Background and Aims:African Americans (AA) historically have a low response rate to hepatitis C therapies,and there is limited information available for this patient population regarding the development and treatment of chronic hepatitis C (CHC).The aim of this study was to evaluate liver disease progression and hepatocellular carcinoma (HCC) development in AA with ***:Between 1995 and 2008,246 AA patients with CHC were identified from a database of patients and followed until 2012-2013 (average 8 years)or the development of HCC after ***:Viral clearance (intent to treat;sustained virus response (SVR)) was achieved in 15% of patients with interferon based therapies with or without *** patients who achieved an SVR (n=22) did not develop HCC or new onset cirrhosis,whereas the HCC incidence in untreated AA patients was 23% (51/203).Patients who achieved an SVR also had improved fibrosis,as defined by the AST Platelet Ratio Index (APRI) and Fibrosis-4 (FIB-4) score,relative to nonresponders and untreated ***:The severity of liver disease at the first visit (except for cirrhosis) correlated with the development of HCC,but because of the overlap in values between patients,these measurements were not useful for predicting individual *** cirrhosis at the first visit was not a predictive factor,treatment with newer antiviral therapies is the best option for reducing the incidence of advanced liver disease and its harmful outcomes in the AA population.

A simple, precise, accurate stability-indicating gradient reversed-phase high-performance liquid chromatographic （RP-HPLC） method was developed for the quantitative determination of zotepine （ZTP） in bulk and pharmaceutical dosage forms in the presence of its degradation products （DPs）. The method was developed using Phenomenex C18 column （250 mm ~ 4.6 mm i.d., 5 mm） with a mobile phase containing a gradient mixture of solvents, A （0.05%trifluoroacetic acid （TFA）, pH ？ 3.0） and B （acetonitrile）. The eluted compounds were monitored at 254 nm;the run time was within 20.0 min, in which ZTP and its DPs were well separated, with a resolution of 41.5. The stress testing of ZTP was carried out under acidic, alkaline, neutral hydrolysis, oxidative, photolytic and thermal stress conditions. ZTP was found to degrade significantly in acidic, photolytic, thermal and oxidative stress conditions and remain stable in basic and neutral conditions. The developed method was validated with respect to specificity, linearity, limit of detection, limit of quantification, accuracy, precision and robustness as per ICH guidelines. This method was also suitable for the assay determination of ZTP in pharmaceutical dosage forms. The DPs were characterized by LC-MS/MS and their fragmentation pathways were proposed.

自然界的复杂流体体系绝大部分由非球形颗粒构成。长期以来,理论和实验都从不同角度对这类体系的流变行为进行了详细探讨。理论上,Jeffrey对椭球体流变行为的理论计算开启了类似体系理论研究的先河,随后Brosema、Brenner等从不同角度对棒状颗粒构成的复杂流体进行了扩充。Batchleor则对这类体系的轴向拉升流场下的流变行为进行了研究。实验上,早期主要集中在表征不同长度的棒状纤维的流变行为,在一定程度上吻合了理论的预测。过去的十几年,人们对由无机颗粒如针状的赤铁矿、棒状的二氧化硅颗粒、碟状的黏土颗粒以及生物相关的棒状体系如F-actin等构成的模型体系进行了广泛深入的实验研究。随着纳米科学的兴起,碳纳米管构成的复合材料的流变行为是近期的一个研究热点。然而,目前为止的实验模型不可避免地具有尺寸不均一性和颗粒表面物化性质的不确定性等问题,导致实验结果的不可重复性以及与理论的不可比性。在本项目中,基于棒状病毒,结合基因控制和表面化学改性,构建了一类尺寸单一、刚性和表面性质可控的非球形颗粒的复杂流体体系,并对这类体系构成的凝胶进行了详细的流变表征,建立起微观结构与宏观流变行为之间的关系。将表面接枝有高分子刷的棒状病毒分散到甘油等高黏度的牛顿流体后,采用两种不同的技术手段对该体系的轴向拉升流场下的流变行为进行了研究,并与Batchleor等人的理论研究进行了比较。

Rare earth ortho-oxomolybdates of composition Ln2/aMoO4 （Ln=La, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Er and Y） were prepared by metathesis reaction. All these materials were characterized by powder X-ray diffraction, Fourier transform-infrared spectroscopy （FT-IR） and UV-Vis diffuse reflectance spectra. The Rietveld analysis of all the samples was carried out to obtain the unit cell parameters and reliability factors. All the materials were crystallized in tetragonal lattice with I41/a space group. The infrared spectra of all the samples were characterized by bands due to MoO42 units. The bandgap energies of all the samples were obtained from UV-Vis DRS spectra. The luminescence spectra of Ln2/3MoO4 （Ln=Pr, Eu and Dy） showed characteristic transitions of the rare earth ions. The visible light photocatalytic activity of all the samples was studied against degradation of methylene blue.

An inherent acceleratory effect of insulin on small intestinal transit and its pharmacological characterization in normal mice

AIM： To study an inherent effect of insulin on small intestinal transit and to explore involvement of various systems/mechanisms in normal mice. METHODS： Insulin at the doses of 2 μU/kg, 2 mU/kg, 2 U/kg or vehicle was subcutaneously administered to four groups of overnight fasted normal male mice. Blood glucose （BG） levels were measured 2 min before insulin administration and 2 min before sacrificing the animals for the measurement of small intestinal transit （SIT）. Charcoal meal was administered （0.3 mL） intragastrically 20 min after insulin administration and animals were sacrificed after 20 min and SIT was determined. For exploration of the various mechanisms involved in insulin-induced effect on SIT, the dose of insulin which can produce a significant acceleration of SIT without altering BG levels was determined. The following drugs, atropine （1 mg/kg）, clonidine （0.1 mg/kg）, ondansetron （1 mg/kg）, naloxone （5 mg/kg）, verapamil （8 mg/kg） and glibenclamide （10 mg/kg）, were administered intravenously 10 min prior to the administration of insulin （2 μU/kg）. RESULTS： The lower doses of insulin （2 μU/kg and 2 mU/kg） produced a significant acceleration of SIT from 52.0% to 70.7% and 73.5% without lowering blood glucose levels （P〈0.01）, while the highest dose of insulin （2 U/kg） produced a fall in blood glucose levels which was also associated with significant acceleration of SIT （P〈0.01）. After pretreatment of insulin （2 μU/kg） group with atropine, insulin could reverse 50% of the inhibition produced by atropine. In clonidine-pretreated group, insulin administration could reverse only 37% of the inhibition produced by clonidine and inhibition of SIT was significant compared with vehicle ＋ insulintreated group, i.e. from 74.7% to 27.7% （P〈0.01）. In ondansetron-pretreated group, insulin administration could produce only mild acceleration of SIT （23.5%）. Impedance (Z), and phase angle (θ) of a parallel-plate capacitor with a single cherry fruit between the plates were measured using a CI meter (Chari’s Impedance meter), at 1 and 9 MHz. Capacitance C was derived from Z and θ, and by using the C, θ, and Z values of a set of cherries whose moisture content (MC) values were later determined by the hot air-oven method, a calibration equation was developed. Using this equation, and their measured C, θ, and Z values, the MC of a group of cherries, not used in the calibration, was predicted. The predicted values were compared with their vacuum-oven values. The method worked well with an R2 value of 0.98 and a standard error of prediction (SEP) of 1.24, in the measured moisture range between 5% and 20%.

Background: The present study was conducted to examine the association between various coronary risk factors and clinical parameters, with special emphasis on ankle-brachial index, in predicting the severity and complexity of coronary artery disease. Methods: Patients diagnosed with coronary artery disease at our hospital between September-2012 and December-2014 were examined in this study. Selected patients were screened for cardiovascular risk factors including diabetes, hypertension, smoking, and alcohol habits as well as for clinical parameters including body-mass index, waist-hip ratio, ankle-brachial index, and ejection fraction. All patients underwent coronary angiography and were evaluated for severity of coronary artery disease (based on number of vessels involved) and complexity of coronary angiographic lesions (measured by computer-assisted Syntax score calculator). The collected data were analyzed to determine the role of cardiovascular risk factors and clinical parameters as predictors of complexity and severity of coronary artery disease. Results: A total of 211 patients (mean age: 54.64 ± 9.9 years;81% males) with coronary artery disease were analyzed. Findings revealed that diabetes mellitus (p < 0.001), hypertension (p < 0.001), smoking habits (p = 0.036), and low ankle-brachial index (p < 0.001) were independent predictors of complex coronary artery disease as measured by Syntax score. Significant associations were also evident between severity of coronary artery disease and diabetes mellitus (p < 0.001), hypertension (p < 0.001), and ankle-brachial index (p < 0.001). Conversely, other cardiovascular risk factors including body-mass index, alcohol habits, wait-hip ratio, and ejection fraction did not exhibit significant associations with severity and complexity of coronary artery.
