Determination of 23 β_2-agonists and 5 β-blockers in animal muscle by high performance liquid chromatography-linear ion trap mass spectrometry

A high performance liquid chromatography-linear ion trap mass spectrometry method using isotope dilution technique has been developed for the simultaneous determination of 23 β2-agonists and 5 β-blockers in animal muscle tissues. Pork and chicken muscle samples were acid hydrolyzed and extracted with 5% trichloracetic acid in water, and then cleaned up using MCX solid phase extraction (SPE) cartridge. Methanol and 0.1% formic acid were used as mobile phases for gradient elution. A Waters AtlantisT3 column was used for separation. ESI positive ion scan mode was used with selective reaction monitoring. 9 β2-Agonists labeled by the deuterium isotope were used as internal standards for quantification. The linear ranges of 23 β2-agonists and 5 β-blockers were 5-200 μg/L, the coefficient of correlation was not less than 0.995, and the limit of detection for each compound in the muscle tissue was below 0.2 μg/kg. The recoveries of each compound in the spiked samples at three levels 5, 10, 20 μg/kg were in the range of 47.3%-123.7%, and the relative standard deviations were in the range of 3.2%-25.7%. The developed method is sensitive and specific for the determination of β2-agonists and β-blockers in pork and chicken muscle samples.

＜正＞This paper investigates the development and evaluation of a robust control system for a wood drying kiln process incorporating decentralized variable structure control（DVSC） such that the moisture content of lumber will reach and be stabilized at the desired set point.A description of the dynamics of the wood drying process by means of the time-delay neural network is also presented,in which the back-propagation algorithm was implemented for testing, training and *** this identified model is used for simulation purpose and controller *** comparison purpose,a conventional proportional-integral-derivative（PID） controller is also employed and system performance is evaluated through *** results are evaluated to tune the controller parameters to achieve good performance in the wood-drying kiln and the DVSC strategy promises improved *** control system developed in this study may be applied in industrial wood-drying kilns,with a clear potential for improved quality and increased speed of drying.

Background In Shiqu County of the Qinghai-Tibet plateau,many wild and domestic mammals are involved in the transmission cycles of Echinococcus *** the adult stage,*** from the foxes is morphologically similar to *** addition,it is difficulty to distinguish ***,E. multilocularis and *** based on morphology of their *** and *** are major sources for human *** is necessary to establish some methods for identifying species. In this study,three species of Echinococcus genus(***,*** and ***) were characterized using a polymerase chain reaction-based restriction fragment length polymorphism technique (PCR-RFLP). Methods The complete 18S rRNA gene was amplified from individuals of each of the three species with PCR and then digested separately with one restriction endonuclease,and the fragments were separated by conventional agarose gel electrophoresis(PCR-RFLP). Results PCR-RFLP produced characteristic patterns for complete 18S rDNA sequences of *** and *** species,but no characteristic patterns were produced for ***. Conclusion The present study indicates the usefulness of the PCR-RFLP of 18S rDNA for systematic, epidemiological and diagnostic ***,this method offers an objective,simple and rapid approach for the discrimination of *** and *** and for study of other parasite complexes.

In this paper,considering the inertia of MRF,a novel method of setting up a 1D transient finite element model for a MR damper is proposed by flexibly using of the moving mesh technology,that is,describing the development of the pre-yield zone by the extension or contraction of the mesh of the post-yield ***,finite elements model is only necessarily built for the post-yield zone,and both the mass conservation in the damper’s gap and the kinetic equation of the pre-yield zone are implemented as the model’s *** this way,difficulties arising from the discontinuities of viscosities and the yield point’s dependence on the pressure are avoided when building a transient finite element *** illustrating example is included in the end of this paper and the agreements between steady values of transient force and results of well-known quasi-static model validate the correctness of the proposed *** also show that magnetic field response is the key factor affecting the transient damping force of a MR damper and the effect of inertia of MRF is negligibly small.

Porcine circovirus type 2 (PCV2) is the primary causative agent of post-weaning multisystemic wasting syndrome in pigs. To generate a genetic marker strain of PCV2, the full-length genome of the virus was amplified using PCR, and two copies of the genome were ligated in tandem to construct an infectious molecular clone. A Sal I restriction enzyme site was inserted into the clone as a genetic marker, and the recombinant plasmid was transfected into porcine kidney cells to generate mutant virus. The antigenicity of the recovered virus was confirmed by immunoperoxidase monolayer assay. The viral antigen was visualized in the nucleus and cytoplasm of the virus-infected cells. The viral genome could be differentiated from the wild-type parent by PCR and restriction fragment length polymorphism (PCR-RFLP). The mutant virus was stable on multiplication through 60 passages in cell culture;the highest titer reached was 106.6 TCID50/mL. Four 35-day-old unvaccinated piglets were inoculated with the virus by the intranasal and intravenous routes. Two of the virus-infected pigs developed high temperatures, progressive weight loss, and swollen lymph nodes. The viral antigen and nucleic acid were detected in numerous tissues of the pigs. The results indicate that the genetic marker strain should be a useful tool in studies on pathogenesis, vaccination, and molecular diagnosis of PCV2.

Porcine teschovirus 1 (PTV-1) (Swine/CH/IMH/03) was isolated from piglets in a farm in Inner Mongolia Province, P.R. china. It was confirmed by electron microscopy, RT-PCR, and sequencing. Comparison of the sequences of the amino acid and nucleotides and phylogenetic analysis of the polyprotein showed that PTV Swine/CH/IMH/03 strain is PTV-1. The isolated virus has closest relationship with Talfan strain, they shared 98.9 % and 99.5 % homology of amino acids and nucleotides, respectively, in the ORF of polyprotein. To our knowledge, this is the first report about isolation and identification of a PTV in china.

A licensed vaccine is currently not available against serotype A foot-and-mouth disease(FMD) in china since A/WH/CHA/09 was isolated in 2009,partly because it does not replicate well in BHK *** this end,a novel plasmid-based reverse genetics system was employed to construct a chimeric strain by replacing the nearly complete P1 gene in the vaccine strain O/CHA/99 with that in the epidemic stain A/WH/CHA/*** chimeric vius displayed similar growth kinetics with its parental strains and was chosen as the vaccine candidate strain after 12 passages in BHK cells. Subsequently,immunization with the inactivated vaccine was conducted in cattle and humoral immune responses in most of vaccinated cattle were induced at day *** result of virus challenge by A/WH/CHA/09 at day 28 indicates that the group vaccinated by a 10μg dose was fully protected without sub-clinic *** together,our data demonstrate the chimeric vius with O/CHA/99 not only breaks through the bottleneck of propagation in BHK cells compared with A/WH/CHA/09,provides excellent antigenic matching against serotype A FMD,but also is also a potential marker vaccine to distinguish infection and vaccination,which suggests reverse genetics technology is a potential tool to engineer a vaccine candidate for FMD prevention and control.
