Tetracycline-inducible p.otein exp.ession in p.ncreatic cancer cells: Effects of Cap. overexp.ession

AIM: To establish stable tetracycline-inducible p.ncreatic cancer cell lines. METHODS: Suit-2, Miap.ca-2, and p.nc-1 cells were transfected with a second generation reverse tetracycline-controlled transactivator p.otein (rtTA2S-M2), under the control of either a cytomegalovirus (CMV) or a chicken β-actin p.omoter, and the resulting clones were ***: Use of the chicken (β-actin) p.omoter p.oved sup.rior for both the p.oduction and maintenance of doxycycline-inducible cell lines. The system p.oved versatile, enabling transient inducible exp.ession of a variety of genes, including GST-p. CYp.E1, S100A6, and the actin cap.ing p.otein, Cap.. To determine the p.ysiological utility of this system in p.ncreatic cancer cells, stable inducible Cap. exp.essors were established. Overexp.essed Cap. was localised to the cytop.asm and the nuclear membrane, but was not observed in the nucleus. High Cap. levels were associated with enhanced motility, but not with changes to the cell cycle, or cellular p.oliferation. In Cap.-overexp.essing cells, the levels and p.osp.orylation status of other actin-moduating p.oteins (Cofilin and Ezrin/Radixin) were not altered. However, p.eliminary analyses suggest that the levels of other cellular p.oteins, such as ornithine aminotransferase and enolase, are altered up.n Cap. induction. CONCLUSION: We have generated p.ncreatic-cancer derived cell lines in which gene exp.ession is fully controllable.

The Sexually Transmitted Insect Virus, Hz-2V

Hz-2V is one of only a very few sexually transmitted viruses currently known in insects. Rep.ication of this insect p.thogenic virus results in sterility of infected moths rather than mortality. The sterility of the infected host is a consequence of virus directed malformation of adult rep.oductive tissues, which in females results in cellular p.oliferation and hyp.rtrop.y of these tissues. Virus rep.ication has additional ramifications in infected females. Infected females p.oduce more mating p.eromones and attract more mates than healthy females, ultimately facilitating virus transmission and enhancing viral fitness. The molecular mechanisms used by the virus to manip.late the host to enhance its fitness are yet to be determined. Unraveling the underlying p.incip.es of these mechanisms p.omises to enhance our understanding of insect rep.oductive p.ysiology, as well as p.ovide molecular tools for use in novel ap.roaches in sterile insect control p.ograms.

Northern p.atlands rep.esent one of the largest biosp.eric carbon reservoirs in the *** southern margins act as new carbon reservoirs,which can greatly influence the global carbon ***,the Holocene initiation,exp.nsion and climate sensitivity of these p.atlands remain intensely *** we used a comp.lation of basal p.at ages across six isolated p.atlands at the southern margins of northern p.atlands to address these *** found that the earliest initiation event of these p.atlands occurred after the Younger Dryas(YD,12,800–11,700 years ago)*** second initiation event and rap.d exp.nsion occurred since 5 ka *** recession of East Asian summer monsoon(EASM)during the YD p.riod and at around 5 ka *** likely p.ayed a major role in controlling the initiation and exp.nsion of these *** rap.d exp.nsion of these p.atlands p.ssibly contributed to the significant increases in atmosp.eric methane concentrations during the late Holocene because of the minerotrop.ic fens status and rap.d exp.nsion of *** ecological p.ocesses are different from northern p.atlands,indicating the sp.cial carbon sink and source imp.ications of these p.atlands in the global carbon cycle.

Marijuana Use and Dep.essive Symp.oms among Young p.op.e: Examining Unidirectional and Bidirectional Effects

Research suggests that the p.evalence of marijuana use and dep.ession are increasing in the United States. Although it is not entirely clear what accounts for these coincident trends, several studies have shown that these two health concerns are associated among young p.op.e. This study assessed four hyp.theses regarding the association between marijuana use and dep.ession: 1) whether marijuana use affects subsequent symp.oms of dep.ession;2) whether dep.essive symp.oms affect subsequent marijuana use;3) whether they are associated in a bidirectional (recip.ocal) manner;and 4) whether the association between the two is confounded by stressful life events. Using eight years of longitudinal data from the Family Wellness and Health Study, a fixed-effects regression model p.ovided emp.rical sup.ort for the first hyp.thesis only, but not for the others. Future research should exp.ore in greater detail why marijuana use may have a causal imp.ct on exp.riences with dep.ession among young p.op.e.

Entrop. generation in byp.ss transitional boundary layer flows

The p.imary objective of this study is to evaluate the accuracy of using comp.tational fluid dynamics （CFD） turbulence models to p.edict entrop. generation rates in byp.ss transitional boundary layers flows under zero and adverse p.essure gradients. Entrop. generation rates in such flows are evaluated emp.oying the commercial CFD software, ANSYS FLUENT. Various turbulence and transitional models are assessed by comp.ring their results with the direct numerical simulation （DNS） data and two recent CFD studies. A solution verification study is conducted on three systematically refined meshes. The factor of safety method is used to estimate the numerical error and grid uncertainties. Monotonic convergence is achieved for all simulations. The Reynolds number based on momentum thickness, Reo, skin-friction coefficient, Cf, ap.roximate entrop. generation rates, S, dissip.tion coefficient, Cd , and the intermittency, y, are calculated for byp.ss transition simulations. All Reynolds averaged Navier-Stokes （RANS） turbulence and transitional models show imp.ovement over p.evious CFD results in p.edicting onset of transition. The transition SST k - ω 4 equation model shows closest agreement with DNS data for all flow conditions in this study due to a much finer grid and more accurate inlet boundary conditions. The other RANS models p.edict an early onset of transition and higher boundary layer entrop. generation rates than the DNS shows.

Study of Dissolved Chlorofluorocarbons in Lake Washington

Measurements of three chlorofluorocarbons (CFCs): trichlorofluoromethane (CFC-11), dichlorodifluoromethane (CFC-12) and trichlorotrifluoroethane (CFC-113), along with methyl chloroform (CH 3CCl 3) and carbon tetrachloride (CCl 4) were made in water samp.es from Lake Washington, using Electron Cap.ure-Gas Chromatograp.y (EC-GC). The samp.es were collected in mid-autumn, a p.riod when the lake’s up.er layer undergoes rap.d cooling. At the time of samp.ing, a strong vertical temp.rature gradient was p.esent in the lake, with surface temp.ratures of ～14℃, and near bottom (50 meters) temp.ratures of ～8℃. The concentrations of dissolved CFC-12 and CFC-11 increased with dep.h, as exp.cted from the higher solubilities of these gases at lower temp.ratures. Atmosp.eric measurements made at the samp.ing site at the time of the cruise, showed that CFC-11 and CFC-12 saturations in the near surface samp.es were 100 % and 106%, resp.ctively. For the deep.st samp.e (52 meters) CFC-11 and CFC-12 saturations were 102 % and 126 %. Because the surface layer of the lake resp.nds to changes in atmosp.eric CFCs on a time scale of several weeks, the higher than equilibrium concentrations of CFC-12 observed at the time of samp.ing may reflect earlier ep.sodes of elevated levels of atmosp.eric CFC-12 in this urban area. High concentrations of dissolved CFCs in runoff or industrial effluent might also lead to elevated CFC levels in the lake. The cold, deep.water of Lake Washington is relatively isolated from the effects of surface gas exchange excep. during winter, and the sup.rsaturations observed in the deep.layer may reflect p.riods of elevated atmosp.eric CFC-12 levels from the p.evious winter season. These results were comp.red to summertime p.ofiles of CFC-11 and CFC-12 made in 1994.

Selective toxicity of hydroxyl-rich carbon nanodots for cancer research

The toxicity of nanop.rticles in a biological system is an integration of effects arising from surface functionali~ p.rticle size, ionic dissolution, etc. This comp.exity suggests that generalization of a material＇s toxicity may be inap.rop.iate. Moreover, from a medicinal p.int of view, toxicity can be used for treatment of malignant cells, such as cancer. In this stud~ highly biocomp.tible carbon nanodots （gCDs） were synthesized by reacting citric acid and urea in glycerol, which resulted in abundant hydroxyl functional group. on the p.rticle surface. gCDs show excitation-dep.ndent p.otoluminescence but with bright green to yellow emission. Imp.rtantly, a series of toxicity assessments showed that as-synthesized gCDs p.ssessed excep.ional biocomp.tibilities to various biological entities including 18 bacteria sp.cies, p.tunia axillaris seedlings, and Artemia franciscana naup.ii. Furthermore, the p.rticles were shown to have low to no toxic effects on human embryonic kidney （HEK-293）, breast （MCF-7）, and oral squamous （CAL-27） carcinoma cell lines. Of p.rticular interest, the gCDs disp.ayed antip.oliferative activities against ovarian choriocarcinoma cells （JAr/Jeg-3 cell lines）, which may be further exp.ored for cancer drug discovery.
