Background:since the 1990s,evidence has accumulated of an increased prevalence of epilepsy in onchocerciasisendemic areas in Africa as compared to onchocerciasis-free *** the causal relationship between onchocerciasis and epilepsy has yet to be proven,there is likely an *** we discuss the need for disease burden estimates of onchocerciasis-associated epilepsy(OAE),provide them,detail how such estimates should be refined,and discuss the socioeconomic impact of OAE,including a cost-estimate for anti-epileptic *** body:Providing OAE burden estimates may aid prevention of epilepsy in onchocerciasis-endemic areas by inciting and informing collaboration between onchocerciasis control programmes and mental health *** not only massively impacts the health of those affected,but it also carries a high socioeconomic burden for the households and communities *** used previously published geospatial estimates of onchocerciasis in Africa and a separately published logistic regression model quantifying the association between onchocerciasis and epilepsy to estimate the number of OAE *** then applied disability weights for epilepsy to quantify the burden in terms of years of life lived with disability(YLD)and estimate the cost of *** estimate that in 2015 roughly 117000 people were affected by OAE across onchocerciasis-endemic areas previously under the African Programme for Onchocerciases control(APOC)mandate where OAE has ever been reported or suspected,and another 264000 persons in onchocerciasis-endemic areas where OAE has never been investigated *** total number of YLDs due to OAE was 39300 and 88700 in these areas respectively,based on a weighted mean disability weight of *** burden of OAE is approximately 13%of the total YLDs attributable to onchocerciasis and 10%of total YLDs attributable to *** estimated that by 2015 the total costs of treatment with antiepileptic drug for OAE cases wou

Background:Pulmonary sarcomatoid carcinoma(PsC)is a rare and aggressive subtype of non-small cell lung cancer(NsCLC),characterized by the presence of epithelial and sarcoma-like *** molecular and immune landscape of PsC has not been well ***:Multiomics profiling of 21 pairs of PsCs with matched normal lung tissues was performed through targeted high-depth DNA panel,whole-exome,and RNA *** describe molecular and immune features that define subgroups of PsC with disparate genomic and immunogenic features as well as distinct clinical ***:In total,27 canonical cancer gene mutations were identified,with TP53 the most frequently mutated gene,followed by ***,most TP53 and KRAs mutations were earlier genomic events mapped to the trunks of the tumors,suggesting branching evolution in most PsC *** identified two distinct molecular subtypes of PsC,driven primarily by immune infiltration and *** Immune High(IM-H)subtype was associated with superior survival,highlighting the impact of immune infiltration on the biological and clinical features of localized ***:We provided detailed insight into the mutational landscape of PsC and identified two molecular subtypes associated with ***-H tumors were associated with favorable recurrence-free survival and overall survival,highlighting the importance of tumor immune infiltration in the biological and clinical features of PsCs.

Early efforts to classify Mortierellaceae were based on macro-and micromorphology,but sequencing and phylogenetic studies with ribosomal DNA(rDNA)markers have demonstrated conflicting taxonomic groupings and polyphyletic *** some taxonomic confusion in the family has been clarified,rDNA data alone is unable to resolve higher level phylogenetic relationships within *** this study,we applied two parallel approaches to resolve the Mortierel-laceae phylogeny:low coverage genome(LCG)sequencing and high-throughput,multiplexed targeted amplicon sequenc-ing to generate sequence data for multi-gene *** then combined our datasets to provide a well-supported genome-based phylogeny having broad sampling depth from the amplicon *** the Mortierellaceae phylogeny into monophyletic genera resulted in 13 genera,7 of which are newly ***-coverage genome sequencing proved to be a relatively cost-effective means of generating a high-confidence *** multi-gene phylogenetics approach enabled much greater sampling depth and breadth than the LCG approach,but has limitations *** present this work to resolve some of the taxonomic confusion and provide a genus-level framework to empower future studies on Mortierellaceae diversity and evolution.

The high occurrence rate and difficulties in symptom control are listed as the major problems of oral mucosal disease by medical *** the development of oral mucosal lesions,the oral microenvironment changes,immunity declines,and continuous bacterial stimulation causes wound *** antibacterial drugs are ineffective for oral mucosal *** overcome this problem,a light-responsive antibacterial hydrogel containing sustained-release BMsCs was inspired by the trauma environment in the oral cavity,which is different from that on the body surface since it mostly remains under dark *** the absence of light,the hydrogel seals the wound to form a barrier,exerts a natural bacteriostatic effect,and prevents invasion by foreign ***,mesenchymal stem cells are presented,and the released growth factors and other substances have excellent anti-inflammatory and angiogenic effects,which result in rapid repair of the damaged *** light conditions,after photo-induced shedding of the hydrogel,RuB_(2)A exerts an antibacterial effect accompanied by degradation of the *** in a rat oral mucosal repair model demonstrate that DCs-RuB_(2)A_(2)-BMsCs could rapidly repair the oral mucosa within 4 *** data provide ideas for further analysis of the intrinsic molecular mechanisms and signaling *** together,our results suggest that this light-responsive antibacterial hydrogel loaded with BMsCs can be used for rapid wound repair and may advance the development of therapeutic strategies for the treatment of clinical oral mucosal defects.

Genome mapping has promised much to tree fruit breeding during the last 10 ***,one of the greatest challenges remaining to tree fruit geneticists is the translation of trait loci and whole genome sequences into diagnostic genetic markers that are efficient and cost-effective for use by breeders,who must select genetically optimal parents and subsequently select genetically superior individuals among their *** take this translational step,we designed the apple International RosBREED sNP Consortium OpenArray v1.0(IRsCOA v1.0)assay using a set of 128 apple single nucleotide polymorphisms(sNPs)linked to fruit quality and pest and disease resistance trait *** Thermo Fisher scientific OpenArray®technology enables multiplexed screening of sNP markers using a real-time PCR instrument with fluorescent probe-based Taqman®*** validated the apple IRsCOA v1.0 multitrait assay by screening 240 phenotyped individuals from the Plant&Food Research apple cultivar breeding *** set of individuals comprised commercial and heritage cultivars,elite selections,and families segregating for traits of importance to *** total,33 sNP markers of the IRsCOA v1.0 were validated for use in marker-assisted selection(MAs)for the scab resistances Rvi2/vh2,Rvi4/vh4,Rvi6/vf,fire blight resistance MR5/RLP1,powdery mildew resistance Pl2,fruit firmness,skin colour,flavour intensity,and *** availability of this set of validated trait-associated sNP markers,which can be used individually on multiple genotyping platforms available to various apple breeding programmes or re-designed using the flanking sequences,represents a large translational genetics step from genomics to crop improvement of apple.

Roses,which have been cultivated for at least 5000 years,are one of the most important ornamental crops in the *** of the interspecific nature and high heterozygosity in commercial roses,the genetic resources available for rose are *** effectively identify markers associated with QTL controlling important traits,such as disease resistance,abundant markers along the genome and careful phenotyping are *** genotyping by sequencing technology and the strawberry genome(Fragaria vesca v2.0.a1)as a reference,we generated thousands of informative single nucleotide polymorphism(sNP)*** sNPs along with known bridge simple sequence repeat(ssR)markers allowed us to create the first high-density integrated consensus map for diploid *** maps were first created for populations J06-20-14-3×“Little Chief”(J14-3×LC),J06-20-14-3×“vineyard song”(J14-3×vs)and“Old Blush”×“Red Fairy”(OB×RF)and these maps were linked with 824 sNPs and 13 ssR bridge *** anchor ssR markers were used to determine the numbering of the rose linkage *** diploid consensus map has seven linkage groups(LGs),a total length of 892.2 cM,and an average distance of 0.25 cM between 3527 *** combining three individual populations,the marker density and the reliability of the marker order in the consensus map was improved over a single population *** synteny between the strawberry and diploid rose genomes was *** consensus map will serve as the tool for the discovery of marker–trait associations in rose breeding using pedigree-based *** high level of conservation observed between the strawberry and rose genomes will help further comparative studies within the Rosaceae family and may aid in the identification of candidate genes within QTL regions.

Cultivated potato is a clonally propagated autotetraploid species with a highly heterogeneous *** assemblies of six cultivars including two chromosome-scale phased genome assemblies revealed extensive allelic diversity,including altered coding and transcript sequences,preferential allele expression,and structural variation that collectively result in a highly complex transcriptome and predicted proteome,which are distributed across the homologous *** species contribute to the extensive allelic diversity in tetraploid cultivars,demonstrating ancestral introgressions predating modern breeding *** a clonally propagated autotetraploid that undergoes limited meiosis,dysfunctional and deleterious alleles are not purged in tetraploid *** a quarter of the loci bore mutations are predicted to have a high negative impact on protein function,complicating breeder’s efforts to reduce genetic *** stCDF1 locus controls maturity,and analysis of six tetraploid genomes revealed that 12 allelic variants of stCDF1 are correlated with maturity in a dosage-dependent *** of the complexity of the tetraploid potato genome with its rampant structural variation and embedded deleterious and dysfunctional alleles will be key not only to implementing precision breeding of tetraploid cultivars but also to the construction of homozygous,diploid potato germplasm containing favorable alleles to capitalize on heterosis in F1 hybrids.
