The eIP-Agri multiactor approach was exemplified during a 3-day workshop with 63 project participants from the eU H2020 funded project“Redesigning european cropping systems based on species MIXtures”.The objective was to share firsthand experience of participatory research among researchers who were mostly not familiar with this *** participants were divided into smaller multidisciplinary groups and given the opportunity to interact with representatives from eight actor positions in the value chain of the agrifood cooperative Terrena located in Western *** four stages of the workshop were:(1)key actor interviews,(2)sharing proposed solutions for overcoming barriers,and(3)developing possible interdisciplinary *** of frustration were recorded serving both as a motivation for group members to become more aware of the scientific concerns and practices of their colleagues,as well as a recognition that some researchers have better skills integrating qualitative approaches than ***,the workshop format was an effective way to gain a common understanding of the pertinent issues that need to be addressed to meet overall multiactorapproach *** with the actor networks was identified and emphasized as a means to overcome existing barriers between academia and practice in order to coproduce a shared vision of the benefits of species mixture benefits.

Ultrasensitive detection of DNA and protein markers in cancer cells

cancer cells differ from normal cells in various parameters, and these differences are caused by genomic mutations and consequential altered gene expression. The genetic and functional heterogeneity of tumor cells is a major challenge in cancer research, detection, and effective treatment. As such, the use of diagnostic methods is important to reveal this heterogeneity at the single-cell level. Droplet microfluidic devices are effective tools that provide exceptional sensitivity for analyzing single cells and molecules. In this review, we highlight two novel methods that employ droplet microfluidics for ultrasensitive detection of nucleic acids and protein markers in cancer cells. We also discuss the future practical applications of these methods.

Histone Acetyltransferase AtGcN5/HAG1 Is a Versatile Regulator of Developmental and Inducible Gene expression in Arabidopsis

Histone acetylation/deacetylation is a dynamic process and plays an important role in gene regulation. Histone acetylation homeostasis is regulated by antagonist actions of histone acetyltransferases （HAT） and deacetylases （HDAc）. Plant genome encodes multiple HATs and HDAcs. The Arabidopsis HAT gene AtGcNS/HAGlplays an essential role in many plant development processes, such as meristem function, cell differentiation, leaf and floral organogenesis, and responses to environmental conditions such as light and cold, indicating an important role of this HAT in the regulation of both long-term developmental switches and short-term inducible gene expression. AtGcN5 targets to a large number of promoters and is required for acetylation of several histone H3 lysine residues. Recruitment of AtGcN5 to target promoters is likely to be mediated by direct or indirect interaction with DNA-binding transcription factors and/or by interaction with acetylated histone lysine residues on the targets. Interplay between AtGcN5 and other HATand HDAc is demonstrated to control specific regulatory pathways. Analysis of the role of AtGcN5 in light-inducible gene expression suggests a function of AtGcN5 in preparing chromatin commitment for priming inducible gene activation in plants.

The P-T conditions of auriferous and barren quartz veins from Kopylovsky,Kavkaz and Krasnoye gold deposits in Proterozoic black shales of Bodaybo ore region are presented the first time in this *** inclusions trapped in auriferous quartz are aqueous Na±K-Mg chloride with salinity of 6 wt.%-8.8 wt.%*** temperatures vary from 260 to 350℃,and calculated trapping pressures are 1.2-1.6 *** fluids trapped in barren quartz have more complicated compositions with Na,K,Mg and fe chlorides,salinity up to 13 wt.%Nacleqv,and homogenization temperatures ranging between 140 and 280℃.The volatiles in fluids are dominated by H2O,followed by cO2 with minor amounts of cH4 and *** suppose that auriferous and barren quartz veins have been formed due to the basic metamorphogenic fluid as evidenced by the close slat and gas fluid composition.

The flow velocity of gases in gobs directly influences the kinetics and intensity of gaseous components release during heating and cooling of *** assessment of fire hazard is performed on the basis of concentrations of particular gases in a mine *** concentrations differ in coal heating and cooling phase which was proven in the *** paper presented the results of the experimental study on temperature distribution in a simulated coal bed in heating(50–250°c)and cooling(250–35°c)phases as well as its correlation to variations in concentration of gases released in these phases and flow rates of gases flowing through the coal *** research was performed on twenty-two samples of bituminous coals acquired from various coal beds of Polish coal *** differences were observed between heating and cooling phases in terms of the concentrations of gases taken into account in calculations of self-combustion *** the heating phase temperature increase resulted in the decrease of concentrations ratios of ethane,ethylene,propane,propylene and acetylene,while in the cooling phase these ratios increased *** effect of air(in heating phase)and nitrogen(in cooling phase)flow rate on the self-ignition index cO/cO2 was also determined.

Translational approaches: from fatty liver to non-alcoholic steatohepatitis

Over the past few decades, non-alcoholic fatty liver disease(NAfLD) has become one, if not the most common,cause of chronic liver disease affecting both adults and children. The increasing number of cases at an early age is the most worrying aspect of this pathology, since it provides more time for its evolution. The spectrum of this disease ranges from liver steatosis to steatohepatitis, fibrosis and in some cases, hepatocellular carcinoma. NAfLD may not always be considered a benign disease and hepatologists must be cautious in the presence of fatty liver. This should prompt the use of the available experimental models to understand better the pathogenesis and to develop a rational treatment of a disease that is dangerously increasing. In spite of the growing efforts, the pathogenesis of NAfLD is still poorly understood. In the present article we review the most relevant hypotheses and evidence that account for the progression of NAfLD to non-alcoholic steatohepatitis(NASH) and fibrosis. The available in vitro and in vivo experimental models of NASH are discussed and revised in terms of their validity in translational studies. These studies must be aimed at the discovery of the still unknown triggers or mediators that induce the progression of hepatic inflammation, apoptosis and fibrosis.

Role of chromosomal instability and clonal heterogeneity in the therapy response of breast cancer cell lines

Objective:chromosomal instability(cIN)is a hallmark of cancer characterized by cell-to-cell variability in the number or structure of chromosomes,frequently observed in cancer cell populations and is associated with poor prognosis,metastasis,and therapeutic *** cancer(Bc)is characterized by unstable karyotypes and recent reports have indicated that cIN may influence the response of Bc to chemotherapy ***,paradoxical associations between extreme cIN and improved outcome have been ***:This study aimed to 1)evaluate cIN levels and clonal heterogeneity(cH)in Mcf7,ZR-751,MDA-MB468,BT474,and KPL4 Bc cells treated with low doses of tamoxifen(TAM),docetaxel(DOc),doxorubicin(DOX),Herceptin(HT),and combined treatments(TAM/DOc,TAM/DOX,TAM/HT,HT/DOc,and HT/DOX)by using fluorescence in situ hybridization(fISH),and 2)examine the association with response to treatments by comparing fISH results with cell ***:Intermediate cIN was linked to drug sensitivity according to three characteristics:estrogen receptorα(eRα)and HeR2 status,pre-existing cIN level in cancer cells,and the cIN induced by the ***α+/HeR2-cells with intermediate cIN were sensitive to treatment with taxanes(DOc)and anthracyclines(DOX),while eRα-/HeR2-,eRα+/HeR2+,and eRα-/HeR2+cells with intermediate cIN were resistant to these ***:A greater understanding of cIN and cH in Bc could assist in the optimization of existing therapeutic regimens and/or in supporting new strategies to improve cancer outcomes.
