Association of ITPA polymorphism with outcomes of peginterferon-α plus ribavirin combination therapy

AIM:To analyzed the association between inosine triphosphatase(ITPA)(rs1127354) genotypes and sustained virological response(SVR) rates in peginterferon(Peg-IFN)α + ribavirin(RBV) ***:Patients who underwent Peg-IFNα + RBV combination therapy were enrolled(n = 120) and they had no history of other IFN-based *** in hemoglobin levels during therapy,cumulative reduction of RBV dose,frequency of treatment withdrawal,and SVR rates were investigated in each ITPA ***:In patients with ITPA CC genotype,hemoglobin decline was significantly greater and the percentage of patients in whom total RBV dose was < 60% of standard and/or treatment was withdrawn was significantly higher compared with CA/AA ***,SVR rates were equivalent between CC and CA/AA genotypes,and within a subset of patients with Interleukin 28B(IL28B)(rs8099917) TT genotype,SVR rates tended to be higher in patients with ITPA CC genotype,although the difference was not ***:ITPA CC genotype was a disadvantageous factor for Peg-IFNα + RBV treatment in relation to completion rates and RBV ***,CC genotype was not inferior to CA/AA genotype for SVR *** full-length treatment is accomplished,it is plausible that more SVR is achieved in patients with ITPA CC variant,especially in a background of IL28B TT genotype.

AIM： To evaluate the role of intestinal microflora in the effects of multi-herbal medicine on gene expression in the gut and liver. METHODS： The multi-herbal medicine Juzentaihoto （JTX） was administered to five germ-free mice and regular mice for 2 wk. Among the results of the comprehensive gene chip analysis of the intestine and liver, we featured heat shock proteins （HSPs） 70 and 105 because their gene expression changed only in the presence of microflora. Real-time RT-PCR was performed to confirm the expression levels of these HSP genes. To determine whether JTX acts directly on the HSP genes, sodium arsenite （SA） was used to induce the heat shock proteins directly. To examine the change of the intestinal microflora with administration of JTX, the terminal restriction fragment polymorphism （T-RFLP） method was used. To identify the changed bacteria, DNA sequencing was *** by gene chip and real-time RT-PCR, changed with the administration of JTX in the regular mice but not in the germ-free mice. JTX did not suppress the direct induction of the HSPs by SA. T-RFLP suggested that JTX decreased unculturable bacteria and increased Lactobacillus johnsoni. These data suggested that JTX changed the intestinal microflora which, in turn, changed HSP gene ***： Intestinal microflora affects multi-herbal product JTX on the gene expression in the gut and liver.

Objective The changes in serum adipokines and cytokines related to oxidative stress were examined during 3 months ‘Off to On’ and ‘On to Off’ periods using negatively charged particle-dominant indoor air conditions（NCPDIAC）.Methods Seven volunteers participated in the study,which included ‘OFF to 3 months ON’ periods（ON trials） for a total of 16 times,and ‘ON to 3 months OFF’（OFF trials） periods for a total of 13 *** With the exception of one case,serum amyloid A（SAA） levels decreased significantly during the ON *** Considering that SAA is an acute phase reactive protein such as C reactive protein（CRP）,this observed decrease might indicate the prevention of cardiovascular and atherosclerotic changes,since an increase in high-sensitive CRP is associated with the subsequent detection of these events.

Objective The custom-homebuilding company, Cosmic Garden Co. Ltd., located in Okayama City, Japan was established in 1997 and uses specific natural ore powder （SNOP） in wall materials and surveys customers in order to improve allergic symptoms. Methods To investigate the biological effects of SNOP, patients with a pollen allergy were recruited to stay in a room surrounded by cloth containing SNOP （CCSNOP）, and their symptoms and various biological parameters were compared with those of individuals staying in a room surrounded by control non-woven cloth （NWC）. Each stay lasted 60 min. Before and immediately after the stay, a questionnaire regarding allergic symptoms, as well as POMS （Profile of Mood Status） and blood sampling, was performed. Post-stay minus pre-stay values were calculated and compared between CCSNOP and NWC groups. Results Results indicated that some symptoms, such as nasal obstruction and lacrimation, improved, and POMS evaluation showed that patients were calmer following a stay in CCSNOP. Relative eosinophils, non-specific Ig E, epidermal growth factor, monocyte chemotactic protein-I, and tumor necrosis factor-a increased following a stay in CCSNOP. Conclusion This ore powder improved allergic symptoms, and long-term monitoring involving 1 to 2 months may be necessary to fully explore the biological and physical effects of SNOP on allergic patients.

Relationship between Self-Care and Hand Foot Syndrome Specific Quality of Life in Cancer Patients

Purpose: The purpose of this study was to describe adherence to self-care for hand-foot syndrome (HFS) and the relationship between self-care and health-related quality of life (QOL). Methods: A cross-sectional study was conducted on 105 consecutive outpatients with HFS. To assess self-care for HFS and QOL, the participants were asked to complete self-care and HFS-14 (hand-foot syndrome specific QOL) questionnaires, respectively. Multiple regression models were used to identify the relationship between self-care and QOL. Results: Adherence to self-care for HFS was low, less than 50% for most of the items. Multivariate analysis revealed that poorer self-care (βs = -0.19;P = 0.03), depressive symptoms (βs = 0.43;P βs = 0.20;P = 0.04) were independent predictors of poorer QOL scores. Conclusions: We have identified a need for further research to develop effective programs of self-care in HFS.

Tomato Seed Extract Containing Lycoperoside H Improves Skin Elasticity in Japanese Female Subjects: A Randomized, Placebo-Controlled, Double-Blind Trial

Background and Objective: Tomato seeds are edible seeds unconsciously ingested with the fruit. However, there are few reports regarding the constituents and biological activities of tomato seed extract (TSE). Recently, we found that saponins are major constituents of TSE including lycoperoside H. Previous reports have described that several plant-derived saponins improve skin diseases such as wounds and microangiopathy. Therefore, to discover the effect of TSE on the skin condition, we conducted a clinical trial of TSE (Tomato Seed Extract-P) standardized with lycoperoside H when orally ingested. Methods: The study was performed as a randomized, double-blind, placebo-controlled study. TSE (200 mg daily) containing 1 mg of lycoperoside H was used as the active sample. We enrolled 44 Japanese women who have concerns about facial elasticity and relatively low facial skin elasticity. All subjects were randomly allocated into either the active group (n = 22) or the placebo group (n = 22) using a computerized random-number generator. Capsules containing either the active sample or a placebo were administered for 8 weeks between October 12, 2020, and January 16, 2021. Facial elasticity, specifically the R7 value, was evaluated as the primary outcome. The remaining facial R parameters, upper arm R parameters, and other skin parameters including epidermal moisture, trans epidermal water loss, dermal parameters, and advanced glycation end products (AGEs) parameters were measured at 0, 4, and 8 weeks of ingestion. Blood, urine, and body parameters were also evaluated for safety. Results: Forty-three subjects completed the trial, and the per protocol set comprised 21 subjects in the TSE group and 22 subjects in the placebo group. After ingesting TSE for 8 weeks, the R7 value was significantly higher in the TSE group compared to the placebo group. Subsurface exploration is one of the most ambitious scientific objectives of the Hayabusa2 mission.A small device called small carry-on impactor(SCI)was developed to create an artificial crater on the surface of asteroid *** enables us to sample subsurface materials,which will provide a window to the *** physical properties of the resulting crater are also useful for understanding the internal structure of *** understanding of the crater and ejecta properties,including the depth of excavation of subsurface materials,requires accurate information on impact *** particular,the impact angle is a critical factor because it greatly influences the size and shape of the *** April 5,2019,the Hayabusa2 spacecraft deployed the SCI at 500 m of altitude above the asteroid *** SCI gradually reduced its altitude,and it shot a 2 kg copper projectile into the asteroid 40 min after *** the position of the released SCI is essential for determining the impact *** study describes the motion reconstruction of the SCI based on the actual operation *** results indicate that the SCI was released with high accuracy.

The Japanese interplanetary probe Hayabusa2 was launched on December 3,2014 and the probe arrived at the vicinity of asteroid 162173 Ryugu on June 27,*** its 1.4 years of asteroid proximity phase,the probe successfully accomplished numbers of record-breaking achievements including two touchdowns and one artificial cratering experiment,which are highly expected to have secured surface and subsurface samples from the asteroid inside its sample container for the first time in *** Hayabusa2 spacecraft was designed not to orbit but to hover above the asteroid along the sub Earth *** orbital and geometrical configuration allows the spacecraft to utilize its high-gain antennas for telecommunication with the ground station on Earth while pointing its scientific observation and navigation sensors at the *** paper focuses on the regular station-keeping operation of Hayabusa2,which is called“home position”(HP)-keeping ***,together with the spacecraft design,an operation scheme called HP navigation(HPNAV),which includes a daily trajectory control and scientific observations as regular activities,is *** the description on the guidance,navigation,and control design as well as the framework of optical and radiometric navigation,the results of the HP-keeping operation including trajectory estimation and delta-V planning during the entire asteroid proximity phase are summarized and evaluated as a first ***,this paper states that the *** operation in the framework of HPNAV had succeeded without critical incidents,and the number of trajectory control delta-V was planned fficiently throughout the period.
