Modeling the intracellular dynamics for Vif-APO mediated HIV-1 virus infection

The viral infectivity factor (Vif) was found to be essential for controlling HIV-1 virus infectivity. It targets cellular antiviral proteins in APOBEC family (APO) to trigger its fast degradation and inhibits APO packaging into nascent virion. In the present study, we propose a mathematical model to quantitatively study the intracellular dynamics of these typical virus-host interactions. Four sets of published experimental data were compared with simulation results to justify the model. Systematic parameter sensitivity and perturbation analysis showed that parameters related to APO are crucial to the infectivity of newly synthesized HIV-1 virus. Interestingly, we discovered that the synthesis rate of the viral structure protein Gag and the required number per nascent virion are optimized to achieve high virion production with minimal level of packaged APO, and large portion of model parameters are beneficial to virus only within a relatively small range. Furthermore, minor variations in several parameters related to viral protein Tat, the activator of viral RNA synthesis, were found to induce switch-like behaviors on both incorporated Vif and APO. These findings may provide new insights for understanding the high mutation rate of HIV-1 virus and its latency, as well as help identify key targets for therapeutic design.

Characteristics and evolution of strike-slip tectonics of the Liaohe Western Sag,Bohai Bay Basin

Because of its rich oil and gas resources and the special tectonic location of the Liaohe Western Sag （the Tanlu Fault traverses the sag）, Bohai Bay Basin, a detailed study of its strike-slip tectonics is significant in revealing the sag＇s tectonic evolution, its control on hydrocarbon accumulation, and the activity history of the northern section of the Tanlu Fault in the Cenozoic. Through systematic structure analysis of 3D seismic data of the Liaohe Western Sag, combined with balanced section analysis, a variety of structural features in relation to right-lateral strike-slip faults, such as echelon normal faults, ＂comb＂ structure, ＂flower＂ structure,＂interpretable＂ and ＂buried＂ strike-slip faults have been revealed exist in the Liaohe Western Sag. According to the research in this paper, the complex structural phenomena in the Liaohe Western Sag could be reasonably interpreted as right-lateral strike-slip activity and the strike-slip activities of the Liaohe Western Sag began in the early Oligocene. The activity was weak at the beginning （E3s1-2., then strengthened gradually and reached its strongest level in the late Oligocene （E3d1）. In the Miocene, the strike-slip activity was low and then strengthened significantly once again from the Pliocene to the present. It is speculated that the entire northern section of the Tanlu Fault has had a similar evolution history since the Oligocene.

Application of cathodoluminescence to zircon in FEG-ESEM

Cathodoluminescence (CL) image analysis of zircon is a key step of mineral micro-area compositional investigation and U-(Th)-Pb dating. We introduce a system consisting of field emission ESEM and high performance cathodoluminescence. This system has obvious advantages in imaging resolution and CL spectrum. We acquired some zircon CL images and spectra to demonstrate the ability of this system. Our results show that this system has a great potential application to characterize and identify the genesis of zircon.

Direct Synthesis and Spectrum analysis of CeO_2.Nanoparticles Deposited on Carbon Nanotubes

A novel method of direct synthesis of CeO2.nanoparticles onto multi-walled carbon nanotubes （MWNTs） was developed with advantages of simplicity, ease of scale-up, and low costs. The size of CeO2.particles deposited on the MWNTs was less than 6 nm. SEM and TEM were em-ployed to analysis the CeO2.coated MWNTs, and the properties of FTIR spectrum and UV-vis ab-sorption spectrum were investigated. The functional groups on the MWNTs obtained by nitric acid treatment play an important role on the deposition of the CeO2.particles. The carbon nanotubes possess broadened UV absorption function after being coated with CeO2.nanopartilces.

The concept of hydrocarbon accumulation in network was presented on basis of the overall analysis of the formation and evolution characteristics of the continental faulted basin and of the systemic re-search on the major controlling factors on the hydrocarbon accumulation. The hydrocarbon accumu-lation in network can be defined as hydrocarbon accumulation in a three-dimensional network system which is constituted by the hydrocarbon migration passages under multiple dynamics,following the hydrocarbon generation from source rocks. The research shows that the hydrocarbon accumulation in network is composed of four elements,i.e.,hydrocarbon source (source rock kitchen),hydrocarbon accumulation terminal (trap),network pathway connecting source and terminal (transporting system),and network potential driving hydrocarbon migration in the network pathway (migration dynamics). Compared with other networks,hydrocarbon accumulation in network has three basic characteristics: the irreversible geological process of material and information flow in the network; the loss of material and information in the flow process in the network; the multiple dynamics in the flow process. Interac-tion of all the elements in the geological process can be called hydrocarbon accumulation in network. There are three basic models for hydrocarbon accumulation in network,that is,hydrocarbon accumu-lation in the network source area,hydrocarbon accumulation in the network pathway,and hydrocarbon accumulation in the network terminal. The key in the application of the hydrocarbon accumulation models in network in practice is to confirm the major accumulation stage and the function range of the four elements controlling the hydrocarbon firstly,to predict the profitable accumulation region by su-perposition of the favorable areas confirmed by four elements consequently,and to evaluate the oil-bearing property of the trap as well as confirm drilling targets. Experiments and sensitivity analyses for heat transfer in a meter-scale regularly fractured granite model with water flow

Experiments of saturated water flow and heat transfer were conducted for a meter-scale model of regularly fractured granite. The fractured rock model （height 1502.5 ram, width 904 mm, and thickness 300 mm）, embedded with two vertical and two horizontal fractures of pre-set apertures, was constructed using 18 pieces of intact granite. The granite was taken from a site currently being investigated for a high-level nuclear waste repository in china. The experiments involved different heat source temperatures and vertical water fluxes in the embedded fractures either open or filled with sand. A finite difference scheme and computer code for calculation of water flow and heat transfer in regularly fractured rocks was developed, verified against both the experimental data and calculations from the TOUGH2.code, and employed for parametric sensitivity analyses. The experiments revealed that, among other things, the temperature distribution was influenced by water flow in the fractures, especially the water flow in the vertical fracture adjacent to the heat source, and that the heat conduction between the neighboring rock blocks in the model with sand-filled fractures was enhanced by the sand, with larger range of influence of the heat source and longer time for approaching asymptotic steady-state than those of the model with open fractures. The temperatures from the experiments were in general slightly smaller than those from the numerical calculations, probably due to the fact that a certain amount of outward heat transfer at the model perimeter was unavoidable in the experiments. The parametric sensitivity analyses indicated that the tem- perature distribution was highly sensitive to water flow in the fractures, and the water temperature in the vertical fracture adjacent to the heat source was rather insensitive to water flow in other fractures.

In this paper,the technology of wave process method for sedimentation is first adopted in the research of the foreland thrust fault of Micang Mountain with respect of oil and reservoir’s formation and tectonic and sedimentary evolution. From the fluctuation characteristics,we could make conclusions in the foreland thrust belt of Micang Mountain that,there existed 2.first-order sedimentary cycles (2.0 Ma),corresponding to Caledonian-Hercynian and Indo-Chinese-Yanshan-Himalayan tectonic cycles respec-tively; there existed 4 second-order sedimentary cycles (10 Ma),corresponding to two sedimentation peak period and two denudation peak periods in research zone; there existed 12.third-order sedimen-tary cycles (35 Ma) and 2. fourth-sedimentary cycles (2. Ma). These 33 cycles in the research zone corresponded to the sedimentation-denudation process in different periods,furthermore,their fluctua-tion characteristics bore the genetic relationship with the development law of source,reservoir and cap rocks: the source rock had the tendency to develop at the turning part between wave crest and wave trough,or at the superposition of wave turning part in different periods,presenting like "X"; most res-ervoir rocks developed at the place of wave peak; the development of cap rock was located in the wave trough on the right of sedimentation-denudation datum line. As a result,through the application of wave process method for sedimentation,we could rediscover the understanding of the tectonic and sedimentary evolution from another prospective,meanwhile,it enables to make prediction about the development rule of source,reservoir and cap rocks,which means a significant importance to the re-search of oil and reservoir’s forming condition.

Glucose-6-phosphate dehydrogenase (G6PDH,EC 1.***.1.49) is the first and main regulated enzyme of oxidative pentose phosphate pathway (OPPP),catalyzing the conversion of glucose-6-phosphate to 6-phospho-gluconolactone and playing important roles in the growth and development of plants. It is preciously reported that the enhancement of freezing resistance of Populus suaveolenscuttings is clear related to the distinct increase in cytosolic G6PDH activity. Here,a 1697 bp cDNA fragment (PsG6PDH) is amplified by RT-PCR from cold-induced total RNA of the freezing-tolerant P. suaveolens. A sequence analysis showed that PsG6PDH coding region had 1 530 bp and encoded 510 predicted amino acid residues. Genomic Southern analysis revealed that the isoform is encoded by a few copies of the gene in the poplar genome. The cloned gene PsG6PDHis cloned into binary vector pBI12. and used to transform tobacco. PCR and Southern blotting results verified integration of this gene into the genome of tobacco. Moreover,cold treatment experiments and membrane defense enzymeactivity analysis confirmed that overexpression of the PsG6PDHgene could enhance the tolerance to cold or frigid stresses in transgenic plants.

The onset and advance of the Asian summer monsoon

Based on the daily outgoing longwave radiation (OLR) data from national Oceanic and At- mospheric Administration (NOAA) satellites, the Cli- mate Prediction center’s merged {2. of pre- cipitation (CMAP) data and the national centers for Environmental Prediction/national center for At- mospheric Research (NCEP/NCAR) re{2. dataset, the mean intraseasonal variability of the Asian summer monsoon (ASM) is investigated by using power spectrum {2., band-pass filter, and diagnostic analyses. The processes of the onset and advance of monsoon over the southern part of Indo- china Peninsula, the east coast of Bay of Bengal, the South china Sea and the Indian subcontinent are explored. It is found that there is an abrupt change in OLR, precipitation and zonal wind during the onset and advance of the ASM. It is also indicated that the southern part of Indochina Peninsula and the adja- cent Andaman Sea is the region where the earliest onset of the ASM occurs in the 2.d pentad of May.
