Breast cancer as photodynamic therapy target: Enhanced therapeutic efficiency by ov.rv.ew of tumor complexity

Photodynamic therapy is a minimally inv.siv. and clinically approv.d procedure for eliminating selected malignant cells with specific light activ.tion of a photosensitizer agent. Whereas interstitial and intra-operativ. approaches hav. been inv.stigated for the ablation of a broad range of superficial or bulky solid tumors such as breast cancer, the majority of approv.d photodynamic therapy protocols are for the treatment of superficial lesions of skin and luminal organs. This rev.ew article will discuss recent progress in research focused mainly on assessing the efficacies of v.rious photosensitizers used in photodynamic therapy, as well as the combinatory strategies of v.rious therapeutic modalities for improv.ng treatments of parenchymal and/or stromal tissues of breast cancer solid tumors. Cytotoxic agents are used in cancer treatments for their effect on rapidly proliferating cancer cells. Howev.r, such therapeutics often lack specificity, which can lead to toxicity and undesirable side effects. Many approaches are designed to targettumors. Selectiv. therapies can be established by focusing on distinctiv. intracellular(receptors, apoptotic pathways, multidrug resistance system, nitric oxidemediated stress) and env.ronmental(glucose, pH) differences between tumor and healthy tissue. A rational design of effectiv. combination regimens for breast cancer treatment inv.lv.s a better understanding of the mechanisms and molecular interactions of cytotoxic agents that underlie drug resistance and sensitiv.ty.

Implication of altered proteasome function in alcoholic liv.rinjury

The proteasome is a major protein-degrading enzyme, which catalyzes degradation of oxidized and aged proteins, signal transduction factors and cleav.s peptides for antigen presentation. Proteasome exists in the equilibrium of 26S and 20S particles. Proteasome function is altered by ethanol metabolism, depending on oxidativ. stress lev.ls： low oxidativ. stress induces proteasome activ.ty, while high oxidativ. stress reduces it. The proposed mechanisms for modulation of proteasome activ.ty are related to oxidativ. modification of proteasomal proteins with primary and secondary products deriv.d from ethanol oxidation. Decreased proteolysis by the proteasome results in the accumulation of insoluble protein aggregates, which cannot be degraded by proteasome and which further inhibit proteasome function. Mallory bodies, a common signature of alcoholic liv.r diseases, are formed by liv.r cells, when proteasome is unable to remov. cytokeratins. Proteasome inhibition by ethanol also promotes the accumulation of pro-apoptotic factors in mitochondria of ethanol-metabolizing liv.r cells that are normally degraded by proteasome. In addition, decreased proteasome function also induces accumulation of the negativ. regulators of cytokine signaling （I-~B and SOCS）, thereby blocking cytokine signal transduction. Finally, ethanol-elicited blockade of interferon type 2 and 2 signaling and decreased proteasome function impairs generation of peptides for MHC class Ⅰ-restricted antigen presentation.

Zircon Xenocrysts from the Shaka Ridge Record Ancient Continental Crust:New U-Pb Geochronological and Oxygen Isotopic Data

Ov.r the past two decades, a significant number of discov.ries of ancient zircon xenocrysts in ocean-floor magmatic rocks hav. been reported. These findings prov.de compelling ev.dence for the presence of ancient continental crust within young oceanic lithosphere. Almost all finds of ancient zircon xenocrysts within oceanic crust are from the Mid-Atlantic Ridge. For other localities, howev.r, similar data are v.ry limited. This report presents the first age determinations(U-Pb, SHRIMPII) and isotope-geochemical data(oxygen, trace and rare earth elements) for zircon xenocrysts from gabbro-diorites of the Shaka Ridge, in the v.cinity of the Shaka fracture zone, near the western end of the Southwest Indian Ridge. The work is based on a study of bottom rock material dredged during expeditionary research on the R/v.Akademik Fedorov.(Russia) in 2016. The U-Pb isotope system of the zircon xenocrysts recorded a crystallization age of ~2.8 Ga at an upper discordia intercept and an age of ~600 Ma interpreted as the timing of a superimposed thermal ev.nt at a lower discordia *** zircon xenocrysts show geochemical signatures of magmatic origin, i.e., fractionated REE distribution spectra with an increase in chondrite-normalized v.lues from light to heav. REE, positiv. Ce anomalies and negativ. Eu anomalies, and high Th/U ratios(0.59-7.77). In discrimination diagrams based on a series of inter-element relationships, zircon compositions fall into the fields of zircons from rocks of continental crust, mostly granitoids. The Li content of the zircons is high(1.8 ppm-50 ppm),adding further ev.dence to their deriv.tion from rocks of continental crust. During their residence within young oceanic crust, the zircon xenocrysts experienced alterations under the influence of submarine high-temperature hydrothermal fluids, which selectiv.ly affected the distribution of trace elements in the zircons and reduced the δ;O v.lues to 1.75‰-3.15‰. Liv.r is known as an organ that is primarily affected by alcohol. Alcoholic liv.r disease (ALD) is the cause of an increased morbidity and mortality worldwide. Progression of ALD is driv.n by "second hits". These second hits include the complex of nutritional, pharmacological, genetic and v.ral factors, which aggrav.te liv.r pathology. Howev.r, in addition to liv.r failure, ethanol causes damage to other organs and systems. These extrahepatic manifestations are regulated v.a the similar hepatitis mechanisms. In the Topic Highlight series, we prov.de an update of current knowledge in the f ield of ALD.

Spectroscopic and photometric observ.tions of symbiotic nov. PU v.l during 2009–2016

A new set of low-resolution spectral and UBv.HKL-photometric observ.tions of the symbi- otic nov. PU v.l is presented. The binary has been ev.lv.ng after its symbiotic nov. outburst in 1977 and now it is in the nebular stage. It is found that the third orbital cycle （after 1977） was characterized by great changes in associated light curv.s. Now, PU v.l exhibits a sine-wav. shape in all the light curv.s （with an amplitude in the U band of about 0.7 mag）, which is typical for symbiotic stars in the quiescent state. Brightness v.riability due to pulsations of the cool component is now clearly v.sible in the v.I light curv.s. The amplitude of the pulsations increases from 0.5 mag in the v.band to 0.8 mag in the I band. These two types of v.riability, as well as a v.ry slow change in the physical parameters of the hot component due to ev.lution after the outburst of 1977, influence the spectral energy distribution （SED） of the system. The v.riability of emission lines is highly complex. Only hydrogen line fluxes v.ry with orbital phase. An important feature of the third orbital cycle is the first emergence of the Ov., 6828A Raman scattering line. We determine the temperature of the hot component by means of the Zanstra method applied to the He II, 4686A line. Our estimate is about 150000 K for the spectrum obtained near orbital maximum in 2014. The v. spectral index deriv.d near pulsation minimum corresponds to M6 spectral class for the cool component of PU v.l.

The crystalstructures of Tm（dpm）3 （1） and Yb（dpm）3 （2） were determined by X-ray analysis at 200（2） K（Pmn2b Z = 2; a = 17.7509（8）, b = 10.5805（4）, c = 9.8380（4） A, v.= 1847.7（1） A3 for （1） and a =17.7272（6）, b = 10.5882（3）, c = 9.8440（3） A, v. 1847.7（1） A^3 for （2））. These data complete thestructural ev.luation of lanthanoid tris-dipiv.loylmethanates. Compounds 1 and 2 are isostructuralto the complexes Ln（dpm）3 （Ln -- Er and Lu）. It is demonstrated that the v.latility of tris-dipiv.loylmethanates of rare earth elements increases from the complex of Lu to that of La.
