The organization of the compartment of mesenchymal stem cells is still obscure. Two types of human stromal precursor cells are known. Both of them are analyzed in in v.tro system: mesenchymal multipotent stromal cells (MMSC) and fibroblast colony forming units (CFU-F). The aim of this study was to compare the main characteristics of MMSC and CFU-F deriv.d from the bone marrow of 24 healthy donors. Growth and differentiation parameters, as well as relativ. expression lev.ls of different genes were analyzed in MMSC and CFU-F. MMSC were cultiv.ted for 5 passages. CFU-F concentration was determined for each bone marrow sample. The data obtained demonstrated the heterogeneity and hierarchical organization of both studied populations of stromal precursor cells-MMSC and CFU-F. These two types of stromal precursor cells turned to be different in most parameters studied. Altogether MMSC seemed to be more immature cells than CFU-F and took up the higher position in hierarchical tree of mesenchymal stem cells. The rate of differentiation and proliferativ. potential decreased with the donor’s age in both populations MMSC and CFU-F.

Geochemistry of Thermal Waters of Continental Margin of Far East of Russia

Studied waters belong to warm （T=30-50℃）, alkaline （pH=8.9-9.3）, low mineralized （TDS〈235 mg/l） Na-HCO3 or Na-SO4-HCO3 thermal waters with high content of SiO2 （up to 81 mg/1） and F （up to 3.9 mg/l）, occur on modern v.lcano-tectonic rejuv.nated areas of Eastern Sikhote-Alin orogenic belt. Low 3He concentration as well as N2/O2 and N2/Ar ratios exclude influence of deep mantle fluid. New rare earth element data constrain our understanding of water-rock interaction occurring in the water source region. Meteoric origin of waters is prov.d by stable isotope v.lues v.rying from -71%o to -136.1‰ and from -10.8‰ to -18.8%o for δ^2H and δ^18O respectiv.ly. REE patterns reflect high pH, resultfing from water-rock interaction and oxidativ. conditions. Calculations of deep aquifer temperature using Na-K and quartz geothermometers show 116.8-131.1℃ and 82.2-125.8℃ respectiv.ly. Presence of deep faults both with abnormal thermal gradient （-45- 50 K/km） define unique geochemical shape of thermal waters of Sikhote-Alin, area, where no present v.lcanic activ.ty is registered.

Females of the ectoparasitoid Habrobracon hebetor attack and env.nomate numerous host indiv.duals during ov.position. The v.ctoring of the entomopathogenic fungus Beauv.ria bassiana during the adhesion stage by ectoparasitoid females among the wax moth larv.e Galleria mellonella was explored under laboratory conditions. v.ctoring occurred both from infected parasitoids to wax moth larv.e and from infected to healthy wax moth larv.e by parasitoids. The efficacy of v.ctoring in both cases was dose dependent. Parasitoid females were unable to recognize infected larv.e in a labyrinth test. In addition, the presence of H. hebetor females significantly （1.5-13 fold） increased the mycoses lev.l in clusters of G. mellonella, with 40% of the larv.e infected with ftmgal conidia. Env.nomation by H. hebetor increased conidia germination on the cuticles of the wax moth larv.e by 4.4 fold. An enhanced germination rate （2 fold） was registered in the n- hexane epicuticular extract of env.nomated larv.e compared to that of healthy larv.e. Both env.nomation and mycoses enhanced the phenoloxidase （PO） activ.ty in the integument of G. mellonella and, in contrast, decreased the encapsulation rate in hemolymphs. We hypothesize that changes in the integument property and inhibition of cellular immunity prov.de the highest infection efficacy of entomopathogenic fungi with H. hebetor.

Results of simulations of streamers propagating along the atmospheric-pressure helium plasma jets ejected from a thin tube into ambient air are presented. The computational model includes the equations gov.rning the helium-air mixture composition along and across the jet, as well as the equations, describing streamer propagation in the mixture, for the electric field and the charged species densities. Streamer v.locity and spatial-temporal profiles of plasma parameters are obtained for v.rious v.lues of the helium flow rate. It is shown, in agreement with av.ilable experimental data, that the electric field v.lues in the heads of streamers increase nearly linearly with the distance from the tube exit and that the slope of this dependence decreases with the growth of the helium flow rate.

During germination of barley(Hordeum v.lgare L.) seeds, important morphological and physiological changes take place, including dev.lopment of organs and tissues and activ.tion of metabolic pathways. Germination and dormancy of seeds are regulated by abscisic acid, gibberellins, reactiv. oxygen species(ROS), reactiv. nitrogen species(RNS)and sev.ral other factors. Activ.ties of ascorbate-glutathione cycle enzymes, responsible for scav.nging ROS, strongly increase. Catalase and superoxide dismutase activ.ties, also scav.nging ROS, decrease at the onset of seed germination and then increase. With the increase in aerobic metabolism after radicle protrusion, the activ.ties of the fermentation enzymes lactate and alcohol dehydrogenase decline rapidly. The RNS-scav.nging activ.ty of S-nitrosoglutathione reductase decreases in the course of seed germination, in concert with elev.tion of nitric oxide production and protein nitrosylation. This activ.ty supports the role of RNS in regulating seed germination. Transcription of v.rious genes at different phases of seed germination exhibits phase-specific changes. During imbibition, genes inv.lv.d in cell wall metabolism are highly expressed; in the middle phase of seed germination before radicle protrusion, genes inv.lv.d in amino acid synthesis, protein synthesis, and transport and nucleic acid synthesis are upregulated significantly, and after radicle protrusion, genes inv.lv.d in photosynthetic metabolism are induced. In summary, signal transduction and metabolic regulation of seed germination inv.lv. div.rse reactions and complex regulation at different lev.ls of metabolic organization.

The effect of high fat food on erythrocyte phospholipids, fatty acids composition and glutathione redox-system of rats with alimentary dyslipidemia

To ev.luate the effects of high fat food consisted of tallow (19% of total diets) and cholesterol (2%) on modification of erythrocyte phospholipids, fatty acids composition and glutathione redox- system of male Wistar rats with alimentary dysli- pidemia. The results demonstrated that after 30 and 180 days of high-fat feed erythrocyte phos- phatidylinositol and phosphatidylcholine lev.ls were reduced, phosphatidylserine were in-creased. Only on the 90 days of the experiment phosphatidylinositol lev.l increased. In all grow- ups the erythrocyte 18:0 saturated fatty acids and 20:4n6, 22:4n6 polyunsaturated fatty acids (PUFA) were increased. Deficit of n3 PUFA- 20:5n3 and 22:6n3 after 90 and 180 days high fat feed promoted compensatory synthesis from 18:1n9 on 20:3n9. Erythrocyte maleic dialde-hyde increased, glutathione lev.l decreased in all groups of rats after fed with high-fat feed. Glutathione reductase and glutathione peroxi-dase activ.ty decreased in erythrocytes after 30 and 180 days of high-fat feed. In conclusion: high-fat diet during 30-90 days started adaptiv. answer in lipids of membrane and glutathione redox-system. Important mechanism of adapta-tion of a cellular membrane to high-fat diet is increase major, structuring a membrane phos-phatidylethanolamine and minor, most meta-bolic significant fractions phospholipids (phos- phatidylinositol), keeps homeostasis of 18:2n6 and 22:6n3, 20:3n9 compensatory synthesis, decrease in activ.ty of processes lipid peroxi-dation, activ.tion of enzymes of redox-system glutathione. But prolonging the high-fat feeding (180 days and more) formed failure compensa-tory processes (dysadaptation). It is a risk factor of dev.lopmening atherosclerosis, diabetes, steatogepatitis and other diseases.

BACKGROUND： In recent years, an adaptiv. endoplasmic reticulum （ER） stress response has been activ.ly inv.stigated. The ER membrane, isolated from the intact and regenerating liv.r, may be an appropriate model for inv.stigating the association between structural and functional characteristics of ER in v.v. and their corresponding behav.oral characteristics in v.tro. The rate of lipid synthesis and that of intracellular lipid exchange between the ER and cytosol were inv.stigated in the intact and regenerating liv.r （13 h after partial hepatectomy）. Particularly, membrane characteristics, surface potential, and glucose 6- phosphatase （G6Pase） activ.ty were inv.stigated, along with the degradation rate of G6Pase in v.tro, which was estimated by the loss of G6Pase activ.ty, formation of lipid peroxides, and size of excreted membrane v.sicles. METHODS： The rate of lipid synthesis was determined by measuring the intensity of radioactiv. precursor （C^14-sodium acetate） in different fractions of lipids （phospholipids, non-esterified fatty acids, and triacylglycerides） after 30 min exposure. The rate of lipid metabolism was assessed by measuring the quantity oflipids with radioactiv. labels emerging in the cytosol of hepatocytes （CPM）. v.scosity and surface potential were determined by fluorescent probes. RESULTS： It was observ.d that after 13 h of partial hepatectomy, the rate of lipid synthesis in the ER of hepatocytes in the regenerating liv.r was 3 times lower than that in ER ofhepatocytes in the intact liv.r, wherein the rate of incorporation of newly synthesized lipids in cytosol was sev.ral times higher in the regenerating liv.r. Increase in the rate of exchange of neutral lipids in cells of the regenerating liv.r was accompanied by lipid reconstruction in the ER, changing the structural and functional characteristics of the membrane.
