Serological diagnosis of nasopharyngeal carcinoma by enzyme linked immunosorbant assay:optimization,standardization and diagnostic criteria

Objectives To produce an enzyme linked immunosorbant assay (eLISA) to detect antibodies against epstein Barr virus (eBV) specified nuclear antigen 1 (eBNA 1) and the 18kD eBV matrix protein, and to determine and optimize its sensitivity and specificity for the diagnosis of nasopharyngeal carcinoma (NPC).Methods We used a combination of highly purified glutathione transferase fusion proteins of the 40kD carboxy domain of eBNA1 and the 18kD eBV matrix protein for coating eLISA plates. In three separate studies, we tested for IgA antibodies in serum specimens from 28 eBV seronegative donors, 284 eBV seropositive donors and 160 newly diagnosed NPC patients. By comparing the sensitivity and specificity of diagnosis obtained for different cutoff values, we derived several quantitative parameters to evaluate assay performance, establish objective diagnostic criteria which optimize the intrinsic diagnostic capability of the assay and assess the significance of individual test results, respectively. Optimum cutoff optical density (OD) is defined as the cutoff OD where sensitivity of the assay equals its specificity, and resolution of the assay is indicated by the value of sensitivity (or specificity) determined at the optimum cutoff OD. Diagnosis of NPC was achieved by setting a cutoff zone at +/-20% of this *** All the eBV seronegative donors tested were not reactive, and most of the eBV seropositive donors were weakly reactive, while the majority of NPC patients were moderately or strongly reactive. While the assay was thus shown to be specific for eBV, there was an overlap in the level of these serum antibodies between few individuals of the two latter groups. It was shown that the assay performed equally well in two separate studies conducted under different testing conditions and using different collections of sera in that assay resolution determined on these occasions were 86% and 87% respectively. emerging evidence suggests that microbial dysbiosis plays vital roles in many human ***,knowledge of whether the microbial community in thyroid tumor is related to tumorigenesis remains *** this study,we aimed to explore the microbial community in thyroid tissues and its contribution to papillary thyroid cancer(PTC).In parallel,we performed microbial profiling and transcriptome sequencing in the tumor and adjacent normal tissues of a large cohort of 340 PTC and benign thyroid nodule(BTN)*** microbial signatures were identified in PTC,BTN,and their adjacent nontumor ***-thyroid tissue bacteria were verified by means of bacteria staining,fluorescence in situ hybridization,and immunoelectron *** found that 17 bacterial taxa were differentially abundant in PTC compared with BTN,which included enrichment in PTC of the pathobionts Rhodococcus,Neisseria,Streptococcus,Halomonas,and Devosia,and depletion of the beneficial bacteria *** differentially abundant bacteria could differentiate PTC tumor tissues(PTC-T)from BTN tissues(BTN-T)with an area under the curve(AUC)of 81.66%.Microbial network analysis showed increased correlation strengths among the bacterial taxa in PTC-T in comparison with ***-corresponding bacteria(i.e.,erwinia,Bacillus,and Acinetobacter)were found to be enriched in PTC with Hashimoto’s ***,our integrative analysis revealed that the PTC-enriched bacteria had a positive association with key PTC-oncogenic pathway-related genes,including BRAF,KRAS,IRAK4,CTNNB1,PIK3CA,MAP3K7,and *** conclusion,our results suggest that intratumor bacteria dysbiosis is associated with the thyroid tumorigenesis and oncogenic signaling pathways of PTC.

Tocopherols belong to a subgroup of the vitamin e family. Dietary feeding of a γ-tocopherol-rich mixture of tocopherols(γ-Tm T) inhibits prostate tumorigenesis in TRAMP mice. In this study, we aimed to investigate mechanisms of prostate carcinogenesis in TRAMP mice by identifying differentially expressed pathways and effects of γ-Tm T on these pathways. eight-week-old TRAMP and age-matched C57BL/6 mice were administered either 600 mg/kg of γ-Tm T or a control vehicle via oral gavage. Twelve hours after dosing, prostate tissues were collected for RNA extraction. Whole genome mouse microarrays were used to examine gene expression profiles. The expression of the selected genes were validated using quantitative PCR. Thousands of genes and various pathways were altered in the prostates of TRAMP mice. Compared to C57BL/6 mice, TRAMP mice exhibited enhanced proliferation, suppressed expression of antioxidant and phase II detoxification enzymes, and metabolic reprogramming in the prostate. γ-Tm T differentially regulated the gene expression profiles of TRAMP and C57BL/6 mice, with only a small percentage of genes overlapping. γ-Tm T inhibited genes involved in proliferation and glucose metabolism and induced several antioxidant/phase II detoxification genes in TRAMP mice. γ-Tm T modulates multiple aberrant pathways in the prostate of TRAMP mice, which might represent important mechanisms for prostate cancer prevention.

＜正＞In this paper we investigate the turbulent structures and transport of heat in oscillatory plane Couette flow at Reynolds number = 5,000 by solving the unsteady incompressible Navier - Stokes equations directly. This flow consists of a working fluid sandwiched between a lower stationary wall and an upper wall moving with a prescribed velocity u =U+AUsin（2πft） , which is a combination of steady and oscillating components. With different velocity ratios A , the turbulence characteristics in steady and oscillatory Couette flows are studied.

Ripples are widely formed in river channels and coastal regions, the bed-forms and vortex dynamics accompanied with them are quite important because they are responsible for frictions and sediment transports. The coherent vortex structure play a key role in the process, so how to model them is needed for understanding the formation process of sand ripples. In this paper, 2D Large-eddy-Simulation (LeS) was used to predict the flow structure and the dynamics of coherent vortex. The numerical results show a complete process of formation, evolvement and disappearance for the cases of unidire ctional and oscillatory flows over symmetic and asymmetric ripples

The stability of fluid flow in a horizontal layer of Brinkman porous medium with fluid viscosity different from effective viscosity is investigated. A modified Orr-Sommerfeld equation is derived and solved numerically using the Chebyshev collocation method. The critical Reynolds number Re, the critical wave number ac and the critical wave speed cc are computed for various values of porous parameter and ratio of viscosities. Based on these parameters, the stability characteristics of the system are discussed in detail. Streamlines are presented for selected values of parameters at their critical state.
