Characteristics of escape mutations from occult hepatitis B virus infected patients with hematological malignancies in South Egypt

AIm To investigate the prevalence and virological characteristics of occult hepatitis B virus(HBV) infections in patients with hematological malignancies in South *** Serum samples were collected from 165 patients with hematological malignancies to monitor titers of HBV DNA, hepatitis B surface antigen(HBs Ag), and antibodies to HBV core(anti-HBc) and surface antigens. Serum samples negative for HBs Ag and positive for anti-HBc were subjected to nucleic acid extraction and HBV DNA detection by real-time polymerase chain reaction. DNA sequences spanning the S region were analyzed in cases with occult HBV infection. In vitro comparative study of constructed 1.24-fold wild type and S protein mutant HBV genotype D clones was further performed. RESULTS HBV DNA was detected in 23(42.6%) of 54 patients with hematological malignancies who were HBsA g negative, but anti-HBc positive, suggesting the presence of occult HBV infection. The complete HBV genome was retrieved from 6 occult HBV patients, and P120 T and S143 L were detected in 3 and 2 cases, respectively. Site directed mutagenesis was done to produce 1.24-fold genotype D clones with amino acid mutations T120 and L143. The in vitro analyses revealed that a lower level of extracellular HBsA g was detected by chemiluminescence enzyme immunoassay(CLEIA) with the clone containing T120 mutation, compared with the wild type or the clone with S143 L mutation despite the similar levels of extracellular and intracellular HBs Ag detected by Western blot. Southern blot experiments showed that the levels of intracellular HBV DNA were not different between these clones. CONCLUSION Occult HBV infection is common in patients with hematological malignancies and associated with P120 T and S143 L mutations. 120 T mutation impairs the detection of HBsA g by CLEIA.

AIm:To investigate characteristics of hepatitis B virus(HBV)implicated in HBV reactivation in patients with hematological malignancies receiving immunosuppressive ***:Serum samples were collected from 53 patients with hematological malignancies negative for hepatitis B surface antigen(HBsAg)before the start of and throughout the chemotherapy *** reactivation was diagnosed when the HBsAg status changed from negative to positive after the initiation of chemotherapy and/or when HBV DNA was detected by realtime detection polymerase chain reaction(RTD-PCR).For detecting the serological markers of HBV infection,HBsAg as well as antibodies to the core antigen(antiHBc)and to the surface antigen were measured in the sera by *** acids were extracted from sera,and HBV DNA sequences spanning the S gene were amplified by *** extracted DNA was further subjected to PCR to amplify the complete genome as well as the specific genomic sequences bearing the enhancerⅡ/core promoter/pre-core/core regions(nt1628-2364).Amplicons were sequenced ***:Thirty-five(66%)of the 53 HBsAg-negative patients were found to be negative serologically for antiHBc,and the remaining 18(34%)patients were positive for *** of the 53(9.4%)patients with hematologic malignancies experienced HBV *** D1 was detected in all five *** types of mutant strains were detected in the S gene product of HBV strains and were isolated from 3 patients with HBV reactivation:T/S120,L143,and *** DNA was detected in the pretreatment HBsAg-negative samples in one of the five patients with HBV *** this patient,sequences encompassing the HBV full genome obtained from sera before the start of chemotherapy and at the time of de novo HBV hepatitis were detected and it showed 100%***,in the phylogenetic tree,the sequences were clustered together,thereby indicating that this patient developed reactivation from an occult

Efficiency of Bio-Fertilizing as One of the Natural Alternatives to Improve the Growth of Khaya senegalensis and Swietenia mahagoni Trees and for Sustainability

A field experiment was carried out at Ismailia Research Station, Ismailia Governorate from 2020-2022 to improve the growth of Khaya senegalensis and Swietenia mahagoni by using a combination of mineral fertilizer (NPK) and biological fertilizer (Azotobacter chroococcum, Bacillus megatherium, and Bacillus circulant) as recommended dose under new sandy soils conditions. Split plot designed with four treatments (Control, (50% mineral fertilizer (m.) + 50% Biological fertilizer (Bio.)), 100% m. and 100% Bio.) of each species. Vegetative growth, leaf area, tree biomass, stored carbon, basal area, tree volume, and in the soil both of microbial account and mineral content were determined. The experimental results showed no significant differences between studied species among the most studied parameters except for Khaya senegalensis which gave the highest significant difference in root biomass and below-stored carbon than Swietenia mahagoni. Evidently, the highest significant growth parameters were 100% mineral fertilizer followed by (50% m. + 50% Bio.) as compared with control. No significant difference between 100% m. and (50% m. + 50% Bio.) of shoot dry biomass (15.19 and 12.02 kg, respectively) and above-stored carbon (0.28 and 0.22 mt, respectively). microbial account and mineral content in soil were improved after cultivation of tree species compared to before planting and control, especially with 50% mineral fertilizer and 50% bio-fertilizer treatment. In conclusion, a treatment containing 50% mineral fertilizer and 50% bio-fertilizer has led to the ideal Khaya senegalensis and Swietenia mahagoni growth in sandy soil for cheaper and sustainable.

Investigation of charge transfer complexes formed between 3,3′-dimethylbenzidine (o-toluidine) donor and DDQ, p-chloranil and TCNQ as π-acceptors

Charge-transfer (CT) complex of o-toluidine (o-tol) with the π-acceptor tetrachloro-p-benzoquinone (p-chloranil; CHL) has been synthesized and characterized, along with the products of the elimination reaction of o-toluidine (o-tol) with 2,3-dichloro-5,6dicyano-p-benzoquinone (DDQ) and 7,7,8,8-tetracyanoquinodimethane (TCNQ). Their properties and structures have been investigated using electronic absorption and IR spectroscopy as well as elemental analyses of the isolated compounds. The o-tol/p-CHL CT complex is shown to consist of a CHL radical anion and o-tol + radical cation. The formations of all three compounds result in the appearance of new UV-Vis spectral bands that peaked in intensity at a stoichiometric ratio of 1 : 1. 1 H-NmR and mass spectra were used to confirm the formation of the o-tol/DDQ and o-tol/TCNQ products. The new 7,8-dicyano-7′,8′-o-toluidilequinodimethane and 2,3-dichloro-5-cyano-6-o-toluidil-p-benzoquinone products resulted from the rapid reaction of o-tol with TCNQ and DDQ, respectively. Thermogravimetric studies showed that the CT-complex decomposes in three steps whereas the new compounds decompose in a single step. Thermodynamic parameters were computed from the thermal decomposition data.

Objective: To evaluate the effect of moringa oleifera leaf ethanol extract as an adjunct treatment on lead acetate induced hepatonephrotoxicity in rabbits. methods: Thirty-six male New Zealand White rabbits were assigned into two main groups. The first group(14 rabbits) served as normal control. The second group(22 rabbits) was administered orally with lead acetate at a dose of 40 mg/kg/day, 5 days/week for 8 weeks. At the 4 th and the 8 th week of treatment, 6 animals(3 animals at each period) of the second group were sacrificed while the remaining animals(16 rabbits) were assigned randomly into 2 subgroups(8 rabbits each): treated and non-treated. The first subgroup was orally given 1 m L phosphate-buffered saline for further 4 weeks while the second subgroup was administered orally with moringa oleifera leaf ethanol extract at a dose of 400 mg/kg/day for the same period. Blood samples were collected to determine hematological and serum biochemical indices. Tissue specimens were collected from the liver and kidney for evaluation of the oxidant/antioxidant markers and for histopathological ***: Lead acetate exposure decreased the mean body weight gain, hematocrit, hemoglobin, mean corpuscular volume, and lymphocytes count. moreover, it markedly increased counts of monocytes and platelets, serum enzyme activity, levels of creatinine, total cholesterol, triglycerides, and low-density lipoprotein cholesterol. malondialdehyde level was markedly increased while the reduced glutathione content was significantly decreased in liver tissue of lead intoxicated-rabbits. Histopathological alterations were also noticed in the liver and kidney of lead intoxicated rabbits. moringa oleifera leaf ethanol extract significantly improved hematological and serum biochemical parameters and histopathological structure of the liver and kidney. Preemptive living donor kidney transplantation:Access,fate,and review of the status in

BACKGROUND Preemptive living donor kidney transplantation(PLDKT)is recommended as the optimal treatment for end-stage renal *** To assess the rate of PLDKT among patients who accessed KT in our center and review the status of PLDKT in *** We performed a retrospective review of the patients who accessed KT in our center from November 2015 to November *** addition,the PLDKT status in Egypt was reviewed relative to the *** Of the 304 patients who accessed KT,32 patients(10.5%)had preemptive access to KT(PAKT).The means of age and estimated glomerular filtration rate were 31.7±13 years and 12.8±3.5 mL/min/1.73 m2,***-nine patients had KT,including 3 PLDKTs only(5.1%of total KTs and 9.4%of PAKT).Twenty-nine patients(90.6%)failed to receive PLDKT due to donor unavailability(25%),exclusion(28.6%),regression from donation(3.6%),and patient regression on starting dialysis(39.3%).In multivariate analysis,known primary kidney disease(P=0.002),patient age(P=0.031)and sex(P=0.001)were independent predictors of achievement of KT in our ***,PAKT was not significantly(P=0.065)associated with the achievement of *** of the literature revealed lower rates of PLDKT in Egypt than those in the *** Patient age,sex,and primary kidney disease are independent predictors of achieving living donor *** its non-significant effect,PAKT may enhance the low rates of *** main causes of non-achievement of PLDKT were patient regression on starting regular dialysis and donor unavailability or exclusion.

Background: In the developing world, pneumonia is not only more common than it is in Europe or North America;it is also more severe and considered the leading cause of under-five mortality. Effective management of pneumonia in children under 3-year-old-age is still challenging due to various causes. Some authors believed in non-calcemic role of vitamin D as a potential factor in the pathogenesis, prevention, or therapy of pneumonia in this specific age. Purpose: Investigate the Vitamin D status in pneumonia among children between 6 months to 3 years of age. Subjects and Methods: This study was conducted in a Pediatric point of care at Al-Zahraa University Hospital on 90 children aged 6 months to 3 years old;divided into two groups: group I (study group) including 47 patients presented with pneumonia and group II (control group) including 43 apparently healthy children of matched age and sex with no evidence of pneumonia. Prognostic Value of Combined;Cox-2, Cyclin D1 and P21 Expression in Colorectal Cancer (CRC) Patients: An Immunohistochemical Study

Background: Cyclooxygenase (COX) is a rate limiting enzyme in synthesis of prostanoids pathway and it has 2 isoforms (COX-1 and COX-2). It has many biological roles in inflammation and oncogenesis. Cox-2 was incriminated in performing disturbances in the cell cycle control in CRC, but its role of in CRC needs clarification of the mechanisms by which Cox-2 might affect the process of colorectal carcinogenesis. Cyclin D1 is an oncogene that regulates G1 phase progression to S phase of the cell cycle. Its stimulatory role on cell cycle is antagonized by Cyclin D1-dependent kinase (CDK) inhibitors like p21. P21 plays an essential role in cell cycle regulation;it may have a pro-apoptotic or an antiapoptotic role in cancer. P21 was found to have many roles in cancer;invasion metastases, cellular senescence and stem cells aging. The roles of combined expression of Cox-2, Cyclin D1 and P21 in CRC tissues and their role of prognosis and patients survival are not sufficiently clarified. Aim of the Study: To evaluate tissue expression of Cox-2, Cyclin D1 and P21 in CRC and to correlate such expression with pathological parameters, clinical and prognostic data of the patients. methods: Cox-2, Cyclin D1 and P21 are evaluated in colon cancer tissues. Correlations between their level of expressions pathological parameters, clinical and prognostic data of patients were analyzed. Results: Cox-2, Cyclin D1 over-expression was associated with higher grade, higher incidence of occurrence of lymph node & distant metastasis and advanced stage (P = 0.000). Cox-2 was related to higher tumor recurrence rate (P = 0.04) and decreased overall patients survival rate (p = 0.002). (r correlation coefficient = +0.987). Conclusion: Cox-2 and Cyclin D1 are markers of poor prognosis colon cancer patients.
