<i>In Vitro</i>Effect of Soy Isoflavone and Equol on Soluble CD40L Release Stimulated by Ristocetin in Platelets from Postmenopause Women

The inhibition of specific flavonoid on the in vitro platelet aggregation induced by collagen, arachidonic acid and thromboxane A2 (TxA2) agonist, seems to be related mostly to their ability to compete for binding to the TxA2 receptor (TP). The aim of this study was to analyze the effect of soy isoflavone and equol in terms of inhibiting the platelet aggregation and sCD40L release stimulated by ristocetin, an in vitro-activator of glycoprotein Ib/IX/V, in platelets from postmenopausal women. When platelets were stimulated by 0.75 mg/ml ristocetin, equol (10 μm) exhibited a greater inhibitory activity on platelet aggregation (~68%) than genistein or daidzein. The effect of equol was dependent on the concentration of platelet aggregation agonist. In the presence of ristocetin (0.75 mg/ml, 1.125 mg/ml and 1.5 mg/ml), the inhibitory effect of 10 μm equol was 68% ± 5%, 54% ± 4% and 31% ± 5%, respectively. Equol (10 μm) was a potent inhibitor (~35%) of sCD40L release when stimulated with 1.5 mg/ml ristocetin. However, no significant differences were noted in platelets incubated in the presence of genistein or daidzein and stimulated by ristocetin. On the other hand, SQ29548, a high TP antagonist, also inhibited the sCD40L release stimulated by ristocetin. Finally, 10 μm of genistein, daidzein or equol did not significantly affect the thromboxane B2 production when platelets were incubated with 1.5 mg/ml ristocetin. The relevance of this study was to find that equol exhibits a potent activity by inhibiting ristocetin-induced sCD40L release, suggesting that soy isoflavone has important biological effects on the hemostatic system. However, clinical trials will be necessary to assess the effect of equol on platelet and their impact on inflammatory markers.

mucopolysaccharidosis type II （mPS II） is a rare X-linked disorder caused by alterations in the iduronate-2-sulfatase （IDS） gene. In this study, IDS activity in peripheral mononuclear blood monocytes （PmBCs） was measured with a fluorimetric enzyme assay. Urinary glycosaminoglycans （GAGs） were quantified using a colorimetric assay. All IDS exons and intronic flanks were bidirectionally sequenced. A total of 15 mutations （all exonic region） were found in 17 mPS II patients. In this cohort of mPS II patients, all alterations in the IDS gene were caused by point nucleotide substitutions or small deletions. mutations ***88His and ***172＊ occurred twice. All mu- tations were inherited except for ***489Alafs＊7, a germline mutation. We found four new mutations （***142Phe, ***233Gly, ***430＊, and ***360Tyrfs＊31）. In Epstein-Barr virus （EBV）-immortalized PmBCs derived from the mPS II patients, no IDS protein was detected in case of the ***142Phe and ***360Tyrfs＊31 mutants. For ***233Gly and ***430＊, we observed a residual expression of IDS. The ***233Gly and ***430＊ mutants had a residuary enzymatic activity that was lowered by 14.3 and 76-fold, respectively, compared with healthy controls. This observation may help explain the mild disease phenotype in mPS II patients who had these two mutations whereas the ***142Phe and ***360Tyrfs＊31 mutations caused the severe disease manifestation.

Low-Temperature Peculiarities of Electron Transport Properties of Carbon Nanotubes

The large dispersion of data on electrical resistivity (from 10-4 ?·m to 20 ?·m) and low-temperature peculiarities of electrical resistivity (decreasing when temperature increases) in different carbon nanotubes (CNTs) are shown to be described by the classical Drude low with the electron relaxation time calculated by the quantum mechanical techniques, taking into account multiple elastic scattering of electrons by impurities and structural irregularities which are formed in nanotubes during their synthesis. In the frame of our approach it is also possible to describe the wide spread of magnitudes of thermal conductivity, its temperature dependence and to understand why thermopower of these systems does not have any dispersion in magnitude for different CNTs. For resistance and thermal conductivity the magnitude of carrier concentration is determinant but for thermopower it is not important.

Expression of Acyl-lipid 12-desaturase Gene in Prokaryotic and Eukaryotic Cells and Its Effect on Cold Stress Tolerance of Potato

We report the expression profile of acyl-lipid △12-desaturase （desA） gene from Synechocystis sp. PCC6803 and its effect on cell membrane lipid composition and cold tolerance in prokaryotic （Escherichia coil） and eukaryotic （Solanum tuberosum） cells. For this purpose, a hybrid of desA and reporter gene encoding thermostable lichenase （licBm3） was constructed and used to transform these cells. The expression of this hybrid gene was measured using qualitative （Petri dish test, electrophoregram and zymogram） and quantitative methods （spectrometry and gas liquid chromatography assays）. The maximum level of linoleic acid in the bacterial cells containing hybrid gene was 1.9% of total fatty acids. Cold stress tolerance assays using plant damage index and growth parameters showed that cold tolerance was enhanced in primary transgenic lines because of increased unsaturated fatty acid concentration in their lipids. The greatest content of 18：2 and 18：3 fatty acids in primary transgenic plants was observed for lines 2 （73%） and 3 （41%）. Finally, our results showed that desaturase could enhance tolerance to cold stress in potato, and desaturase and lichenase retain their functionality in the structure of the hybrid protein where the enzymatic activity of target gene product was higher than in the case of reporter lichenase gene absence in the construction.

Objective:To determine an algorithm for molecular diagnosis of visceral leishmaniasis(VL)by kinetoplast DNA(kDNA)(RV1/RV2)and internal transcriber spacer(ITS1)(LITSR/L5.8 S)polymerase chain reaction(PCR),complemented by ITS 1 PCR restriction fragment length polymorphism(RFLP),using peripheral blood or bone marrow aspirate from patients with suspected ***:Biological samples were submitted to the gold standard for the diagnosis of VL and molecular diagnosis represented by ITS 1 PCR,kDNA PCR,and ITS 1 PCR *** samples were obtained from seven groups:groupⅠ,82 samples from patients with confirmed VL;groupⅡ,16 samples from patients under treatment for VL;groupⅢ,14 samples from dogs with canine visceral leishmaniasis(CVL);groupⅣ,a pool of six experimentally infected sandflies(Lutzomya longipalpis);group V,18 samples from patients with confirmed tegumentary leishmaniasis(TL)and groupsⅥandⅦwere from control groups without ***:The following gold standard and molecular examination results were obtained for each of the seven groups:groupⅠ:parasitologic and immunochromatographic tests showed a sensitivity of 76.3%(61 of 80)and 68.8%(55 of 80),respectively,and a sensitivity of 97.6%(80 of 82)and 92.7%(76 of 82)by ITS1 and kDNA PCR,*** ITS1 PCR RFLP(HaeⅢ)analysis of the 80 positive samples,52.5%(42 of 80)generated three fragments of 180,70,and 50 bp,corresponding to the pattern of Leishmania infantum infantum;groupⅡ:negative for the parasitologic methods and positive for IrK39(100%,16 of 16),presented 12.5%(2 of 16)of positivity by ITS 1 PCR and 25.0%(4 of 16)by kDNA PCR;groupⅢ:positive in the parasitologic and serologic tests(100%,14 of 14),presented 85.7%(12 of 14)of positivity by ITS1 PCR and kDNA ***1 PCR RFLP showed that 83.3%(10 of 12)of the canine samples contained parasites with profiles similar to ***;groupⅣpresented amplifications by ITS1 PCR and kDNA ***1 PCR products were analyzed by RFLP,generating a profile similar to

Peculiarities of Calculating Bridges with Seismic Isolation Including Spherical Bearings and Hydraulic Dampers in Russia

The peculiarities of calculating isolated structures with spherical bearings are analyzed in this paper. Some of peculiarities are caused by the lack of data at the moment when engineering solutions had to be made, Other peculiarities are connected with physical peculiarities of the device behaviour. To provide the analysis of structure hehaviour under the condition of the lack of input information, two types of design models of seismic protection devices were considered. They are the dampers linearization and the modelling of real dampers by dry friction ones. The dampers linearization makes it possible to use the existing software for calculating linear strongly-damped systems. To calculate structures with dry friction dampers, a new software was worked out. In this case, the structure is described as a piecewise-linear system of a relay-type. The investigations of the structure oscillations take into account both horizontal and vertical components of earthquake input. Under this condition, horizontal oscillation equations of structures are the maRie-Hill ones. The input and structure parameters which caused the structure instability are estimated. To exclude the structure instability, high damping devices should be used. These methods were used for seismic resistant analysis of bridges with spherical bearings and hydraulic dampers applied in Sochi.

Bioremediation of Oil-Polluted Cultivated Soils in the Euro-Arctic Region

It has been studied restoration processes in oil products-polluted soils at high northern latitudes （the murmansk region, Russia）. mineral and organic fertilizers and a bacterial preparation （based on the local strains of hydrocarbon-oxidizing bacteria） were applied for restore polluted soils. Periods of removing OP （oil products） from soil were determined by the reduction of the pollutant concentration and by soil biological activities--the dynamics of bacteria number and CO2 emission from soil. The soil OP even at such a high concentration （as 10 L/m^2） had stimulated bacterial reproduction. In three summer month levels in the control variant without ameliorators of OP content decreased by 59% from the initial level, in the variant with mineral and organic fertilizers by 86%, in the variant with the bacterial preparation by 84%. Stimulating of indigenous microorganisms activity with additional nutrients was no less effective technique of OP-polluted soil bioremediation, than applying the bacterial preparation, which requires considerable financial investment. moderately contaminated of OP soil is a source of additional carbon dioxide emission in the atmosphere. Pollution soil with OP caused for increasing of share of potentially pathogenic fungi in the fungal community.

Numerical modeling of the Initial Formation of Cyclonic Vortices at Tropical Latitudes

To investigate the initial formation of large-scale vortices at tropical latitudes a regional non-hydrostatic mathematical model of the wind system of the lower atmosphere, developed earlier in the Polar Geophysical Institute, is utilized. Three-dimensional distributions of the atmospheric parameters in the height range from 0 to 15 km over a limited region of the Earth’s surface are produced by the utilized model. Simulations are performed for the case when the limited three-dimensional simulation domain is intersected by an intertropical convergence zone in the west-east direction. Simulation results indicated that the origin of two convexities in the north direction in the configuration of the intertropical convergence zone can lead to the formation of three distinct tropical cyclones during the period of about four days.
