Sperm fine-needle aspiration (FNA)mapping after failed microdissection testicular sperm extraction (TESE):location and patterns of found sperm

We sought to evaluate the ability of fine-needle aspiration (FNA)mapping to find sperm and to guide sperm retrieval after failed microdissection testicular sperm extraction (micro-TESE)in nonobstructive azoospermic *** this study of consecutive male infertility cases,interventions included testicular FNA mapping and subsequent sperm *** included the frequency and location of found sperm on FNA maps after failed micro-TESE and the salvage sperm retrieval *** 548 patients undergoing FNA mapping from 2010 to 2016,82 men with previous micro-TESE procedures were *** mean time between micro-TESE and FNA mapping was 2.2 years.A total of 2825 (1424 on right and 1401 on left)sites were *** least one site revealed mature sperm in 24 (29.3%)of 82 men with prior failed micro-TESE *** was an equal likelihood of detecting sperm in either testis (6.1%right;5.7%left;P=0.58).Digital "heat maps"revealed differences in sperm findings within the testis with mature sperm more likely found in the testis periphery rather than *** (62.5%)patients subsequently underwent sperm retrieval procedures guided by FNA *** sperm were retrieved in all cases,and in 10 (66.7%)of 15 cases,extra sperm were frozen for future *** a significant proportion of failed micro-TESE procedures representing the largest study to date,sperm were detected by FNA mapping and could be reliably retrieved through FNA map-guided surgical sperm *** present,sperm were more likely to be found in the testis periphery rather than centrally with FNA mapping.

Microstructure Characterization of a Biogenic Shale Gas Formation——Insights from the Antrim Shale,Michigan Basin

Biogenic gas shales,predominantly microbial in origin,form an important class of organicrich shale reservoirs with a significant economic *** large gaps remain in the understanding of their gas generation,storage,and transport mechanisms,as previous studies have been largely focused on mature thermogenic shale *** this study,the pore structure of 18 Antrim Shale samples was characterized using gas adsorption(CO2 and N2).The results show that most of the Antrim Shale samples are rich in organic matter content(0.58 wt.%to 14.15 wt.%),with highest values found in the Lachine and Norwood *** from the Paxton Member,characterized by lower organic content,have smaller micropore surface area and micropore volume but larger meso-macro pore surface area and *** deconvolution results of the pore size distribution from the N2 adsorption indicate that all of the tested Antrim Shale samples have similar pore *** matter in the Antrim Shale hosts micro pores instead of meso-macro pores,while clay minerals host both micro and meso-macro *** pores play a primary role in the total *** biogenic Antrim Shale,therefore,has different pore structures from other well-studied thermogenic gas shales worldwide.

Insulin secretion is a complex and highly regulated *** much progress has been made in understanding the cellular mechanisms of insulin secretion and regulation,it remains unclear how conclusions from these studies apply to living *** few studies have been done to address these issues is largely due to the lack of suitable tools in detecting secretory events at high spatial and temporal resolution in *** combined with genetically encoded biosensor,optical imaging is a powerful tool for visualization of molecular events in *** this study,we generated a DNA construct encoding a secretory granule resident protein that is linked with two spectrally separate fluorescent proteins,a highly pH-sensitive green pHluorin on the intra-granular side and a red mCherry in the *** exocytosis of secretory granules,the dim pHluorin inside the acidic secretory granules became highly fluorescent outside the cells at neutral pH,while mCherry fluorescence remained constant in the process,thus allowing ratiometric quantification of insulin secretory ***,mCherry fluorescence enabled tracking the movement of secretory granules in living *** validated this approach in insulin-secreting cells,and generated a transgenic mouse line expressing the optical sensor specifically in pancreaticβ-*** transgenic mice will be a useful tool for future investigations of molecular mechanism of insulin secretion in vitro and in vivo.

High-temperature transport properties of BaSn_(1−x)Sc_(x)O_(3−δ) ceramic materials as promising electrolytes for protonic ceramic fuel cells

Protonic ceramic fuel cells(PCFCs)offer a convenient means for electrochemical conversion of chemical energy into electricity at intermediate temperatures with very high *** BaCeO_(3)-and BaZrO_(3)-based complex oxides have been positioned as the most promising PCFC electrolytes,the design of new protonic conductors with improved properties is of paramount *** the present work,we studied transport properties of scandium-doped barium stannate(Sc-doped BaSnO_(3)).Our analysis included the fabrication of porous and dense BaSn_(1−x)Sc_(x)O_(3−δ)ceramic materials(0≤x≤0.37),as well as a comprehensive analysis of their total,ionic,and electronic conductivities across all the experimental conditions realized under the PCFC operation:both air and hydrogen atmospheres with various water vapor partial pressures(p(H2O)),and a temperature range of 500–900℃.This work reports on electrolyte domain boundaries of the undoped and doped BaSnO_(3)for the first time,revealing that pure BaSnO_(3)exhibits mixed ionic–electronic conduction behavior under both oxidizing and reducing conditions,while the Sc-doping results in the gradual improvement of ionic(including protonic)conductivity,extending the electrolyte domain boundaries towards reduced *** latter property makes the heavilydoped BaSnO_(3)representatives attractive for PCFC applications.

Application of solvent pH under pressurized conditions using accelerated solvent extraction and green solvents to extract phytonutrients from wild berries

Berry fruits are known as“superfoods”due to health benefits associated with the high content of bioactive compounds or *** extraction solvents are fundamentally volatile compounds acquired from non-renewable resources,i.e petroleum-based solvents are presumed to be harmful to both human health and the *** to the new emphasis on environmental and safety protections and the development of green chemistry,finding alternative solvents to replace the organic ones has recently become a major ***,there is a great interest in the scientific community to develop a suitable green approach to allow the extraction and recovery of the bioactive phytonutrients from berry sources for applications in improving human health and *** this study,the phytonutrient composition of three popular wild berries:blueberry(Vaccinium angustifolium),partridgeberry(Vaccinium vitis-idaea L.)and Rosehipberry(Rosa canina),were evaluated following extraction using biphasic green solvents at pH 2.5,7.0,and 11.5 under pressurized conditions using Accelerated Solvent Extraction(ASE)*** total phenolic content and composition,antioxidant activity,vitamin and mineral content of the berry extracts were evaluated using Folin-Ciocalteu assay,Ferric Reducing Antioxidant Power Assay(FRAP),Ultra-High-pressure Liquid Chromatography-Mass Spectrometry(UHPLC-MS)and inductively coupled plasma-mass spectrometry(ICP-MS),*** highest total antioxidant activity(p≤0.05)and phenolic content were observed in rosehip berry followed by blueberry and partridgeberry in the hydroethanolic extracts at pH ***,the highest extraction efficiency of total fat-soluble vitamins(FSV)and total B vitamins were also observed in the hydroethanolic extract at pH *** in rosehipberry and partridgeberry the highest vitamin C content was observed in hydroethanolic extract at pH 11.5,whereas in blueberry it was observed in the pH 2.5 hydroethanolic extract.I

Influence of poloxamer 407 on fractional and subfractional composition of serum lipoproteins of mice

Using a novel small-angle X-ray scattering (SAXS) method for determination of fractional and subfractional composition of lipoproteins (LPs), a significant elevation of total cholesterol-lipop- roteins (C-LP) and, especially, total triglyceride- lipoproteins (TG-LP), was shown in this work. Among the LP fractions, poloxamer 407 was shown to significantly increase proatherogenic total C-LDL, TG-LDL and, especially, their precursors C-VLDL and TG-VLDL, while only exhibiting a moderate increase in the antiatherogenic C-HDL and TG-HDL fractions. With regard to the VLDL subfractions, significant elevations were observed in both subfractions studied;namely, C-VLDL1-2 and C-VLDL3-5. Similar chang- es were noted in the TG-VLDL1-2 and TG- VLDL3-5 subfractions. The C-IDL and TG-IDL subfractions were increased significantly (?20- to 30- fold), while the C-LDL1-3 subfraction was moderately (?3- to 5-fold) increased at 48 hrs and at day 4. In the moderately elevated (?2- to 4-fold) anti-atherogenic HDL fraction, the C-HDL2 subfraction was increased more significantly (?4- fold) compared to the C-HDL3 subfraction;how- ever, both C-HDL subfractions returned to base- line by day 4. The elevation in the TG-HDL2 subfraction was observed only at 24 hrs. Mouse models of hyperlipidemia and atherosclerosis are useful to evaluate the role of “individual” LPs, as well as their fractions and subfractions, in hyperlipidemia and the genesis of atherosclerosis.

Model experiments with low melting point liquid metals are an important tool to investigate the flow structure and related transport processes in melt flows relevant for metallurgical applications. We present recent results from the three LIMMCAST facilities working either with room-temperature alloy Ga In Sn or with the alloy Sn Bi at temperatures of 20050°C. The main value of cold metal laboratory experiments consists in the capabilities to obtain quantitative flow measurements with a reasonable spatial and temporal resolution, which is essential for code validation. Experimental results are presented covering the following phenomena: contactless electromagnetic tomography of the flow in the mold, flow monitoring by ultrasonic sensors, mold flow under the influence of an electromagnetic brake, injection of argon bubbles through the stopper rod, X-ray visualization of gas bubble two-phase flow in the nozzle and in the mold.
