Transmission of cryptococcosis by liver transplantation:A case report and review of literature

bACKGROUND Cryptococcosis is a fungal infection caused by the yeast-like encapsulated basidiomycetous fungus of the Cryptococcus neoformans(***)species *** fungi are ubiquitous in soil and bird droppings,and infection by them is an important global health concern,particularly in immunosuppressed patients,such as organ transplant recipients and those infected by the human immunodeficiency *** fungus usually enters the body through the respiratory tract,but extremely rare cases of infection acquired by transplantation of solid organs have been *** SUMMARY We report a case of disseminated cryptococcosis in a liver transplant recipient,diagnosed 2 wk after the *** patient initially presented with fever,hyponatremia and elevated transaminase levels,manifesting intense headache after a few *** cultures were positive for *** biopsy showed numerous fungal elements surrounded by gelatinous matrix and sparse granulomatous *** resonance imaging of the brain showed multiple small lesions with low signal in T2,peripheric enhancement and edematous halo,diffuse through the parenchyma but more concentrated in the subcortical *** with amphotericin b for 3 wk,followed by maintenance therapy with fluconazole,led to complete resolution of the *** recipients of both kidneys from the same donor also developed disseminated cryptococcosis,confirming the transplant as the source of *** organ donor lived in a rural area,surrounded by tropical rainforest,and had negative blood cultures prior to organ *** This case highlights the risk of transmission of fungal diseases,specifically of ***,through liver graft during liver transplantation.

The 19 kDa Protein from <i>Mycobacterium avium subspecies paratuberculosis</i>Is a Glycolipoprotein

This study characterizes the 19 kDa protein expressed by Mycobacterium avium subspecies paratuberculosis (MAP) as a glycolipoprotein, providing the foundation for future experiments regarding its antigenicity and role in disease pathogenicity. We have previously shown that a 4.8 kb insert from MAP will produce a 16 kDa recombinant protein when expressed in Escherichia coli and 19 kDa recombinant protein when expressed in M. smegmatis (smeg19K). The difference of 3 kDa in size of these expressed proteins may be related to post translational modifications that occur in Mycobacterium species. We hypothesized that smeg19K is a glycolipoprotein since bLAST analysis revealed approximately 76% amino acid identity between the MAP 19 kDa protein and a known lipoglycoprotein, the 19 kDa protein of M. tuberculosis. This prediction was confirmed by the following positive staining of smeg19K with Sudan black 4b, a postelectrophoresis dye used to stain for lipids. Smeg19K has also stained positively for glycosylation with the lectin concavalin A, a highly specific stain for mannose residues. As expected, treatment with tunicamycin (an antibiotic known to inhibit N-glycosylation) and treatment with deglycosylation assay (non-specific for mannose), showed no reduction in size of 19 kDa glycolipoprotein.

Variation of the Content of Ethyl Esters in Extra Virgin Olive Oils during Their Shelf Life

Fatty acids＇ ethyl and methyl esters are not natural components of edible vegetable oils and therefore should not be present in virgin olive oils. Among the quality requirements for extra virgin olive oils, the International Olive Council （IOC） Norm, 2015 review, set limits for the ethyl ester content at ≤ 435 ppm for the 2014/2015 and 2015/2016 harvests and at ≤ 30 ppm for the 2016/2017 harvest. The purpose of this paper is to：（a） determine the alkyl ester content in two extra virgin olive oils （of the Arbequina and Coratina varieties） as parameters of quality at the moment of their elaboration and to assess the length of their shelf life, over a 12 month period at room temperature; （b） determine extra virgin olive oils＇ free alcohols content as these may esterify due to the free fatty acids present during storage. After 6 months of storage, the Arbequina oil＇s ethyl ester content was 32 ppm, and the Coratina＇s was 46 ppm. As a result, the shelf life for both was only about half a year （labeling usually indicates it must be consumed ＂before 1 year＂）. However, parameters related to oxidation （peroxide index, K 232, K 270 and AK） remained within the limits set by the International Olive Council （IOC） throughout the year. In conclusion： ethyl ester content is a very fine parameter for assessing the quality of extra virgin in an olive oil and determining the length of its shelf life.

Efficacy of Afoxolaner Alone or in Combination with Milbemycin Oxime against Rhipicephalus sanguineus Lato Sensu in Naturally Infested Dogs in Colombia

The brown dog tick (Rhipicephalus sanguineus lato sensu) is the tick that most affects dogs worldwide and is therefore the main blood pathogen vector in dogs. The efficacy of afoxolaner 2.7 to 7.1 mg/kg NexGard® (group A) and afoxolaner plus milbemycin oxime 2.5 to 5.4 mg/kg and 0.5 to 1.1 mg/kg respectively NexGard Spectra® (Group b) against R. sanguineus, was evaluated in naturally infected sheltered dogs under high challenging conditions in four different areas of Colombia (Antioquia, Córdoba, Santander, and Meta). Tick counts (alive, dead, attached, and unattached) were performed on treated dogs, the average was calculated for the different areas to evaluate the efficacy of each treatment at six different times post-treatment (24 h, 48 h, 7 d, 14 d, 21 d, 30 d). None of the dogs showed adverse events related to the treatments. The average tick number pre-treatment was 68 in group A and 78.3 for group b indicating a strong natural infection of the dogs and their environment. Efficacy after 24 h against R. sanguineus was always above 90% with ≥97.4% for NexGard® and ≥93.7% for NexGard Spectra®. Consistent results were observed along all the observation periods with final efficacies (day 30) of ≥99.8% and 98.3% for NexGard® and NexGard Spectra®, respectively. In conclusion, both NexGard® and Nexgard Spectra® provided a curative effect and sustained efficacy against Rhipicephalus sanguineus for at least 30 days in highly contaminated shelter environments.

We report the isolation of a cold-adapted bacterium belonging to the genus Janthinobacterium （named AU 11）, from a water sample collected in Lake Uruguay （King George Island, South Shetlands）. AUI 1 （growth between 4℃ and 30℃） produces a single cold-active extracellular protease （ExPAU11）, differentially expressed at low temperature. ExPAU11 was identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry （MALDI-ToF MS） as an alkaline metallo-protease （70% coverage with an extracellular protease of Janthinobacterium sp. PI12）, and by protease-inhibitor screening identified as a serine-protease. To the best of our knowledge this is the first experimental evidence of a cold-active extracellular protease produced by Janthinobacterium. Furthermore, we identified a serine-protease gene （named JSP8A） showing 60% identity （98% query coverage） to subtilisin peptidases belonging to the $8 family （S8A subfamily） of many cyanobacteria. A phylogenetic analysis of the JSP8A protease, along with related bacterial protein sequences, confirms that JSP8A clusters with S8A subtilisin sequences from different cyanobacteria, and is clearly separated from SSA bacterial sequences of other phyla （including its own）. An analysis of the genomic organization around JSP8A suggests that this protease gene was acquired in an event that duplicated a racemase gene involved in transforming L- to D-amino acids. Our results suggest that AU11 probably acquired this subtilisin-like protease gene by horizontal gene transfer （HGT） from a cyanobacterittrn. We discuss the relevance of a bacterial protease-HGT in the Antarctic environment in light of this hypothesis.

Like other optical astrometric techniques,the Photographic Zenith Tube(PZT) has played a key role in the past observations of the Earth rotation,and it also has a potential to be applied to several other observations by taking advantage of automatic observations with self compensation of tilt of the *** here propose In-situ Lunar Orientation Measurement(ILOM) to study lunar rotational dynamics by direct observations of the lunar rotation from the lunar surface by using a small telescope like PZT with an accuracy of 1 milli-seconds of arc(1 mas) in the post-SELENE *** second application is to obtain local gravity field on the Earth by combining deflection of the vertical measured by PZT and the position measured by Global Positioning System(GPS) or Global Navigation Satellite System(GNSS).The accuracy required for this purpose is not as strict as *** have already developed a bread board Model(bbM) of the telescope for ILOM and made some experiments in order to know the performance of the driving mechanism under a similar condition to the lunar environment showing high vacuum,large temperature change and dusty *** have also shown that it is possible to correct the effects of uniform temperature change upon the optical system by using a simple model with an accuracy of better than 1 *** model has the potential to attain the accuracy of 1 mas,based on the results of the experiments and the simulations.

Sorghum (Sorghum bicolor (L.) Moench) is one of the world’s leading cereal crops in agricultural production, which has a special importance in the arid regions. However, unlike other cereals, sorghum grain has a lower nutritional value, which is caused, inter alia, by the resistance of its seed storage proteins (kafirins) to protease digestion. One of the effective approaches to improve the nutritional value of sorghum grain is to obtain mutants with partially or completely suppressed synthesis or altered amino acid composition of kafirins. The employment of genome editing may allow to solve this problem by introducing mutations into the nucleotide sequences of the α- and γ-kafirin genes. In this study, genomic target motifs (23 bp sequences) were selected for the introduction of mutations into the α- and γ-KAFIRIN genes of sorghum. The design of the gRNAs was conducted using the online tools CRISPROR and CHOPCHOP. Two most suitable targets were chosen for α-KAFIRIN (k1C5) and two for γ-KAFIRIN (gKAF1) genes. The insertion of respective sequences in the generic vector pSH121 was performed at the bsaI (Eco31I) sites. Validation of the cloning procedure was performed by DNA sequencing. Subcloning of the resulting constructs was performed using the SfiI restriction sites into the compatible binary vector b479p7oUZm-LH. The correct assembly of binary vectors was confirmed by restriction analysis using the MluI and SfiI cleavage sites. The four vectors created (1C - 4C) were transferred by electroporation into the Agrobacterium tumefaciens strain AGL0. Currently, this vector series is used for stable

The Role of Eumelanin in Generating Reactive Oxygen and Reactive Nitrogen in Solution: Possible Relevance to Keloid Formation

Recently, nitric oxide (NO) has been implicated as an epigenetic factor in keloids, a scarring disease occurring primarily in dark skinned people who have relatively high amounts of pigment melanin. In this work, we tested whether a melanin-mediated redox reaction involving adsorbed NO and O2 can couple NO oxidation with O2 reduction to form reactive oxygen species (ROS) or reactive nitrogen species (RNS) in vitro at pH 7.4. We measured the formation of reactive species that oxidize dihydrorhodamine123 (DHR) to fluorescent rhodamine123 in the presence and absence of sepia melanin. In separate experiments, we monitored NO concentration with 4,5-diaminofluorescein (DAF) by measuring the highly fluorescent NO-adduct, DAF-2T. We attempted to detect peroxynitrite with 5 μM 3-methyl-1,2-cyclopentanedione (MCP), a selective scavenger of peroxynitrite (IC50 = 3.6 μM for ONOO- vs. 63.8 μM and >> 100 μM for NO and O2- respectively). However, MCP itself oxidized DHR. We found that in the absence of NO, melanin itself oxidizes DHR, with no loss of DAF-fluorescence (i.e. no net consumption of NO). In the presence of NO, there was a ~ 57% loss of DAF fluorescence, indicating that NOx is formed at the expense of NO. The data provided good fit (r2 = 0.94) to a Langmuir adsorption isotherm, with pseudo first order rate k' = 8.2 × 107) s-1 and adsorption coefficient Kad = 4.04 M-1. both of these parameters are consistent with a facile chemisorption reaction between NO and O2 on the melanin surface. Possible reactions are a) NO and O2- → ONOO- and/or b) 2NO + O2 → 2NO2. The latter reaction is disfavored in solution but is significantly accelerated on the melanin surface via an entropy effect.

以4-氨基-3-(4-甲基-1,2,3-噻二唑)-1,2,4-三唑-5-硫酮为原料,与醛在冰乙酸中回流制得15个新型4-取代亚氨基-3-(4-甲基-1,2,3-噻二唑)-1,2,4-三唑-5-硫酮化合物,其结构经IR、1H NMR及元素分析表征,其中,化合物5c的结构经单晶测试确证,该晶体属单斜晶系,P2(1)/c空间群,晶胞参数a=1.403 7(3)nm,b=1.570 5(3)nm,c=0.6864(14)nm,β=102.06(3)°,V=1.4798(5)nm3,Z=4,F(000)=656。初步的抑菌活性测试结果表明;所有化合物对黄瓜灰霉病菌botrytis cinerea都有较好的抑制作用,化合物5l的抑制率达87%,5c、5d和5f的抑制率在78%左右;5a对小麦赤霉病菌Gibberella zeae的抑制率为78.7%;5m对西瓜炭疽病菌Colletotrichum lagenarium的抑制率为65.6%。
