Red blood cells(RBCs)are the most abundant human blood *** aggregation and deformation strongly determine blood viscosity which imp.cts hemorheology and *** turn,RBC p.op.rties dep.nd on di®erent endogenous and exogenous *** such factor is nitric oxide(NO),which is mainly p.oduced by endothelial cells(EC)from L-arginine amino acid in the circulatory *** the mechanisms of the RBC-endothelium interp.ay are not clear up.to date and considering its p.ssible clinical imp.rtance,the aims of this study are to investigate in vitro:(1)The e®ect of L-arginine induced NO on RBC aggregation and adhesion to endothelium;(2)the NO e®ect on RBC aggregation and deformation induced by L-arginine and sodium nitrop.usside without the p.esence of endothelium in the *** RBC aggregation and adhesion to a monolayer of EC were studied using op.ical tweezers(OT).The RBC deformability and aggregation without endothelium in the samp.es were studied using the°ow chamber method and myrenne *** con¯rmed that NO increases deformability and decreases aggregation of *** showed that the soluble guanylate cyclase p.thway ap.ears to be the only NO signaling p.thway *** the samp.es with the endothelium,the\bell-shap.d"dep.ndence of RBC aggregation force on L-arginine concentration was observed,which imp.oves our knowledge about the p.ocess of NO p.oduction by ***,data related to L-arginine accumulation by endothelium were obtained:Necessity of the p.esence of extracellular L-arginine stated by other authors was p.t under *** our study,NO decreased the RBC-endothelium adhesion,however,the tendency ap.eared to be weak and was not con¯rmed in another set of *** our knowledge,this is the¯rst attemp. to measure the forces of RBC adhesion to endothelium monolayer with OT.

p.rp.se: The oxidative stress (OS) hyp.thesis of overtraining syndrome argues that increased p.oduction of free radicals through exercise cause muscle fatigue and damage resulting in lower athletic p.rformance. Several studies have investigated OS immediately before and after exercise bouts in a training macrocycle. Our study aimed to comp.re OS of endurance athletes between a comp.tition macrocycle and the immediate p.st-season recovery macrocycle. In addition, we aimed to identify athletes who exp.rienced an unexp.ainable drop.in athletic p.rformance during the comp.tition season in order to comp.re their OS to those who exp.rienced no drop.in p.rformance. methods: Fifteen members of the University of Alaska Fairbanks cross country ski team volunteered for this study. Blood samp.es were taken in early February (“mid-season”) and late Ap.il (“p.st-season”). p.rticip.nts comp.eted questionnaires regarding p.ysical activity and athletic p.rformance at the time of the blood draws. p.asma was analyzed for 4-hydroxynonenalp. p.(HNE), nitrotyrosine,p. p.nitric oxide (NOX), and sup.roxide dismutase (SOD). Significance was determined by Wilcoxon and mann-Whitney tests. Results: p.rticip.nts disp.ayed significantly higher (p.Conclusion: Signs of oxidative stress and mitigation during the p.st-season recovery macrocycle were higher in athletes who rep.rted exp.riencing a drop.in athletic p.rformance during the comp.tition season macrocycle.

Biochemical Liver Functions and molecular Identification of Fasciola hep.tica from Exp.rimentally Infected Rat model

The current study was p.rformed to evaluate the liver function status as well as molecular characterization of the recovered worms in rats exp.rimentally infected with F. hep.tica. Sixteen male Wister rats aged 30 days were randomly allocated into two group. (n = 8). The first group.was infected orally with 15 viable encysted metacercaria of F. hep.tica p.r animal. The other group.was kep. non-infected (control group.. At zero time (before infection), the 2p.ndp., 4p.thp., 6p.thp., 8p.thp., 10p.thp., 12p.thp. and 14p.thp. weeks p.st-infection (Wp.), blood and serum samp.es were collected via p.ncture of retro-orbital p.exus of veins from each rat. Serum enzyme level (AST and ALT) and total p.otein were measured, and the serum p.otein p.ofile was carried out using agarose gel electrop.oresis. During the p.riod of the exp.riment, serum ALT and AST activities and serum total globulins significantly increased while serum total p.oteins and albumin markedly decreased in the infected group. On the 14p.thp. Wp., the data of the electrop.erogram showed that globulin fractions (α1-, β- and γ-globulin) levels were significantly increased while α2-globulin was markedly decreased in the infected group. The molecular analysis confirmed the amp.ification of the ITS1, ITS2 and NDI genes of F. hep.tica recovered from the infected liver of rats with amp.icon sizes of 630, 510 and 560 bp. resp.ctively. Sequencing of the amp.ified ITS gene resulted in the determination of 3 strains (p.108836, p.108837, and p.108838). Also, analysis of the ITS2 gene resulted in obtaining 3 isolates under the accession numbers (p.109065, p.109066, and p.109067). In conclusion, fasciolosis in the rat model is suitable for routine exp.rimental infections and caused a p.onounced liver dysfunction with discharging of the Fasciola eggs in the faeces and the develop.ent of adult stages in the bile ducts.
