Increased virulence of the oral microbiome in oral squamous cell carcinoma revealed by metatranscriptome analyses

oral squamous cell carcinoma(oSCC) is the most prevalent and most commonly studied oral cancer. However, there is a void regarding the role that the oral microbiome may play in oSCC. Although the relationship between microbial community composition and oSCC has been thoroughly investigated, microbial profiles of the human microbiome in cancer are understudied. Here we performed a small pilot study of community-wide metatranscriptome analysis to profile mRNA expression in the entire oral microbiome in oSCC to reveal molecular functions associated with this disease. Fusobacteria showed a statistically significantly higher number of transcripts at tumour sites and tumour-adjacent sites of cancer patients compared to the healthy controls analysed. Regardless of the community composition, specific metabolic signatures were consistently found in disease. Activities such as iron ion transport, tryptophanase activity, peptidase activities and superoxide dismutase were over-represented in tumour and tumour-adjacent samples when compared to the healthy controls. The expression of putative virulence factors in the oral communities associated with oSCC showed that activities related to capsule biosynthesis, flagellum synthesis and assembly, chemotaxis, iron transport, haemolysins and adhesins were upregulated at tumour sites. Moreover, activities associated with protection against reactive nitrogen intermediates, chemotaxis, flagellar and capsule biosynthesis were also upregulated in non-tumour sites of cancer patients. Although they are preliminary, our results further suggest that Fusobacteria may be the leading phylogenetic group responsible for the increase in expression of virulence factors in the oral microbiome of oSCC patients.

Thomas S. Kuhn is one of the leading philosophers and historians of science that investigated in-depth cases of simultaneous discoveries in science. Although his analysis of the discovery of energy conservation and oxygen did not focus sharply on the priority disputes involved, it is within such contexts that controversy about which scientist was the first to make a discovery takes place. Evidently, Kuhn’s recourse to historical case studies is a clear departure from the standpoint of traditional mainstream philosophies of science (namely, logical positivism and falsificationism), which cavalierly dismissed such concerns as irrelevant to philosophical reconstructions of science. Challenges to orthodox logistic approaches were prompted by the realisation that the two dominant traditions mentioned above, in their excessive preoccupation with "the logical skeleton of science", have lost contact with real science. As a contribution to what Michael Polanyi referred to as post-critical philosophy, the present study reanalyses the tension-generating potentials of bipolar values shared by members of scientific communities. It traces the origins of the rebellion against logic-dominated philosophies of science, and identifies different post-positivist approaches that have emerged over the years which legitimise broadening the frontiers of the philosophy of science. Consequent upon that, some conflicting values or norms shared by members of scientific communities and how they affect the quest for scientific knowledge are underscored. Using as a case study the acrimonious priority dispute between Isaac newton and Gottfried Leibniz concerning the discovery of calculus, the paper demonstrates that excessive concern for recognition which sometimes leads to protracted priority disputes tends to bring out the worst kind of behaviours towards colleagues even from the greatest scientists. The motion of incompressible fluid of a variable fluid viscosity and variable thermal conductivity with thermal radiation, Dufour, Soret with heat and mass transfer over a linearly moving porous vertical semi-infinite plate with suction is investigated. The governing equations are transformed into a system of coupled nonlinear ordinary differential equations using similarity transformations with dimensionless variables and solved numerically using shooting method with Runge-Kutta fourth-order method and newton-Raphson’s interpolation scheme implemented in MATLAB. The result showed that with increase in Dufour and Soret parameter, fluid velocity increases and temperature increases with increase in variation of Dufour while, temperature decreases with increase in Soret. The effects of variable fluid viscosity, variable thermal conductivity, thermal radiation, Soret, Dufour, Prandtl and Schmidt parameters on the dimensionless velocity, temperature and concentration profiles are shown graphically.

A new family of numerical integration formula is presented, which uses the function evaluation at the midpoint of the interval and odd derivatives at the endpoints. Because the weights for the odd derivatives sum to zero, the derivative calculations cancel out for the interior points in the composite form, so that these derivatives must only be calculated at the endpoints of the overall interval of integration. When using N subintervals, the basic rule which uses the midpoint function evaluation and the first derivative at the endpoints achieves fourth order accuracy for the cost of N/2 function evaluations and 2 derivative evaluations, whereas the three point open newton-Cotes method uses 3N/4 function evaluations to achieve the same order of accuracy. These derivative-based midpoint quadrature methods are shown to be more computationally efficient than both the open and closed newton-Cotes quadrature rules of the same order. This family of derivative-based midpoint quadrature rules are derived using the concept of precision, along with the error term. A theorem concerning the order of accuracy of quadrature rule using the concept of precision is provided to justify its use to determine the leading order error term.

Though well-known for its simplicity and efficiency, newton’s method applied to a complex polynomial can fail quite miserably, even on a relatively large open set of initial guesses. In this work, we present some analytic and numerical results for newton’s method applied to the complex quartic family where is a parameter. The symmetric location of the roots of?allows for some easy reductions. In particular, when λ is either real or purely imaginary, standard techniques from real dynamical systems theory can be employed for rigorous analysis. Classifying those λ-values where newton’s method fails on an open set leads to complex and aesthetically intriguing geometry in the λ-parameter plane, complete with fractal-like figures such as Mandelbrot-like sets, tricorns and swallows.
