Immunoproteome analysis of soluble and membrane proteins of Shigella flexneri 2457T

AIM: To profile the immunogenic proteins of Shigella flexneri (S. flexneri) expressed during human infection using a proteomic approach. METHODS: Soluble and membrane protein extractions of S. flexneri 2457T were separated by two-dimensional gel electrophoresis (2-DE). Proteins were transferred to PVDF membrane and immunoblotted with sera from shigellosis patients. Reactive protein spots were matched to Coomassie stained gels run in parallel, cut out and trypsin digested. Matrix-assisted laser desorption/ionization time of flight-mass spectrometry (MALDI-TOF-MS) was used to determine the peptide mass fingerprints, which were searched in the MASCOT database to identify the protein. RESULTS: A total of 8 immunoreactive proteins were successfully identified from the Coomassie stained gels in three repeats. Six of these proteins have not previously been reported as immunogenic in S. flexneri. These proteins could be potential candidates for vaccine or attenuation studies. CONCLUSION: Soluble and membrane proteins of S. flexneri 2457T have been screened by 2-DE and immunoblotting with sera from shigellosis patients. Eight proteins are identified as immunogenic.

Fault tree analysis is an effective method for predicting the reliability of a system. It gives a pictorial representation and logical framework for analyzing the reliability. Also, it has been used for a long time as an effective method for the quantitative and qualitative analysis of the failure modes of critical systems. In this paper, we propose a new general coverage model （GCM） based on hardware independent faults. Using this model, an effective software tool can be constructed to detect, locate and recover fault from the faulty system. This model can be applied to identify the key component that can cause the failure of the system using failure mode effect analysis （FMEA）.

This study introduces a two-level strategy for efficient execution of multiple sequence alignment (MSA) of complex heterogeneous sequences. The two levels of the proposed technique are comprised of: designing the discrete firefly algorithm (DFFA) for the formation and implementation of makespan minimisation on parallel machines, followed by performing Ctrie-based caching for pairwise alignment to reduce the load on the data servers for handling multiple queries. The proposed strategy addresses a multi-client problem that aims to acquire the full advantage of the computational power of parallel connected machines. Further, it is shown that the inclusion of Ctrie as caching mechanism successively improves the performance of the system with accretion in several sequences. Performance of proposed DFFA is also compared with discrete versions of four swarm intelligence based algorithms at the criteria of makespan minimisation and the rate of convergence on two kinds of complex and diverse datasets. The work is unique in this sense: it is the first swarm-intelligence-based implementation for the addressed problem;it is so far the first approach for Ctrie based caching of the MSA on the scheduled parallel machines;hybridisation of DFFA with Ctrie for caching the MSA results is also a novel implementation.

Introduction:The study was conducted in Mudumalai Tiger Reserve,in the Western Ghats to understand the effect of a single fire event on tree diversity and regeneration *** forest patches were selected which were unburned,2-year-old burn,5-year-old burn,and 15-year-old *** 0.1 ha square plots were laid randomly in all four patches and analyzed for tree diversity,stand structure,and regeneration of tree ***:A total of 4129 individuals of tree species were recorded in field surveys,comprising 3474 seedlings,121 saplings,and 534 ***,40 tree species were recorded in study plots,from which 28 species were seedlings,16 species were saplings,and 37 species were at tree ***:Tree diversity decreased in 2-year-old and 5-year-old burnt plots and was reached to the level of unburnt plots in 15 years of *** of small size classes started increasing after the *** density increased linearly in subsequent years after fire but sapling and tree density recorded less than control in B2 but was higher in B5 and *** overall fire affected diversity,but regeneration showed a positive trend.

Influence of tool rotation speeds on mechanical and morphological properties of friction stir processed nano hybrid composite of MWCNT-Graphene-AZ31 magnesium

The ever-increasing demand for light weighted hard materials for transportation industries encouraged researchers to develop composites with excellent mechanical properties which can transform it into more economical and *** the metals with carbonaceous nanomaterials are progressively in focus due to their excellent capability to inculcate and tailor the properties of *** the present research,a hybrid nanocomposite of MWCNT-Graphene-AZ31 Mg alloy has been developed by using variable tool rotation speeds with friction stir processing(FSP).Optimized reinforcement ratio of 1.6%*** and 0.3%*** graphene have been used with variable tool rotation speeds,whereas other processing parameters are kept *** developed specimens were investigated using standard testing equipment for evaluating and comparing the mechanical properties on the basis of the microstructure of the processing regions and their morphological analysis,according to the ASTM *** obtained results revealed an improvement of 19.72%in microhardness and 77.5% of compressive strength in comparison with the base metal AZ 31 Magnesium alloy,with a tool rotational speed of *** values of tensile stress and percentage area reduction were recorded as less than that of the base metal matrix,but an increasing trend has been observed in the values of both with the improvement on rotational speeds of the *** effectual strengthening mechanisms are analyzed on the bases of SEM images and observed that discussed and found that grain refinement strengthening is the major contributor to the strength of the nanocomposite.

Performance of concrete structures is significantly influenced and governed by its durability and resistance to environmental or exposure conditions, apart from its physical strength. It can be monitored, evaluated and predicted through modeling of physical deterioration mechanisms, performance characteristics and parameters and condition monitoring of in situ concrete structures. One such study has been conducted using Non-destructive testing equipment in the city of Bhopal and around located in India. Some selected parameters influencing durability of reinforced concrete （RC） structures such as concrete cover, carbonation depth, chloride concentration, half cell potential and compressive strength have been measured, for establishing correlation among various parameters and age of structures. Effects of concrete cover and compressive strength over the variation of chloride content with time are also investigated.

In this reported work,we study a major X-class flare(X9.3) that arose from NOAA Active Region(AR) 12673 on 2017 September 6,from 11:53 UT to 12:10 UT in multi-wavelength *** event also produced a fast coronal mass ejection(CME).NOAA AR 12673 emerged at S09 W30 on 2017 September6 and grew rapidly to a large *** 2017 September 9,the maximum area of this AR was 1060 millionth of the solar *** group of sunspots disappeared over the west limb of the Sun(S09 W83) on September *** was a fast emerging flux *** group of sunspots showed magnetic configuration category *** identified their earliest signatures of eruption in AIA 94A images with initialization and successive rapid growth from low coronal heights of hot channeled *** the other hand,the CME associated with this flare event triggered the intense Dst at 1 AU(–142 nT).We have acquired observations and analyze the reported event from the Sun’s surface,corona(source AR),interplanetary space and in-situ measurement near *** addition,here we analyze the complex processes of CMECME interaction that have contributed a significant role to make the reported event so geoeffective.

The network of Himalayan roadways and highways connects some remote regions of valleys or hill slopes,which is vital for India’s socio-economic *** to natural and artificial factors,frequency of slope instabilities along the networks has been increasing over last few *** of stability of natural and artificial slopes due to construction of these connecting road networks is significant in safely executing these roads throughout the *** rock mass classification methods are generally used to assess the strength and deformability of rock *** study assesses slope stability along the NH-1A of Ramban district of North Western *** structurally and non-structurally controlled rock mass classification systems have been applied to assess the stability conditions of 14 *** evaluating the stability of these slopes,kinematic analysis was performed along with geological strength index(GSI),rock mass rating(RMR),continuous slope mass rating(CoSMR),slope mass rating(SMR),and Q-slope in the present *** SMR gives three slopes as completely unstable while CoSMR suggests four slopes as completely *** stability of all slopes was also analyzed using a design chart under dynamic and static conditions by slope stability rating(SSR)for the factor of safety(FoS)of 1.2 and 1 respectively.Q-slope with probability of failure(PoF)1%gives two slopes as stable *** slope angle has been determined based on the Q-slope safe angle equation and SSR design chart based on the *** value ranges given by different empirical classifications were RMR(37-74),GSI(27.3-58.5),SMR(11-59),and CoSMR(3.39-74.56).Good relationship was found among RMR&SSR and RMR&GSI with correlation coefficient(R 2)value of 0.815 and 0.6866,***,a comparative stability of all these slopes based on the above classification has been performed to identify the most critical slope along this road.
