Achievements and prospects of grass pea(Lathyrus sativus L.) improvement for sustainable food production

Grass pea offers an attractive choice for sustainable food production, owing to its intrinsic properties including limited water requirement and drought tolerance. However, low productivity and the presence of a neurotoxin(ODAP) have posed major obstacles to its genetic improvement. Also, biotechnological investments remain limited and the genome is complex and not well understood. Strategies that allow identification of genotypes with reduced ODAP content, coupling of low ODAP content with enhanced yield, and effective seed detoxification methods merit immediate attention. Breeder-friendly genomic tools are being increasingly made available to improve the efficiency of breeding protocols. To this end, the application of next-generation sequencing has provided a means of leveraging the repertoire of genomic resources for this somewhat neglected crop. In this review, we describe progress achieved in Lathyrus genetic improvement. We also explore potential opportunities in Lathyrus research and identify urgent research needs.

Identification,development,and application of cross-species intron-spanning markers in lentil(Lens culinaris Medik.)

Lentil(Lens culinaris Medik) is one of the most important food legumes in the world. The use in lentil of molecular marker-assisted breeding is limited, owing to the low availability of polymorphic markers. In the present study, we developed a set of polymorphic intron-spanning markers(ISMs) using a cross-species mapping approach. In this approach, putative unique transcripts(PUTs) of L. culinaris were mapped onto the Medicago truncatula genome, exploiting its closeness with the lentil genome. Spliced alignment of the PUTs resulted in a total of 25,717 alignments, allowing the development of 1703 ISMs. From these, a subset of 105 ISMs were synthesized and validated with a 51% amplification success rate in 32 lentil genotypes. Of these ISMs, 40(74%) were polymorphic and generated 2–11 alleles per locus in a genetically diverse panel of 32 lentil genotypes including wild *** set of polymorphic ISMs along with their functional annotation data will be useful in lentil breeding.

Novel CMS lines in pigeonpea[Cajanus cajan(L.)Millspaugh]derived from cytoplasmic substitutions,and their effective restoration and deployment in hybrid breeding

The availability of stable cytoplasmic male sterile(CMS or A) lines coupled with a robust restoration system(R lines) is an essential prerequisite for efficient hybrid ***-enabled hybrid technology holds immense potential to enhance the long-stagnant productivity of pigeonpea. In the present investigation, cytoplasmic substitutions were made in the nuclear backgrounds of early-maturing pigeonpea varieties or lines. Three new CMS lines(ICPL 88039 A, Pusa 992 A, and DPP 3-2A) resulted from genetic crosses involving cytoplasmic donors from A2(GT 288A) and A4(ICPA 2089) categories. In addition to visual inspection of anthers, pollen-staining techniques and scanning electron microscopy(SEM)analysis were used to confirm pollen sterility. Further, given the relevance of the plant mitochondrial genome to CMS manifestation, 25 mitochondrion-specific DNA markers were assayed on these newly developed A lines and isogenic maintainer(B) lines. DNA polymorphism between Pusa 992 A and Pusa 992 B as revealed by the nad7a_del marker confirmed the successful combination of sterilizing cytoplasm(A4) and nonrestoring nuclear background(Pusa 992). Such cytoplasm-specific DNA markers are required for A2-CMS as well. Further, to assess restoration ability, potential restorers were crossed with these CMS lines, and as a consequence, promising A × R combinations exhibiting 100%pollen fertility could be identified. In parallel, we also analyzed the inheritance patterns underlying fertility restoration using ICPL 88039A-derived F2 and BC1F1populations,and established a monogenic dominant model to explain the phenomenon of A2-CMS restoration. In summary, we report the successful development of new CMS lines and describe their effective deployment in hybrid breeding of pigeonpea.

Beach pea or beach cowpea(Vigna marina(Burm.)Merr.)belongs to the family *** is a close relative of cultivated Vigna species such as adzuki bean(***),cowpea(***),mung bean(***),and blackgram(***),and is distributed throughout the *** its ability to tolerate salt stress,beach pea has great potential to contribute salt-tolerance genes for developing salt-tolerant cultivars in cultivated Vigna ***,it is still underutilized in Vigna breeding programs.A draft genome sequence of beach pea was generated using a high-throughput next-generation sequencing platform,yielding 23.7 Gb of sequence from 79,929,868 filtered reads.A de novo genome assembly containing 68,731 scaffolds gave an N50 length of 10,272 bp and the assembled sequences totaled 365.6 Mb.A total of 35,448 SSRs,including 3574 compound SSRs,were identified and primer pairs for most of these SSRs were *** analysis identified 50,670 genes with mean coding sequence length 1042 *** analysis revealed highest sequence similarity with ***,followed by *** with the *** genome revealed 16,699 SNPs and 2253 InDels and comparison with the *** genome revealed 17,538 SNPs and 2300 *** our knowledge this is the first draft genome sequence of beach pea derived from an accession(ANBp-14-03)adapted locally in the Andaman and Nicobar Islands of *** draft genome sequence may facilitate the genetic enhancement in cultivated Vigna species.

Ameliorative Effects of Paclobutrazol via Physio-Biochemical and Molecular Manifestation in Rice under Water Deficit Stress

To comprehensively explore the physio-biochemical and molecular changes of paclobutrazol(PBZ) at the ideal dose under water deficit stress(WDS) conditions, we investigated the effects of 100 mg/kg PBZ applied via drenching on various physio-biochemical and molecular parameters in three rice varieties(N22, IR64, and IR64 DTY1.1) under both mild [75%-80% relative water content(RWC)] and severe(60%-65% RWC) WDS conditions. The results showed that PBZ treatment positively influenced the physio-biochemical parameters, significantly increasing dry matter(16.27%-61.91%), RWC(6.48%-16.34%), membrane stability index(4.37%-10.35%), and total chlorophyll content(8.97%-29.09%) in the rice varieties under both mild and severe WDS. Moreover, PBZ treatment reduced drought susceptibility(0.83-0.95) and enhanced drought tolerance efficiency(60.92%-86.78%), indicating its potential as a stress-mitigating agent. Global methylation analysis revealed changes in DNA methylation patterns, indicating the regulatory influence of PBZ on gene expression. The expression analysis of genes involved in the diversification of geranylgeranyl pyrophosphate towards the biosynthesis of abscisic acid, gibberellin acid, and chlorophyll showed alterations in their expression levels, suggesting the involvement of PBZ in the isoprenoid pathway. Overall, this study provides valuable insights into the potential mechanisms by which PBZ modulates physiological and molecular responses in rice plants under WDS. The findings highlight the importance of PBZ as a promising agent for enhancing drought tolerance in rice and offer valuable information for future research in crop stress management.

Spatio-temporal variation and propagation direction of coal fire in Jharia Coalfield,India by satellite-based multi-temporal night-time land surface temperature imaging

In this paper,the spatio-temporal variation and propagation direction of coal fire were studied in the Jharia Coalfield(JCF),India during 2006–2015 through satellite-based night-time land surface temperature(LST)*** LST was retrieved from Advanced Spaceborne Thermal Emission and Reflection Radiometer(ASTER)night-time thermal-infrared data by a robust split-window algorithm based on scene-specific regression coefficients,band-specific hybrid emissivity,and night-time atmospheric *** LST-profile-based coal fire detection algorithm was formulated through statistical analysis of the LST values along multiple transects across diverse coal fire locations in the JCF in order to compute date-specific threshold temperatures for separating thermally-anomalous and background *** algorithm efficiently separates surface fire,subsurface fire,and thermally-anomalous transitional *** the observation period,it was noticed that the coal fire area increased significantly,which resulted from new coal fire at many places owing to extensive opencast-mining *** was observed that the fire propagation occurred primarily along the dip direction of the coal *** places,lateral-propagation of limited spatial extent was also observed along the strike direction possibly due to spatial continuity of the coal seams along ***,the opencast-mining activities carried out during 2009–2015 and the structurally weak planes facilitated the fire propagation.

Cd-bearing solid wastes are considered to be a serious threat to the environment,and effective strategies for their treatment are urgently *** sintering has been considered as a promising method for efficiently incorporating heavy metal-containing solid wastes into various ceramic ***-rich dredged river sediment,especially Al and Si-containing oxides,can be treated as alternative ceramic precursors rather than being disposed of as solid *** examine the feasibility of using waste sediment for Cd stabilization and the phase transition mechanisms,this study conducted a sintering scheme for the mixtures of CdO and dredged river sediment with different(Al+Si):Cd mole *** investigations have been performed on phases transformation,Cd incorporation mechanisms,elemental distribution,and leaching behaviors of the sintered *** showed that Cd incorporation and transformation in the sintered products were influenced by the mole ratio of(Al+Si):*** the high-Cd series((Al+Si):Cd=6:1),CdSiO_(3),Cd_(2)SiO_(4),CdAl_(2)(SiO_(4))_(2) and Cd_(2)Al_(2)Si_(2)O_(9) were predominant Cd-containing product phases,while Cd2Al2Si2O9 was replaced by CdAl_(4)O_(7) when the mole ratio of(Al+Si):Cd was 12:1(low-Cd series).Cd was efficiently stabilized in both reaction series after being sintered at≥900℃,with<5%leached ratio even after a prolonged leaching time,indicating excellent long-term Cd *** study demonstrated that both Cd-containing phases and the amorphous Al-/Si-containing matrices all played critical roles in Cd stabilization.A promising strategy can be proposed to simultaneously reuse the solid waste as ceramic precursors and stabilize heavy metals in the ceramic products.

Intestinal loop stoma is a common surgical procedure performed for various benign and malignant abdominal problems,but it rarely undergoes spontaneous closure,without surgical *** male patients presented to our emergency surgical department with acute abdominal *** of them was diagnosed as having rectosigmoid perforation and underwent diversion sigmoid loop colostomy after primary closure of the *** other was a known case of carcinoma of the rectum who had already undergone low anterior resection with covering loop ileostomy;the patient underwent second loop ileostomy,this time for complicated intestinal *** our surprise,both the loop colostomy and ileostomy closed spontaneously at 8 weeks and 6 weeks,respectively,without any *** stoma closure is a rare and interesting *** exact etiology for spontaneous closure remains unknown,but it may be hypothesized to result from slow retraction of the stoma,added to the concept of a tendency towards spontaneous closure of enterocutaneous fistula.
