Genetic linkage mapping and QTL identification for salinity tolerance in Indian mustard(Brassica juncea L.Czern and Coss.)using SSr markers

Soil salinity is one of the major environmental constraints that limits crop yield and nearly 7%of the total area worldwide is affected by salinity.Salinity-induced oxidative stress causes membrane damage during germination and seedling growth.Indian mustard is a major oilseed crop in India and its production and productivity are severely affected by salt stress.Breeding Brassica cultivars for salinity tolerance by conventional means is very difficult and time-consuming.Therefore,understanding the molecular components associated with salt tolerance is needed to facilitate breeding for salt tolerance in Brassica.In this investigation,quantitative trait loci(QTLs)associated with salt tolerance were identified using F_(2:3)mapping population developed from a cross between CS52(salinity tolerant)and rH30(salinity sensitive).Parents and F_(2:3)were evaluated under controlled and salinity stress conditions for 14 morpho-physiological traits for two consecutive generations(F2 and F_(2:3)),explaining proportion of the phenotypic variance under control condition.Simple sequence repeat(SSr)markers were used for mapping studies.A genetic linkage map based on 42 simple sequence repeats(SSrs)markers was constructed covering 2298.5 cM(Haldane)to identify the loci associated with salt tolerance in Brassica juncea.Forty-one SSrs showing polymorphism in the parents(CS52 and rH30)were mapped on 8 linkage groups(C1–C8).One marker(nga 129)did not map to any of the linkage group and was excluded from mapping.Linkage group 5(C5;317.9 cM)was longest and linkage group 1(C1,255.0 cM)was shortest.Further,we identified 15 QTLs controlling 8 traits using F_(2:3)population.These QTLs explained 12.44–60.63%of the phenotypic variation with a LOD score range of 3.62–5.97.Out of these QTLs,QMI4.1 related to membrane injury showed 51.28%phenotypic variance with a LOD score of 3.34.QTL QBYP8.1 related to biological yield per plant showed 60.63%phenotypic variance at a LOD score of 3.62.The highest LOD score of 5.97 was recorded for QTL related to

Development of high-throughput phenotyping technologies has progressed considerably in the last 10 years.These technologies provide precise measurements of desired traits among thousands of field-grown plants under diversified environments;this is a critical step towards selection of better performing lines as to yield,disease resistance,and stress tolerance to accelerate crop improvement programs.High-throughput phenotyping techniques and platforms help unrave-ling the genetic basis of complex traits associated with plant growth and development and targeted traits.This review focuses on the advancements in technologies involved in high-throughput,field-based,aerial,and unmanned platforms.Development of user-friendly data management tools and softwares to better understand phenotyping will increase the use of field-based high-throughput techniques,which have potential to revolutionize breeding strategies and meet the future needs of stakeholders.

Management of proximal tibial stress fracture associated with advanced knee osteoarthritis:A systematic review

Purpose:Tibial stress fracture associated with knee osteoarthritis is an unusual and difficult clinical scenario.There is no clear existing treatment guideline for this uncommon clinical disease.The aim of this study is to review the impact of various treatment options for patients with advanced knee osteoarthritis associated with proximal tibial stress fracture.Methods:The study was performed using the databases of PubMed and Scopus.Methodological index for non-randomized studies score was used to evaluate the included studies’bias.The concluded data included the treatment approach,reported outcome measure,and time to fracture union.The literature search was started in December 2021 and accomplished in January 2022.A narrative description of the different methods and comparison of their results were done.results:Out of total assessed 69 studies,9 studies were included in our review.The commonest treatment approach used was total knee arthroplasty by long tibial stem extension.The mean preoperative knee society score and knee functional score were 30.62 and 23.17,respectively.The mean postoperative knee society knee score was 86.87,while the functional score was 83.52.The average reported time to achieve fracture union was 4 months(a range of 2.07-5.50 months).Conclusion:The optimal clinical outcome for treating either acute or mobile tibial stress fracture in patients with advanced knee osteoarthritis can be achieved with long stem total knee arthroplasty.However,due to heterogeneity of data,comparison of different treatment options for chronic proximal tibial stress fracture mal-union/non-union coexisting with knee osteoarthritic and such inferences need to be judged cautiously.

Geospatial Analysis of Urban Heat Island Effects and Tree Equity

In recent decades, Urban Heat Island Effects have become more pronounced and more widely examined. Despite great technological advances, our current societies still experience great spatial disparity in urban forest access. Urban Heat Island Effects are measurable phenomenon that are being experienced by the world’s most urbanized areas, including increased summer high temperatures and lower evapotranspiration from having impervious surfaces instead of vegetation and trees. Tree canopy cover is our natural mitigation tool that absorbs sunlight for photosynthesis, protects humans from incoming radiation, and releases cooling moisture into the air. Unfortunately, urban areas typically have low levels of vegetation. Vulnerable urban communities are lower-income areas of inner cities with less access to heat protection like air conditioners. This study uses mean evapotranspiration levels to assess the variability of urban heat island effects across the state of Tennessee. results show that increased developed land surface cover in Tennessee creates measurable changes in atmospheric evapotranspiration. As a result, the mean evapotranspiration levels in areas with less tree vegetation are significantly lower than the surrounding forested areas. Central areas of urban cities in Tennessee had lower mean evapotranspiration recordings than surrounding areas with less development. This work demonstrates the need for increased tree canopy coverage.

BACKGrOUND Early prediction of response to percutaneous catheter drainage(PCD)of necrotic collections in acute pancreatitis(AP)using simple and objective tests is critical as it may determine patient prognosis.The role of white blood cell(WBC)count and neutrophil-lymphocyte ratio(NLr)has not been assessed as a tool of early prediction of PCD success and is the focus of this study.AIM To assess the value of WBC and NLr in predicting response to PCD in AP.METHODS This retrospective study comprised consecutive patients with AP who underwent PCD between June 2018 and December 2019.Severity and fluid collections were classified according to the revised Atlanta classification and organ failure was defined according to the modified Marshall Score.WBC and NLr were monitored 24 h prior PCD(WBC-0/NLr-0)and 24 h(WBC-1/NLr-1),48 h(WBC-2/NLr-2)and 72 h(WBC-3/NLr-3)after PCD.NLr was calculated by dividing the number of neutrophils by the number of lymphocytes.The association of success of PCD(defined as survival without the need for surgery)with WBC and NLr was assessed.The trend of WBC and NLr was also assessed post PCD.rESULTS One hundred fifty-five patients[median age 40±13.6(SD),64.5%males,53.5%severe AP]were included in the final analysis.PCD was done for acute necrotic collection in 99(63.8%)patients and walled-off necrosis in 56(36.1%)patients.Median pain to PCD interval was 24±69.89 d.PCD was successful in 109 patients(group 1)and 46 patients(group 2)who failed to respond.There was no significant difference in the baseline characteristics between the two groups except the severity of AP and frequency of organ failure.Both WBC and NLr showed an overall decreasing trend.There was a significant difference between WBC-0 and WBC-1(P=0.0001).WBC-1 and NLr-1 were significantly different between the two groups(P=0.048 and 0.003,respectively).The area under the curve of WBC-1 and NLr-1 for predicting the success of PCD was 0.602 and 0.682,respectively.At a cut-off value of 9.87 for NLr-1,the sensitivity and specificity for pred

Dear editor,Globe luxation is defined as complete prolapse of the eyeball outside the orbital cavity with eyelids closed behind the eyeball.Broadly,globe luxation can be categorized as traumatic,spontaneous or voluntary;males are affected more than females with an average age of 30 years.[1]Traumatic globe luxation is quite rare in children,with most cases reported in the adult age group.Orbital fracture comprises 5%to 25%of all facial fractures in the paediatric age group.[2]In cases with luxation of globe,immediate reposition is crucial for globe and vision salvage.We describe the clinical features,evaluation,and management of globe luxation present at birth following normal vaginal delivery conducted elsewhere.

In patients with lagophthalmos due to eyelid trauma,it is imperative to ensure adequate closure of the eye to protect the ocular surface.Sometimes it is difficult to surgically close eyelids by conventional methods of tarsorrhaphy,such as intermarginal suture tarsorrhaphy,Week’s tarsorrhaphy,and tongue-in-groove tarsorrhaphy.We need intact eyelid tissues to perform these procedures and might not be possible in patients with severe lagophthalmos due to lost/damaged eyelid tissues either due to deficient length of tissues or increased tension at the wound site,which can result in surgical failure.[1]

Bi-LSTM-Based Deep Stacked Sequence-to-Sequence Autoencoder for Forecasting Solar Irradiation and Wind Speed

Wind and solar energy are two popular forms of renewable energy used in microgrids and facilitating the transition towards net-zero carbon emissions by 2050.However,they are exceedingly unpredictable since they rely highly on weather and atmospheric conditions.In microgrids,smart energy management systems,such as integrated demand response programs,are permanently established on a step-ahead basis,which means that accu-rate forecasting of wind speed and solar irradiance intervals is becoming increasingly crucial to the optimal operation and planning of microgrids.With this in mind,a novel“bidirectional long short-term memory network”(Bi-LSTM)-based,deep stacked,sequence-to-sequence autoencoder(S2SAE)forecasting model for predicting short-term solar irradiation and wind speed was developed and evaluated in MATLAB.To create a deep stacked S2SAE prediction model,a deep Bi-LSTM-based encoder and decoder are stacked on top of one another to reduce the dimension of the input sequence,extract its features,and then reconstruct it to produce the forecasts.Hyperparameters of the proposed deep stacked S2SAE forecasting model were optimized using the Bayesian optimization algorithm.Moreover,the forecasting performance of the proposed Bi-LSTM-based deep stacked S2SAE model was compared to three other deep,and shallow stacked S2SAEs,i.e.,the LSTM-based deep stacked S2SAE model,gated recurrent unit-based deep stacked S2SAE model,and Bi-LSTM-based shallow stacked S2SAE model.All these models were also optimized and modeled in MATLAB.The results simulated based on actual data confirmed that the proposed model outperformed the alternatives by achieving an accuracy of up to 99.7%,which evidenced the high reliability of the proposed forecasting.

As one of the most important and essential macronutrients next to nitrogen,phosphorus(P)is important for plant development,but it is the least mobile nutrient element in plant and soil.Globally,P is mined from geological sediments and added to agricultural soils so as to meet the critical requirements of crop plants for agronomic productivity.Phosphorus exists in soil in both organic and inorganic forms.The various inorganic forms of the element in soil are salts with calcium,iron,and aluminum,whereas the organic forms come from decaying vegetation and microbial residue.There is a huge diversity of plant microbiomes(epiphytic,endophytic,and rhizospheric)and soil microbiomes that have the capability to solubilize the insoluble P and make it available to plant.The main mechanism for the solubilization of inorganic P is by the production of organic acids,which lowers soil pH,or by the production of acid and alkaline phosphatases,which causes the mineralization of organic P.The P-solubilizing and-mobilizing microorganisms belong to all three domains,comprising archaea,bacteria,and eukarya.The strains belonging to the genera Arthrobacter,Bacillus,Burkholderia,Natrinema,Pseudomonas,rhizobium,and Serratia have been reported as efficient and potential P solubilizers.The use of P solubilizers,alone or in combination with other plant growth-promoting microbes as an eco-friendly microbial consortium,could increase the P uptake of crops,increasing their yields for agricultural and environmental sustainability.

Neurodegenerative diseases are a heterogeneous group of maladies, characterized by progressive loss of neurons. These diseases involve an intricate pattern of cross-talk between different types of cells to maintain specific signaling pathways. A component of such intercellular cross-talk is the exchange of various types of extracellular vesicles (EVs). Exosomes are a subset of EVs, which are increasingly being known for the role they play in the pathogenesis and progression of neurodegenerative diseases, e.g., synucleinopathies and tauopathies. The ability of the central nervous system exosomes to cross the blood-brain barrier into blood has generated enthusiasm in their study as potential biomarkers. However, the lack of standardized, efficient, and ultra-sensitive methods for the isolation and detection of brain-derived exosomes has hampered the development of effective biomarkers. Exosomes mirror heterogeneous biological changes that occur during the progression of these incurable illnesses, potentially offering a more comprehensive outlook of neurodegenerative disease diagnosis, progression and treatment. In this review, we aim to discuss the challenges and opportunities of peripheral biofluid-based brain-exosomes in the diagnosis and biomarker discovery of Alzheimer’s and Parkinson’s diseases. In the later part, we discuss the traditional and emerging methods used for the isolation of exosomes and compare their advantages and disadvantages in clinical settings.
