Y chromosome microdeletions in azoospermic patients with Klinefelter's syndrome

Aim： To study the occurrence of Y chromosome microdeletions in azoospermic patients with Klinefelter＇s syndrome （KFS）. Methods： Blood and semen samples were collected from azoospermic patients with KFS （n = 14） and a control group of men of proven fertility （n = 13）. Semen analysis was done according to World Health Organization （WHO） guidelines. Blood samples were processed for karyotyping, fluorescent in situ hybridization （FISH） and measurement of plasma follicle stimulating hormone （FSH） by radioimmunoassay. To determine Y chromosome microdeletions, polymerase chain reaction （PCR） of 16 sequence tagged sites （STS） and three genes （DFFRY, XKRY and RBM1 Y） was performed on isolated genomic DNA. Testicular fine needle aspiration cytology （FNAC） was done in selected cases. Results： Y chromosome microdeletions spanning the azoospermia factor （AZF）a and AZFb loci were found in four of the 14 azoospermic patients with KFS. Karyotype and FISH analysis revealed that, of the four cases showing Y chromosome microdeletion, three cases had a 47,XXY/46,XY chromosomal pattern and one case had a 46,XY/47,XXY/48,XXXY/48,XXYY chromosomal pattern. The testicular FNAC of one sample with Y chromosome microdeletion revealed Sertoli cell-only type of morphology. However, no Y chromosome microdeletions were observed in any of the 13 fertile men. All patients with KFS had elevated plasma FSH levels. Conclusion： Patients with KFS may harbor Y chromosome microdeletions and screening for these should be a part of their diagnostic work-up, particularly in those considering assisted reproductive techniques. （Asian JAndrol 2006 Jan; 8： 81-88）

Prediction of daily sediment discharge using a back propagation neural network training algorithm:A case study of the narmada River,India

Most of the studies on Artificial Neural Network(ANN)models remain restricted to smaller rivers and *** this paper,an attempt has been made to correlate variability of sediment loads with rainfall and runoff through the application of the Back Propagation Neural Network(BPNN)algorithm for a large tropical *** algorithm and simulation are done through MATLAB *** methodology comprised of a collection of data on rainfall,water discharge,and sediment discharge for the narmada River at various locations(along with time variables)and application to develop a threelayer BPNN model for the prediction of sediment *** training and validation purposes a set of549 data points for the monsoon(16 June-15 November)period of three consecutive years(1996-1998)was *** testing purposes,the BPNN model was further trained using a set of 732 data points of monsoon season of four years(2006-07 to 2009-10)at nine *** model was tested by predicting daily sediment load for the monsoon season of the year *** evaluate the performance of the BPNN model,errors were calculated by comparing the actual and predicted *** validation and testing results obtained at all these locations are tabulated and *** obtained from the model application are robust and encouraging not only for the sub-basins but also for the entire *** results suggest that the proposed model is capable of predicting the daily sediment load even at downstream locations,which show nonlinearity in the transportation ***,the proposed model with further training might be useful in the prediction of sediment discharges for large river basins.

Higher frequency of Yq microdeletions in sperm DNA as compared to DNA isolated from blood

Aim： To determine if Yq microdeletion frequency and loci of deletion are similar in two tissues （blood and sperm） of different embryological origin. Methods： The present study included 52 infertile oligozoospermic cases. In each case, DNA was isolated from blood and sperms and polymerase chain reaction （PCR） microdeletion analysis was done from genomic DNA isolated from both the tissues. The PCR products were analyzed on a 1.8% agarose gel. PCR amplifications found to be negative were repeated at least three times to confirm the deletion of a given marker. Results： Only 1 case harbored microdeletion in blood DNA, whereas 4 cases harbored microdeletion in sperm DNA. Conclusion： The frequency of Yq microdeletions is higher in germ cells as compared to blood. As the majority of infertile couples opt for assisted reproduction procreation techniques （ART）, Yq microdeletion screening from germ cells is important to understand the genetic basis of infertility, to provide comprehensive counseling and most adapted therapeutics to the infertile couple.

Submergence Analysis Using Geo-Informatics Technology for Proposed Dam Reservoirs of Par-Tapi-narmada River Link Project, Gujarat State, India

The Par-Tapi-narmada river link envisages transfer of surplus water from west flowing rivers between Par and Tapi in Gujarat State, India to water deficit areas in North Gujarat. The scheme is located mainly in southern Gujarat but it also covers part of the areas of Maharashtra, North of Mumbai on the Western Ghats of India. The main aim of Par-Tapi-narmada link is to transfer the surplus waters of Par, Auranga, Ambica and Purna River basins to take over part of narmada Canal command (Miyagam branch) after providing enroute irrigation. It is proposed that water saved in Sardar Sarovar Project, as a result of this transfer, would be taken further northwards to benefit water scarce areas of north Gujarat and also westwards in Saurashtra and Kutch regions. The construction of seven reservoirs on Par-Tapi-narmada River Link Project would affect land use/land cover, settlements and infrastructure facilities within and around reservoir area. Thus, the submergence impact analysis of all the seven reservoirs of this project have been carried out by using remote sensing and GIS techniques for planning and designing of the structures. Out of the seven reservoirs, the paper discusses submergence analysis of Kelwan Dam reservoir which is located in geologically complex region of the Dangs district. The study attempts to assess the present problems of submergence of land, forest, agriculture, settlements and infrastructure facilities by using GIS techniques for taking alternative remedial measures prior and during construction of the dams.
