Gasdermin D-mediated pyroptosis in myocardial ischemia and reperfusion injury:Cumulative evidence for future cardioprotective strategies

Cardiomyocyte death is one of the major mechanisms contributing to the development of myocardial infarction(MI)and myocardial ischemia/reperfusion(MI/R)injury.Due to the limited regenerative ability of cardiomyocytes,understanding the mechanisms of cardiomyocyte death is necessary.Pyroptosis,one of the regulated programmed cell death pathways,has recently been shown to play important roles in MI and MI/R injury.Pyroptosis is activated by damage-associated molecular patterns(DAMPs)that are released from damaged myocardial cells and activate the formation of an apoptosisassociated speck-like protein containing a CARD(ASC)interacting with NACHT,LRR,and PYD domains-containing protein 3(NLRP3),resulting in caspase-1 cleavage which promotes the activation of Gasdermin D(GSDMD).This pathway is known as the canonical pathway.GSDMD has also been shown to be activated in a non-canonical pathway during MI and MI/R injury via caspase-4/5/11.Suppression of GSDMD has been shown to provide cardioprotection against MI and MI/R injury.Although the effects of MI or MI/R injury on pyroptosis have previously been discussed,knowledge concerning the roles of GSDMD in these settings remains limited.In this review,the evidence from in vitro,in vivo,and clinical studies focusing on cardiac GSDMD activation during MI and MI/R injury is comprehensively summarized and discussed.Implications from this review will help pave the way for a new therapeutic target in ischemic heart disease.

Adaptive predictor-based control for a helicopter system with input delays:Design and experiments

In this paper,we consider a 2-degrees-of-freedom(DOF)helicopter system subject to long input delays and uncertain system parameters.To address the challenges including unknown system parameters and input delays in control design,we develop an adaptive predictor-feedback control law to achieve trajectory tracking.Stability of the closed-loop system is further established,where the tracking errors are shown to converge towards zero.Through simulation and experiments on the helicopter system,we illustrate that tracking of a desired trajectory is achieved with the proposed control scheme.

Mountains are sometimes referred to as’sky islands’since their biodiversity may be geographically isolated at high altitudes in a sea of uninhabitable lowlands (first used by [1]).It is a nice expression because it gives an immediate picture of how mountain systems may behave like oceanic islands,especially when it comes to patterns of dispersal and colonization.

Nano-TRAIL:a promising path to cancer therapy

Tumor Necrosis Factor-Related Apoptosis-Inducing Ligand,also called apo-2 ligand(TRAIL/Apo-2L),is a cytokine that triggers apoptosis by binding to TRAIL-R1(DR4)and TRAIL-R2(DR5)death receptors.Apoptosis occurs through either the extrinsic or intrinsic pathway.The administration of recombinant human TRAIL(rhTRAIL)or TRAIL-receptor(TRAIL-R)agonists promotes apoptosis preferentially in cancerous cells over normal cells in vitro;this phenomenon has also been observed in clinical studies.The limited efficacy of rhTRAIL in clinical trials could be attributed to drug resistance,short half-life,targeted delivery issues,and off-target toxicities.Nanoparticles are excellent drug and gene delivery systems characterized by improved permeability and retention,increased stability and biocompatibility,and precision targeting.In this review,we discuss resistance mechanisms to TRAIL and methods to overcome TRAIL resistance by using nanoparticle-based formulations developed for the delivery of TRAIL peptides,TRAIL-R agonists,and TRAIL genes to cancer cells.We also discuss combinatorial approaches of chemotherapeutic drugs with TRAIL.These studies demonstrate TRAIL’s potential as an anticancer agent.

Analysis of the Effectiveness of Laparoscopic Appendectomy Compared to Laparotomy in the Treatment of Acute Appendicitis

Objective:To analyze the clinical effects of laparoscopic appendectomy compared to traditional laparotomy in treating acute appendicitis.Methods:90 patients with acute appendicitis were selected as research subjects.They were divided into a control group and an observation group,with 45 cases in each group.The control group underwent traditional laparotomy,while the observation group underwent laparoscopic appendectomy.The intraoperative indicators,postoperative recovery indicators,postoperative stress indicators,and postoperative complications of the two groups were compared.Results:The operative time of the observation group was longer,but the incision length was shorter and the blood loss was lesser(P<0.05);the observation group had shorter postoperative first gas-passing time,recovery of gastrointestinal function,ambulation time,and lower postoperative pain score.The observation group had lower postoperative stress index levels(P<0.05);the observation group had a lower postoperative complication rate(P<0.05).Conclusion:Aside from prolonging the operative time,laparoscopic appendectomy is more ideal than traditional laparotomy in all other indicators and has better therapeutic effects in treating acute appendicitis.

A topology-based bounded rationality day-to-day traffic assignment model

This paper analyzes the day-to-day adjustment process of users’behaviors in a transport network which is affected by relevant alterations such as disruptions due to critical events which cause the impossibility to use one or more links.For representing the progressive adjustment of the flows on the network to reach a new equilibrium,a dayto-day discrete-time model is proposed,based on the idea that people are bounded rational in their choices,i.***.they often do not behave according to the optimal solution but they accept solutions they consider satisfying.Users,in their choice process,are influenced by the topological similarity between the route they are currently using and others.This means that they tend to prefer the solutions that are more similar to the one they are already using.In parallel,users exhibit a myopic behavior,i.e.,they tend to overestimate the goodness of a route if,when using it,they suddenly experience a significant reduction in travel time compared to what they are used to.In the paper it is shown that such route choice behaviour implies that the steady state of the system corresponds to a Bounded Rational User Equilibrium,i.e.,a state that does not diverge from the user equilibrium more than a certain value which increases when the relative importance given to the topological similarity grows.The model also assumes that these biases vanish,at least with respect to those routes that are most frequently used by users,after a sufficient amount of time.Under certain conditions,it is then shown that the steady state can eventually collapse into a User Equilibrium.The effectiveness of the proposed model is assessed via simulation results in which two test networks are analyzed in detail to show the evolution of the users’behaviour in a transport network after a disruption.

[目的]为筛选出适合通辽地区推广种植的优质高产青贮玉米品种。[方法]选取22个青贮玉米品种在试验田内采取随机区组试验设计,对不同品种生育期、抗逆性(倒伏倒折率、瘤黑粉病、大斑病)、生物产量(鲜草产量、干草产量、籽粒产量)、农艺性状(株高、茎粗,绿叶数、枯叶数、有效穗数)和营养品质(淀粉、粗蛋白、中性洗涤纤维、酸性洗涤纤维)等进行综合比较分析。[结果]不同品种生育期不同,为98~122 d。抗倒伏倒折最强的是蒙青贮17272,倒伏倒折率为0.14%,瘤黑粉病发生率最低的是先玉1611和京科968,病害率为0。干物质产量最高的是先玉1710,为1 487.44 kg/667m2。先玉1620、先玉1611、金岭青贮377、先玉1710干物质含量都达到了30%以上,其淀粉含量也较高,均达到了35%以上,但中性洗涤纤维和酸性洗涤纤维含量均较低。不同品种粗蛋白含量差距不大,北农青贮368含量最高,为10.03%。[结论]先玉系列(1710、1620、1611)、金岭青贮系列(377、97)、蒙青贮(17272、266)、京九青贮16和京科968符合通辽地区在青贮玉米种植上的产量、品质和适应性的需求,适合在本区域大面积推广种植。

In order to support advanced vehicular Internet-of-Things(IoT)applications,information exchanges among different vehicles are required to find efficient solutions for catering to different application requirements in complex and dynamic vehicular environments.Federated learning(FL),which is a type of distributed learning technology,has been attracting great interest in recent years as it performs knowledge exchange among different network entities without a violation of user privacy.However,client selection and networking scheme for enabling FL in dynamic vehicular environments,which determines the communication delay between FL clients and the central server that aggregates the models received from the clients,is still under-explored.In this paper,we propose an edge computing-based joint client selection and networking scheme for vehicular IoT.The proposed scheme assigns some vehicles as edge vehicles by employing a distributed approach,and uses the edge vehicles as FL clients to conduct the training of local models,which learns optimal behaviors based on the interaction with environments.The clients also work as forwarder nodes in information sharing among network entities.The client selection takes into account the vehicle velocity,vehicle distribution,and the wireless link connectivity between vehicles using a fuzzy logic algorithm,resulting in an efficient learning and networking architecture.We use computer simulations to evaluate the proposed scheme in terms of the communication overhead and the information covered in learning.

Numerical Solutions for Heat Transfer of An Unsteady Cavity with Viscous Heating

The mechanism of viscous heating of a Newtonian fluid filled inside a cavity under the effect of an external applied force on the top lid is evaluated numerically in this exploration.The investigation is carried out by assuming a two-dimensional laminar in-compressible fluid flow subject to Neumann boundary conditions throughout the numerical iterations in a transient analysis.All the walls of the square cavity are perfectly insulated and the top moving lid produces a constant finite heat flux even though the fluid flow attains the steady-state condition.The objective is to examine the effects of viscous heating in the fully insulated lid-driven cavity under no-slip and free-slip Neumann boundary conditions coupled with variations in Reynolds and Prandtl numbers.The partial differential equations of time-dependent vorticity-stream function and thermal energy are discretized and solved using a self-developed finite difference code in MATLAB®environment.Time dependence of fluid thermodynamics is envisaged through contour and image plots.A commercial simulation software,Ansys Fluent®utilizing a finite element code is employed to verify the finite difference results produced.Although the effect of viscous heating is very minimal,Neumann no-slip and free-slip boundary conditions are able to trap the heat inside the fully insulated cavity as the heat flux is constantly supplied at the top lid.A lower Reynolds number and a greater Prandtl number with free-slip effects reduce temperature distribution in the cavity with a faster velocity than in the no-slip condition as the free-slip behaves as a lubricant.
