The combination of hydrogen/deuterium(H/D)formaldehyde-based isotopic methyl labeling with solid-phase extraction and high-performance liquid chromatography–high resolution mass spectrometry(HPLC-HRMS)is a powerful analytical solution for nontargeted analysis of trace-level amino-containing chemicals in water *** the huge amount of chemical information generated in HPLC-HRMS analysis,identifying all possible H/Dlabeled amino chemicals presents a significant challenge in data *** address this,we designed a streamlined data processing pipeline that can automatically extract H/D-labeled amino chemicals from the raw HPLC-HRMS data with high accuracy and ***,we developed a cross-correlation algorithm to correct the retention time shift resulting from deuterium isotopic effects,which enables reliable pairing of H-and D-labeled ***,we implemented several bioinformatic solutions to remove false chemical features generated by in-source fragmentation,salt adduction,and natural13C ***,we used a data mining strategy to construct the AMINES library that consists of over 38,000 structure-disjointed primary and secondary amines to facilitate putative compound ***,we integrated these modules into a freely available R program,*** rationale of each module was justified and its performance tested using experimental H/D-labeled chemical standards and authentic water *** further demonstrated the application of HDPairFinder to effectively extract N-containing contaminants,thus enabling the monitoring of changes of primary and secondary N-compounds in authentic water *** is a reliable bioinformatic tool for rapid processing of H/D isotopic methyl labeling-based nontargeted analysis of water samples,and will facilitate a better understanding of N-containing chemical compounds in water.

In Canada,the periodic circulation of west Nile Virus(wNV)is difficult to predict and,beyond climatic factors,appears to be related to the migratory movements of infected birds from the southern United *** hypothesis has not yet been explored in a spatially distributed *** main objective of this work was to develop a spatially explicit dynamic model for the transmission of wNV in Canada,that allows us to explore non-climate related hypotheses associated with wNV transmission.A Cellular Automata(CA)approach for multiple hosts(birds and humans)is used for a test region in eastern Ontario,*** tool is designed to explore the role of host and vector spatial heterogeneity,host migration,and vector feeding *** developed a spatialized compartmental SEIRDS-SEI model for wNV transmission with a study region divided into 4 rectangular *** used 2010–2021 bird data from the ebird project and 2010–2019 mosquito data collected by Ontario Public Health to mimic bird and mosquito seasonal *** considered heterogeneous bird densities(high and low suitability areas)and homogeneous mosquito and human *** high suitability areas for birds,we identified 5 entry points for wNV-infected *** compared our simulations with pools of wNV-infected field collected *** and sensitivity analyses were performed using MATLAb *** results showed good correspondence between simulated and observed epidemics,supporting the validity of our model assumptions and *** analysis showed that a 5%increase or decrease in each parameter of our model except for the biting rate of bird by mosquito(c^(b,M)and mosquito natural mortality rate(d^(M)),had a very limited effect on the total number of cases(newly infected birds and humans),prevalence peak,or date of *** demonstrate the utility of the CA approach for studying wNV transmission in a heterogeneous landscape with multiple hosts.

Given a compact and regular Hausdorff measure space (X, μ), with μ a Radon measure, it is known that the generalised space M(X) of all the positive Radon measures on X is isomorphic to the space of essentially bounded functions L∞(X, μ) on X. we confirm that the commutative von Neumann algebras M⊂b(H), with H=L2(X, μ), are unitary equivariant to the maximal ideals of the commutative algebra C(X). Subsequenly, we use the measure groupoid to formulate the algebraic and topological structures of the commutative algebra C(X) following its action on M(X) and define its representation and ergodic dynamical system on the commutative von Neumann algebras of M of b(H) .

Objective:Myocardial infarction(MI)is linked to an imbalance in the supply and demand of blood oxygen in the heart ***-blockers and calcium antagonists are just two of the common medications used to treat ***,these have reportedly been shown to be either ineffective or to have undesirable side *** of Ginkgo biloba leaves(GbE),a Chinese herbal product offers special compatibility benefits in therapeutic settings relating to inflammatory diseases and oxidative *** order to better understand how GbE affects MI in rats insulted by isoprenaline(ISO),the current study was ***:The heart weight index,serum lipid profile,cardiac marker enzymes,endogenous antioxidants[catalase(CAT),superoxide dismutase(SOD),glutathione(GSH),nitrites and malondialdehyde(MDA)],inflammatory mediators[tumour necrosis factor alpha(TNF-a)and interleukin-6(IL-6)],immunohistochemical expressions of b-cell lymphoma factor-2(bcl-2),extracellular signal-regulated kinase(ERK1/2),and mammalian target of rapamycin(mTOR)and histopathological analysis were used to assess the cardioprotective properties of ***:The findings showed that GbE effectively attenuated myocardial infarction by boosting the body’s natural antioxidant defense system and reducing the release of inflammatory cytokines as well as heart injury marker *** expression of bcl-2,ERK1/2 and mTOR was increased while the histomorphological alterations were ***:The cardioprotective effects of GbE may be due to a mechanism involving increased bcl-2/mTOR/ERK1/2/Na^(+),K^(+)-ATPase activity.

Cirrhotic portal hypertension: From pathophysiology to novel therapeutics

Portal hypertension and bleeding from gastroesophageal varices is the major cause of morbidity and mortality in patients with cirrhosis. Portal hypertension is initiated by increased intrahepatic vascular resistance and a hyperdynamic circulatory state. The latter is characterized by a high cardiac output, increased total blood volume and splanchnic vasodilatation, resulting in increased mesenteric blood flow. Pharmacological manipulation of cirrhotic portal hypertension targets both the splanchnic and hepatic vascular beds. Drugs such as angiotensin converting enzyme inhibitors and angiotensin Ⅱ type receptor 1 blockers, which target the components of the classical renin angiotensin system(RAS), are expected to reduce intrahepatic vascular tone by reducing extracellular matrix deposition and vasoactivity of contractile cells and thereby improve portal hypertension. However, these drugs have been shown to produce significant offtarget effects such as systemic hypotension and renal failure. Therefore, the current pharmacological mainstay in clinical practice to prevent variceal bleeding and improving patient survival by reducing portal pressure is non-selective-blockers(NSbbs). These NSbbs work by reducing cardiac output and splanchnic vasodilatation but most patients do not achieve an optimal therapeutic response and a significant proportion of patients are unable to tolerate these *** statins, used alone or in combination with NSbbs, have been shown to improve portal pressure and overall mortality in cirrhotic patients, further randomized clinical trials are warranted involving larger patient populations with clear clinical end points. On the other hand, recent findings from studies that have investigated the potential use of the blockers of the components of the alternate RAS provided compelling evidence that could lead to the development of drugs targeting the splanchnic vascular bed to inhibit splanchnic vasodilatation in portal hypertension. Plasticity in the antipredator behavior of the orange-footed sea cucumber under shifting hydrodynamic forces

Marine invertebrates that move too slowly to evade unfavorable environmental change may instead exhibit phenotypic plasticity,allowing them to adjust to varying *** orange-footed sea cucumber Cucumaria frondosa is a slow-moving suspension feeder that is preyed on by the purple sunstar Solaster *** sea cucumber's antipredator behavior involves changing shape and detaching from the substratum,which might increase its probability of being displaced by water motion into an unsuitable *** hypothesized that sea cucumbers'antipredator responses would be diminished under stronger hydrodynamic forces,and that behavioral strategies would be flexible so that individuals could adjust to frequent changes in water *** a natural orange-footed sea cucumber habitat,individuals lived along a pronounced hydrodynamic gradient,allowing us to measure antipredator behavior under different water flow *** placed purple sunstars in physical contact with sea cucumbers living at various points along the gradient to elicit antipredator *** then repeated this procedure in a laboratory mesocosm that generated weak and strong hydrodynamic forces similar to those observed at the field *** in the mesocosm experiment were tested in both wave conditions to determine if their antipredator behavior would change in response to sudden environmental change,as would be experienced under deteriorating sea *** responses did not covary with hydrodynamic forces in the ***,antipredator responses in the mesocosm experiment increased when individuals were transplanted from strong to weak forces and decreased when transplanted from weak to strong ***,our results indicate environmentally induced plasticity in the antipredator behavior of the orange-footed sea cucumber.

Mass data processing of time series Landsat imagery:pixels to data products for forest monitoring

Free and open access to the Landsat archive has enabled the implementation of national and global terrestrial monitoring ***,we summarize a project characterizing the change history of Canada’s forested ecosystems with a time series of data representing *** the Composite2Change approach,we applied spectral trend analysis to annual best-available-pixel(bAP)surface reflectance image composites produced from Landsat TM and ETM+imagery.A total of 73,544 images were used to produce 29 annual image composites,generating∼400 Tb of interim data products and resulting in∼25 Tb of annual gap-free reflectance composites and change *** average,10%of pixels in the annual bAP composites were missing data,with 86%of pixels having data gaps in two consecutive years or *** detection overall accuracy was 89%.Change attribution overall accuracy was 92%,with higher accuracy for standreplacing wildfire and *** were assigned to the correct year with an accuracy of 89%.Outcomes of this project provide baseline information and nationally consistent data source to quantify and characterize changes in forested *** methods applied and lessons learned build confidence in the products generated and empower others to develop or refine similar satellite-based monitoring projects.
