Overactivation of histone deacetylases and EZH2 in Wilms tumorigenesis

Wilms tumor (WT) is the most common childhood kidney cancer. Although WT is largely curable, current treatments fail in up to 15 percent of patients. Moreover, survivors suffer from the complications and late effects of the aggressive treatments. Thus, there is a critical need to improve our understanding of tumorigenesis to develop novel therapies to reduce the treatment burden while maintaining excellent survival rates. WT is believed to arise from the immature kidney cells, nephron progenitor cells (NPCs), which have failed to differentiate properly. Previous studies revealed that Wilms cells share a transcriptional and epigenetic landscape with normal renal stem cells.1 Although several studies have shown the positive associations between WT in children and embryonic exposure to adverse environments, the underlying mechanisms remain unknown. Altered epigenetics is central to oncogenesis in many pediatric cancers. The critical contribution of epigenetic dysregulation to pediatric tumors provides a compelling rationale for the therapeutic potential of epigenetic drugs. Histone deacetylases (HDACs) and Enhancer of Zeste Homolog 2 (EZH2, a histone H3K27 methyltransferase), have been demonstrated to play a critical role in self-renewal and differentiation of mouse NPCs.2,3 In addition, altered expression and mutations of HDACs and EZH2 have been linked to many human cancers, including WT. Thus, they are among the most promising therapeutic targets for cancer treatment. We reasoned that WT would result from the unrestrained proliferation of progenitor cells due to overactive HDAC1/2 (HDAC1 and HDAC2), and EZH2. We tested this hypothesis by analyzing a clinical specimen received from the left kidney tumor of an 11-year-old male patient diagnosed with WT and four other human WT specimens. A Correlation Coefficient Approach for Evaluation of Stiffness Degradation of Beams Under Moving Load

This paper presents a new approach using correlation and cross-correlation coefficients to evaluate the stiffness degradation of beams under moving *** theoretical study of identifying defects by vibration methods showed that the traditional methods derived from the vibration measurement data have not met the needs of the actual *** show that the correlation coefficients allow us to evaluate the degree and the effectiveness of the defects on *** the same time,the cross-correlation model is the basis for determining the relative position of *** results of this study are experimentally conducted to confirm the relationship between the correlation coefficients and the existence of the *** particular,the manuscript shows that the sensitivity of the correlation coefficients and cross-correlation is much higher than the parameters such as changes in stiffness(EJ)and natural frequency values(Δf).This study suggests using the above parameters to evaluate the stiffness degradation of beams by vibration measurement data in practice.

Rainfall data with an appropriate spatial resolution is a key input to hydrological ***,networks of rain gauges are often sparsely and unevenly distributed in large catchments,especially in developing ***-resolution rainfall datasets,such as the Asian Precipitation Highly Resolved Observational Data Integration Towards Evaluation of Water Resources(APHRODITE),the Climate Forecast System Reanalysis(CFSR),the Climatic Research Unit Time Series(CRUTS),the Global Precipitation Climatology Centre(GPCC)and the Tropical Rainfall Measuring Mission(TRMM),have become available to overcome such *** objective of this study was to evaluate the impacts of four land-based rainfall products(APHRODITE,CFSR,CRU-TS,and GPCC)and a satellite-based rainfall product(TRMM)on streamflow of the upper catchment of Tri An reservoir in Vietnam using the Hydrological Modeling System(HEC-HMS).In addition,the available rain gauges data were used for comparison *** indicates that the TRMM and GPCC data show their best match to rain gauges data in simulating the streamflow in the period 1999–***,the results indicate that the TRMM and GPCC data could be alternative solutions.

Small cell lung cancer(SCLC)is a common type of primary lung cancer that contributes to approximately 15%of *** is closely associated with tobacco risk *** is also known as a type of lung cancer that has a high mortality rate within a short time due to its rapid growth rate(with tumor doubling time of 30 days)and its tendency to metastasize early in the disease *** primary sites of metastasis in SCLC are similar to those in other primary lung cancers and often include the brain,bones,adrenal glands,liver,and lymph ***,there are a few clinical reports of uncommon metastases,such as gastric metastasis from *** the incidence of this clinical presentation is very low,reported cases have generally resulted in early mortality due to inadequate *** purpose of this letter is to discuss the knowledge related to gastric metastasis from SCLC and remind clinical doctors not to miss atypical symptoms,thereby providing the right attitude to improve the prognosis for these patients.

Push forward LC-MS-based therapeutic drug monitoring and pharmacometabolomics for anti-tuberculosis precision dosing and comprehensive clinical management

A new species of the genus Leptolalax (Anura： Megophryidae) from southern Vietnam

We describe a new species of megophryid frog from Phu yen Province in southern Vietnam. Leptolalax macrops sp. nov. is distinguished from its congeners by a combination of the following morphological attributes：（1） body size medium （SVL 28.0-29.3 mm in three adult males, 30.3 mm in single adult female）; （2） supra-axillary glands present, creamy white; ventrolateral glands indistinct; （3） tympanum externally distinct; （4） dorsal skin roughly granular with larger tubercles, dermal ridges on dorsum absent; （5） rudimentary webbing present between fingers I-II and I1-111; rudimentary webbing between all toes; fingers and toes without dermal fringes; （6） in life ventral surface greyish-violet with white speckling; （7） supratympanic fold distinct, dark brown in life; （8） iris bicolored, typically golden in upper half, fading to golden green in lower half; （9） tibia short （TbL/SVL 0.44-0.45 in males）; and （10） eyes large and protuberant （ED/SVL 0.15-0.16 in males） From all congeners for which comparable sequences are available, the new species differs markedly in the 16S rRNA mitochondrial gene sequence （P-distance〉5.7%）. The new species is currently known only from montane evergreen tropical forests of Song Hinh District, Phu yen Province, and M＇Drak District of Dak Lak Province at elevations of 470-630 m a.s.l.. We suggest the new species should be considered as Data Deficient following the IUCN＇s Red List categories. We also report a previously unknownLeptolalax mtDNA lineage from an evergreen tropica forest in the Hoa Thinh District of Phu yen Province which may also represent an undescribed species.
