nAT10-mediated ac 4 C-modified AnKZF1 promotes tumor progression and lymphangiogenesis in clear-cell renal cell carcinoma by attenuating YWHAE-driven cytoplasmic retention of YAP1

Background:Lymphatic metastasis is one of the most common metastatic routes and indicates a poor prognosis in clear-cell renal cell carcinoma(ccRCC).n-acetyltransferase 10(nAT10)is known to catalyze n4-acetylcytidine(ac4C)modification of mRnA and participate in many cellular ***,its role in the lymphangiogenic process of ccRCC has not been *** study aimed to elucidate the role of nAT10 in ccRCC lymphangiogenesis,providing valuable insights into potential therapeutic targets for ***:ac4C modification and nAT10 expression levels in ccRCC were assessed using public databases and clinical *** investigations involved manipulating nAT10 expression in cellular and mouse models to study its role in *** insights were gained through a combination of RnA sequencing,mass spectrometry,co-immunoprecipitation,RnA immuno-precipitation,immunofluorescence,and site-specific mutation ***:We found that ac4C modification and nAT10 expression levels increased in ***10 promoted tumor progression and lymphangiogene-sis of ccRCC by enhancing the nuclear import of Yes1-associated transcriptional regulator(YAP1).Subsequently,we identified ankyrin repeat and zinc fin-ger peptidyl tRnA hydrolase 1(AnKZF1)as the functional target of nAT10,and its upregulation in ccRCC was caused by nAT10-mediated ac4C *** analyses demonstrated that AnKZF1 interacted with tyro-sine 3-monooxygenase/tryptophan 5-monooxygenase activation protein epsilon(YWHAE)to competitively inhibit cytoplasmic retention of YAP1,leading to transcriptional activation of pro-lymphangiogenic ***:These results suggested a pro-cancer role of nAT10-mediated acetylation in ccRCC and identified the nAT10/AnKZF1/YAP1 axis as an under-reported pathway involving tumor progression and lymphangiogenesis in ccRCC.

Carbon nanocages bridged with graphene enable fast kinetics for dual-carbon lithium-ion capacitors

Lithium-ion capacitors(LICs) combining the advantages of lithium-ion batteries and supercapacitors are considered a promising nextgeneration energy storage device. However, the sluggish kinetics of battery-type anode cannot match the capacitor-type cathode, restricting the development of LICs. Herein, hierarchical carbon framework(HCF) anode material composed of 0D carbon nanocage bridged with 2D graphene network are developed via a template-confined synthesis process. The HCF with nanocage structure reduces the Li^(+) transport path and benefits the rapid Li^(+) migration, while 2D graphene network can promote the electron interconnecting of carbon nanocages. In addition, the doped n atoms in HCF facilitate to the adsorption of ions and enhance the pseudo contribution, thus accelerate the kinetics of the anode. The HCF anode delivers high specific capacity, remarkable rate capability. The LIC pouch-cell based on HCF anode and active HCF(a-HCF) cathode can provide a high energy density of 162 Wh kg^(-1) and a superior power density of 15.8 kW kg^(-1), as well as a long cycling life exceeding 15,000cycles. This study demonstrates that the well-defined design of hierarchical carbon framework by incorporating 0D carbon nanocages and 2D graphene network is an effective strategy to promote LIC anode kinetics and hence boost the LIC electrochemical performance.

Effect of red clover isoflavones on hormone, immune, inflammatory,and plasma biochemistry in lactating dairy cows

This study was to conducted to investigate the effect of red clover isoflavones on the health indicated by immune status and blood biochemistry in dairy cows. Sixty-eight healthy Holstein lactating cows were randomly divided into four treatments(n = 17 per treatment) from 5 blocks according to milk yield using a randomized complete block design. no initial differences in parity(2.13 ± 1.21), days in milk(165 ± 21 d), and milk yield(33.93 ± 3.81 kg/d) between groups. Cows were fed the basal diet supplemented with 0, 2, 4, or 8 g/kg red clover extract(RCE) in diet(dry matter based). Feeding, refusal feed weights, and milk yield were recorded three consecutive days in weeks 0, 4, 8, and 12. Blood was collected from the tail vein of the cows on the last day of weeks 4, 8 and 12, 1 h after the morning feeding,and analyzed for hormones, immunoglobulins, inflammatory markers, and markers of liver and kidney activities. The dry matter intake was significantly decreased by 3.7% in the 8 g/kg group(P nificantly higher in both of the 2 and 4 g/kg groups(P nd prolactin showed a quadratic effect with increasing RCE levels, with the highest in the 4 g/kg group(P necrosis factor(TnF)-a, interleukin(IL)-6, and IL-1β levels decreased linearly with increasing dietary RCE levels. Plasma IL-18 levels showed a quadratic effect with increasing dietary RCE levels, with significantly lower levels in both of the 2 and 4 g/kg groups(P noglobulin A and D-lactic acid levels showed a quadratic effect with increasing dietary RCE levels,with significantly higher level in the 4 g/kg group(P nction and kidney activity makers were similar(P > 0.05). These results recommend the supplementation of RCE at a level from 2 to4 g/kg DM.

●AIM:To explore the combined application of surgical navigation nasal endoscopy(nnE)and three-dimensional printing technology(3DPT)for the adjunctive treatment of orbital blowout fractures(OBF).●METHODS:Retrospective analysis was conducted on the data of patients with OBF who underwent surgical treatment at the Affiliated Eye Hospital of nanchang University between July 2012 and november *** control group consisted of patients who received traditional surgical treatment(n=43),while the new surgical group(n=52)consisted of patients who received nnE with *** difference in therapeutic effects between the two groups was evaluated by comparing the duration of the operation,best corrected visual acuity(BCVA),enophthalmos difference,recovery rate of eye movement disorder,recovery rate of diplopia,and incidence of postoperative complications.●RESULTS:The study included 95 cases(95 eyes),with 63 men and 32 *** patients’age ranged from 5 to 67y(35.21±15.75y).The new surgical group and the control group exhibited no statistically significant differences in the duration of the operation,BCVA and enophthalmos *** recovery rates of diplopia in the new surgical group were significantly higher than those in the control group at 1mo[OR=0.03,95%CI(0.01–0.15),Pnd 3mo[OR=0.11,95%CI(0.03–0.36),Pnt disorders at 1 and 3mo after surgery were OR=0.08,95%CI(0.03–0.24),Pnd OR=0.01,95%CI(0.00–0.18),Pncidence of postoperative complications was lower in the new surgical group compared to the control group[OR=4.86,95%CI(0.95–24.78),PnCLUSIOn:The combination of nnE and 3DPT can shorten the recovery time of diplopia and eye movement disorder in patients with OBF.

Cobalt selenide(CoSe_(2))has garnered considerable attention as a prospective anode candidate for advanced lithium-ion storage,prompting comprehensive ***,CoSe_(2)-based anodes usually suffer from significant volume variation upon lithiation leading to unsatisfactory cycling ***,a versatile synthesis route is proposed for the in-situ fabrication of CoSe2nanoparticles embedded in n-dope carbon skeleton(CoSe_(2)@nC)through annealing treatment and selenization of a metal–organic framework-derived(MOF)*** n-doped carbon derived from the MOF serves not only as an excellent conductive substrate but also as a confined reactor,effectively inhibiting the structural instability and alleviating the inevitable volume change of CoSe_(2).Owing to their unique nanostructure,the as-prepared CoSe_(2)@nC exhibits a high capacity of 745.9mAh·g^(-1)at 0.1 A·g^(-1),while maintaining excellent rate capability and an impressive ***,the assembled lithium-ion capacitor(LIC)based on CoSe_(2)@nC demonstrates an energy density of 130Wh·kg^(-1),a power density of 24.6 kW·kg^(-1),and remarkable capacity retention of 90.8%after 8000 *** results highlight the great potential of CoSe_(2)@nC for practical applications.

Metal tellurides(MTes) are highly attractive as promising anodes for high-performance potassium-ion batteries. The capacity attenuation of most reported MTe anodes is attributed to their poor electrical conductivity and large volume variation. The evolution mechanisms, dissolution properties, and corresponding manipulation strategies of intermediates(K-polytellurides, K-pTe_(x)) are rarely mentioned. Herein,we propose a novel structural engineering strategy to confine ultrafine CoTe_(2) nanodots in hierarchical nanogrid-in-nanofiber carbon substrates(CoTe_(2)@nC@nSPCnFs) for smooth immobilization of K-pTe_(x) and highly reversible conversion of CoTe_(2) by manipulating the intense electrochemical reaction process. Various in situ/ex situ techniques and density functional theory calculations have been performed to clarify the formation, transformation, and dissolution of K-pTe_(x)(K_(5)Te_(3) and K_(2)Te), as well as verifying the robust physical barrier and the strong chemisorption of K_(5)Te_(3) and K_(2)Te on S, n co-doped dual-type carbon substrates. Additionally, the hierarchical nanogrid-in-nanofiber nanostructure increases the chemical anchoring sites for K-pTe_(x), provides sufficient volume buffer space, and constructs highly interconnected conductive microcircuits, further propelling the battery reaction to new heights(3500 cycles at 2.0 A g^(-1)). Furthermore, the full cells further demonstrate the potential for practical applications. This work provides new insights into manipulating K-pTe_(x) in the design of ultralong-cycling MTe anodes for advanced PIBs.
