In vivo femtosecond laser nanosurgery of the cell wall enabling patch-clamp measurements on filamentous fungi

Studying the membrane physiology of filamentous fungi is key to understanding their interactions with the environment and crucial for developing new therapeutic strategies for disease-causing ***,their plasma membrane has been inaccessible for a micron-sized patch-clamp pipette for pA current recordings due to the rigid chitinous cell ***,we report the first femtosecond IR laser nanosurgery of the cell wall of the filamentous fungi,which enabled patch-clamp measurements on protoplasts released from hyphae.A reproducible and highly precise(diffraction-limited,submicron resolution)method for obtaining viable released protoplasts was *** release from the nanosurgery-generated incisions in the cell wall was achieved from different regions of the *** plasma membrane of the obtained protoplasts formed tight and high-resistance(GΩ)contacts with the recording *** entire nanosurgical procedure followed by the patch-clamp technique could be completed in less than 1 *** to previous studies using heterologously expressed channels,this technique provides the opportunity to identify new ionic currents and to study the properties of the ion channels in the protoplasts of filamentous fungi in their native environment.

Maize cobs are valuable plant residues that can be used for many different purposes,including as a renewable energy *** their value,maize cobs are still underutilized,mainly because people are not aware of their potential and harvesting techniques for their collection are still being ***,this research focused on finding the most suitable energy-and time-efficient method for mechanical collection of maize *** the combine harvester with an additional hopper proved to be the most promising solution,as it allows harvesting and storage of maize grain and maize cobs in one pass,while the harvesting time remains almost the same and the maize cobs are of good quality,as they do not fall on the field.A prototype with enlarged openings in the straw walkers,an additional grab container for maize cobs,a transport auger,a transport cleaning fan with built-in knives,a transport tube and some other assemblies was fabricated and used in *** first test results are very encouraging(94.4%-96.0%purity of maize cobs)and the system has been patented as a promising solution,especially in times when the environmental sustainability of the planet and energy independence are becoming increasingly important.

BACKGROUND Distal humerus elbow fractures are one of the most common traumatic fractures seen in pediatric patients and present as three main types:Supracondylar(SC),lateral condyle(LC),and medial epicondyle(ME)*** To evaluate the epidemiology of pediatric distal humerus fractures(SC,LC,and ME)from an American insurance claims *** A retrospective review was performed on patients 17 years and younger with the ICD 9 and 10 codes for SC,LC and ME fractures based on the IBM Truven MarketScan®Commercial and IBM Truven MarketScan Medicare Supplemental *** from 2015 to 2020 were queried for treatments,patient age,sex,length of hospitalization,and *** A total of 1133 SC,154 LC,and 124 ME fractures were *** fractures had the highest percentage of operation at 83%,followed by LC(78%)and ME fractures(41%).Male patients were,on average,older than female patients for both SC and ME *** In the insurance claims databases used,SC fractures were the most reported,followed by LC fractures,and finally ME *** was identified to be a factor for how a pediatric distal humerus fractures,with patients with SC and LC fractures being younger than those with ME *** peak age per injury per sex was similar to reported historic central tendencies,despite reported trends for younger physiologic development.

Background: We currently have international and national guidelines regarding the assessment and monitoring of clinical signs and humane endpoints in animals used in teaching and research, which make the performance of these activities mandatory for any experiment and professional working in this area. Assigning the severity of a research experiment is the result of an analysis of records of observations of the animal’s behavior, and clinical signs. The aim of this study was to describe the importance of carrying out a severity assessment associated with clinical and behavioral monitoring of rodents and rabbits during experimentation to maintain the welfare of these animals undergoing scientific research. Methods: The literature search was carried out using the following terms: “Monitoring”;“Humane endpoints”;“Animal welfare”, “Rodents”;“Rabbits”, and as connectors “and”;“or”, in the following databases: PubMed;LILACS/BIREME and SciELO. Results: A total of 987 articles were identified in the databases, and 20 of these studies were included in this review. Conclusions: Humane endpoint protocols and procedure severity tables are of the utmost importance, both from an ethical point and to refine the results of research conducted on laboratory animals. They should be drawn up jointly by the teams responsible for the project and the maintenance of the animals during the research period, and the data obtained should be published so that the scientific community can have access to it, helping to disseminate these practices, as well as helping to draw up new procedures. Monitoring and evaluating the welfare and clinical condition of animals undergoing scientific research procedures is the responsibility of the professors, researchers, veterinarians, and animal facility coordinators. Direct discharge of municipal and industrial waste waters and leachate (originating from the illegal landfills) into recipients without prior purification is unfortunately very common practice in the region of northern Kosovo. In addition, irresponsible and incorrect selection of sites for industrial and municipal dumps, which are often located in vicinity or on actual river banks, contributes significantly to environmental pollution. Analysis of the leachate from such sites was done by direct sampling and by using TCLP (Toxicity Characteristic Leashing Procedures) method. Based on the analysis of physicochemical parameters of the filtrated water from the sites Zitkovac, Grabovac and Balaban and analysis of the samples of surface water and groundwater from the site Grabovac, possible steps for removal and reduction of the existing pollution were proposed. Potential permanent solution in form of purification of municipal and industrial waste waters as well as leachate from illegal landfills was suggested. The potential implementation of remediation with a unified system for water purification, by using Membrane Bio Reactor (MBR), which includes the process of stabilization/solidification of a residual sludge, would have as an end product a neutral powder material completely safe for the environment, suitable for a variety of applications.

The effects of selected printing parameters on the fire properties of additively produced composites from neat polylactic acid(PLA)and wood/PLA filaments were *** reaction to fire of the 3D-printed specimens was tested according to the ISO 5660-1 cone calorimeter test *** results showed that the properties of the specimens when exposed to fire were significantly affected by the incorporation of wood flour into the PLA *** was also interesting that PLA specimens had much better reactions to fire than the wood/PLA *** to ignition was found to be much longer in the 3D-printed PLA *** the maximal heat release rate was a little higher in the PLA than the wood/PLA specimens,the duration of HRR was longer for the wood/PLA *** initial mass of the specimens was smaller in the wood/PLA composites,but during the radiant heat exposure the mass typically decreased slower than in the PLA specimens.

Hypertension in adolescents is, as in other patients, one of the important cardiovascular risk factors, associated with hypertension in adults and with appearance of cardiovascular complications later in life. In recent years it is in increase, in parallel with occurrence of overweight in this age group. There is mostly essential or obesity-related hypertension. It is often asymptomatic, therefore it holds true also for adolescents, that it has to be actively searched, especially in risk groups such as overweight adolescents, adolescents with positive family history or some of the other classical cardiovascular risk factors. If we have normal-weight adolescent with important hypertension or symptomatic adolescent without positive family history, the possibility of secondary hypertension has to be thinking of and extended, on possible cause focused and stepwise diagnostics performed. In this case, the treatment is etiological. Otherwise, the first-line treatment in adolescents comprises nonpharmacological treatment measures. Sometimes, because of poor nonpharmacological treatment compliance and presence of hypertensive-target organ damage, antihypertensive medications have to be introduced. However, preventive actions represent the most important task, in the form of primary and primordial prevention. In this article, diagnostic approach, prevention and treatment strategies in adolescents are shortly presented.
