prostate-specific antigen response was recorded in 18 patients(31.5%).Median overall survival was 13.1 months regardless of age(85 years),but overall survival was reduced in vulnerable(7.2 months)and PS>2 patients(6.8 months).The most frequently documented grade 3-4 toxicities were neutropenia(14%)and diarrhea(10.5%).Six patients(10.5%)dropped out due to severe ***:Octogenarian patients can be treated with cabazitaxel with reduced doses or alternative schedules that are associated with less toxicity and fewer treatment *** geriatric assessment could facilitate more appropriate patient selection.
