CCR5A32 mutation does not influence the susceptibility to HCV infection, severity of liver disease and response to therapy in patients with chronic hepatitis C

AIM： To study whether CCR5△32 mutation was associated with viral infection and severity of liver disease. METHODS： Two hundred and fifty two histologically proven, chronic HCV patients （mean age： 41±14 years; M/F： 164/88） were genotyped. PCR based genotyping of 32 bp deletion at the CCR5 locus was done. Fourhundred and eight matched healthy controls were studied to assess susceptibility to HCV infection. To assess correlation of immune gene polymorphism with severity of HCV related liver disease, patients with chronic HCV infection were divided into those with a fibrosis score of ≤ 2 （mild） or 〉 2 （severe） and histological activity index （HAI） of ≤ 5 or 〉 5. For correlation between CCR5△32 mutations and response to therapy, 129 patients who completed therapy were evaluated. RESULTS： The majority （89.4%） of the patients were infected with genotype 3. The frequency of homozygous CCR5△32 mutants was comparable to HCV patients as compared to the healthy controls （0.7% vs 0%, P = 0.1）. Further more, the frequency of CCR5△32 mutation was comparable in patients with mild or severe liver disease.（P = NS）. There was also no association observed with response to therapy and CCR5△32 mutation. CONCLUSION： CCR5△32 mutation does not have a role in disease susceptibility, severity or response to therapy in patients with chronic hepatitis C infection.

Midostaurin inhibits hormone-refractory prostate cancer PC-3 cells by modulating nPKCs and AP-1 transcription factors and their target genes involved in cell cycle

The development of prostate cancer from a clinically localized, hormone-naive state to a hormonerefractory phenotype involves a complex interplay of protein kinase C (PKC) and activator protein-1 (AP-1). Therefore, the present study aimed to uncover the roles of PKC and AP-1 through midostaurin-mediated regulation—a multi-target protein kinase inhibitor. Androgen Receptor-negative, hormone-refractory prostate cancer cells (PC-3) were used as an in-vitro model system. The effect of midostaurin on cell viability was assessed by an MTT assay. Expression studies on PKC-α, PKC-d, different AP-1 transcription factors, and AP-1 regulating genes were analyzed by semiquantitative RT-PCR, and protein levels of Bcl-2 were evaluated by western blotting. Midostaurin decreased the viability of hormone-refractory PC-3 cells. Furthermore, midostaurin significantly induced the transcripts of apoptotic-mediated PKC-d, tumor suppressor p53, cell cycle inhibitor p21cip1/waf1, death receptor TNF-α, pro-apoptotic Bax, and Caspase-8, and eventually inhibited the expression of pro-survival PKC-ε, pro-oncogene c-Jun, c-Fos, Fra-1, positive growth regulator cyclin D1, and anti-apoptotic Bcl-2. In addition, midostaurin also decreased the protein expression of anti-apoptotic Bcl-2. The present study provided evidence that midostaurin suppresses tumor growth and induces apoptosis in hormone-refractory PC-3 cells via modulation of PKC-d and PKC-ε expression, and regulation of PMA-altered c-Jun, c-Fos, and Fra-1 AP-1 transcription factors and their target genes involved in cell cycle regulation (cyclin D1, p53, p21, Bcl-2, and TNF-α). Thus, pharmacological targeting of PKC and AP-1 factors may have therapeutic potential against hormone-refractory prostate cancer.

Modeling transmission dynamics of lyme disease: Multiple vectors, seasonality, and vector mobility

Lyme disease is the most prevalent tick-borne disease in the United States,which humans acquire from an infected tick of the genus Ixodes(primarily Ixodes scapularis).While previous studies have provided useful insights into various aspects of Lyme disease,the tick's host preference in the presence of multiple hosts has not been considered in the existing *** this study,we develop a transmission dynamics model that includes the interactions between the primary vectors involved:blacklegged ticks(***),white-footed mice(Peromyscus leucopus),and white-tailed deer(Odocoileus virginianus).Our model shows that the presence of multiple vectors may have a significant impact on the dynamics and spread of Lyme *** on our model,we also calculate the basic reproduction number,R 0,a threshold value that predicts whether a disease exists or dies *** extensions of the model consider seasonality of the tick's feeding period and mobility of deer between *** results suggest that a longer tick peak feeding period results in a higher infection ***,while the deer mobility may not be a primary factor for short-term emergence of Lyme disease epidemics,in the long-run it can significantly contribute to local infectiousness in neighboring counties,which eventually reach the endemic steady state.

Full Outline of Unresponsiveness score and the Glasgow Coma Scale in prediction of pediatric coma

BACKGROUND: This study was done to compare the admission Full Outline of Unresponsiveness(FOUR) score and Glasgow Coma Scale(GCS) as predictors of outcome in children with impaired ***: In this observational study, children(5–12 years) with impaired consciousness of <7 days were included. Children with traumatic brain injury, on sedatives or neuromuscular blockade; with pre-existing cerebral palsy, mental retardation, degenerative brain disease, vision/hearing impairment; and seizure within last 1 hour were excluded. Primary outcomes: comparison of area under curve(AUC) of receiver operating characteristic(ROC) curve for in-hospital mortality. Secondary outcomes: comparison of AUC of ROC curve for mortality and poor outcome on Pediatric Overall Performance Category Scale at 3 ***: Of the 63 children, 20 died during hospital stay. AUC for in-hospital mortality for GCS was 0.83(CI 0.7 to 0.9) and FOUR score was 0.8(CI 0.7 to 0.9) [difference between areas –0.0250(95%CI 0.0192 to 0.0692), Z statistic 1.109, P=0.2674]. AUC for mortality at 3 months for GCS was 0.78(CI 0.67 to 0.90) and FOUR score was 0.74(CI 0.62 to 0.87)(P=0.1102) and AUC for poor functional outcome for GCS was 0.82(CI 0.72 to 0.93) and FOUR score was 0.79(CI 0.68 to 0.9)(P=0.2377), which were also comparable. Inter-rater reliability for GCS was 0.96 and for FOUR score ***: FOUR score was as good as GCS in prediction of in-hospital and 3-month mortality and functional outcome at 3 months. FOUR score had a good inter-rater reliability.

An inherent acceleratory effect of insulin on small intestinal transit and its pharmacological characterization in normal mice

AIM： To study an inherent effect of insulin on small intestinal transit and to explore involvement of various systems/mechanisms in normal mice. METHODS： Insulin at the doses of 2 μU/kg, 2 mU/kg, 2 U/kg or vehicle was subcutaneously administered to four groups of overnight fasted normal male mice. Blood glucose （BG） levels were measured 2 min before insulin administration and 2 min before sacrificing the animals for the measurement of small intestinal transit （SIT）. Charcoal meal was administered （0.3 mL） intragastrically 20 min after insulin administration and animals were sacrificed after 20 min and SIT was determined. For exploration of the various mechanisms involved in insulin-induced effect on SIT, the dose of insulin which can produce a significant acceleration of SIT without altering BG levels was determined. The following drugs, atropine （1 mg/kg）, clonidine （0.1 mg/kg）, ondansetron （1 mg/kg）, naloxone （5 mg/kg）, verapamil （8 mg/kg） and glibenclamide （10 mg/kg）, were administered intravenously 10 min prior to the administration of insulin （2 μU/kg）. RESULTS： The lower doses of insulin （2 μU/kg and 2 mU/kg） produced a significant acceleration of SIT from 52.0% to 70.7% and 73.5% without lowering blood glucose levels （P〈0.01）, while the highest dose of insulin （2 U/kg） produced a fall in blood glucose levels which was also associated with significant acceleration of SIT （P〈0.01）. After pretreatment of insulin （2 μU/kg） group with atropine, insulin could reverse 50% of the inhibition produced by atropine. In clonidine-pretreated group, insulin administration could reverse only 37% of the inhibition produced by clonidine and inhibition of SIT was significant compared with vehicle ＋ insulintreated group, i.e. from 74.7% to 27.7% （P〈0.01）. In ondansetron-pretreated group, insulin administration could produce only mild acceleration of SIT （23.5%）. Metathesis synthesis, characterization, spectral and photoactivity studies of Ln_(2/3)MoO_4(Ln=La, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Er and Y)

Rare earth ortho-oxomolybdates of composition Ln2/aMoO4 （Ln=La, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Er and Y） were prepared by metathesis reaction. All these materials were characterized by powder X-ray diffraction, Fourier transform-infrared spectroscopy （FT-IR） and UV-Vis diffuse reflectance spectra. The Rietveld analysis of all the samples was carried out to obtain the unit cell parameters and reliability factors. All the materials were crystallized in tetragonal lattice with I41/a space group. The infrared spectra of all the samples were characterized by bands due to MoO42 units. The bandgap energies of all the samples were obtained from UV-Vis DRS spectra. The luminescence spectra of Ln2/3MoO4 （Ln=Pr, Eu and Dy） showed characteristic transitions of the rare earth ions. The visible light photocatalytic activity of all the samples was studied against degradation of methylene blue.

IBIS-FM radar as a tool for monitoring hill slopes during excavations in hydropower projects:A case study from Bhutan Himalayas

Excavation is common in the construction of hydropower *** for both stable and unstable slopes is a pressing requirement during progressive excavations with or without cut-slope support *** present study is about an excavation carried out in a proposed concrete gravity dam site on the right bank slope of the Punatsangchhu River in western *** the excavation,the right bank abutment witnessed multiple events of slope failures of various *** major landslide occurred on 23 July 2013 in the toe of the right abutment,where foliation/multiple sheared/fractured zones/seams in quartz-feldspathic biotite gneiss dip towards the *** further excavations/piling works,a subsequent landslide on the downstream,within the body of the July 2013 landslide,occurred on 12 August *** a result,a real-time monitoring of the slope became necessary to facilitate further excavations for achieving the dam ***,the advanced slope monitoring instrument like Image By Interferometric Survey e Frequency Modulated(IBIS-FM)radar was deployed for monitoring the right bank slope during the excavation for the construction of~129 m high *** displacement of the hill slope is assessed based on this system by monitoring point locations as well as areas by assigning different threshold values for providing timely *** real-time monitoring was effective in identifying the reactivation of August 2016 landslide that occurred on 22 January ***,this study showcases the efficiency of IBIS-FM radar in monitoring slope instability with sub-millimeter accuracy on a near real-time basis.
