A Study of Rb Gene in Oral Squamous Cell Carcinoma

Objective To study abnormalities of Rb gene in the initiation of oral squamous cell carccinoma (SCC), and the relationship between deletion of Rb gene and some clinico pathological parameters. Methods DIG High Prime labeled Rb 3 6 kb DNA probe was used to detect 28 cases of oral squamous cell carcinomas digested by restriction endonuclease EcoRI using Southern blotting technique. 15 males and 13 females, aged from 34 to 69 patients were enrolled in this study. Results The structural abnormal alterations of Rb gene were found in 7 cases of total samples:6 cases with deletion of 5.4, 4.1 and 2.3 kb fragments and one case was rearrangement. No correlation was found with sex, age, site of primary tumor, level of histopathological differentiation and TNM staging on further analyses. Conclusion The abnormalities of Rb gene may be associated with carcinogenesis of oral SCC.

＜正＞ The time course changes of basic fibroblast growth factor （bFGF） expression in- duced by hypoxia and the effectsof genistein on hypoxia - induced bFGF expression in the human retinal pigment epithelium （RPE） cells werestudied. Hypoxia significantly increased bFGF mRNA expression. The maximal level detected at 24 h was abouttwo times of that at the start of treatment. With pre - treatment of genistein for 30 min. the elevated expression ofbFGF mRNA was suppressed in a concentration - dependent manner. bFGF mRNA expression was reduced to30. 4% by 200 uM genistein when compared with that untreated with genistein. Hypoxia treatment alsoremarkably increased the expression of bFGF protein. At 24 h after hypoxia. the highest expression of bFGF protein was observed, it was about two times as much as that at the start of treatment. Genistein could alsosuppress bFGF protein expression in a concentration- dependent manner. The highest suppression was observedwhen exposed to 200 uM genistein. which was 43 % of control. These results suggested that suppression of bFGF expression in RPE cells might partly account for the inhibitive effect of genistein on retinal neovascularizationinvivo.

hCG/ATP-induced Leydig cell calcium channel reaction at different extracellular calcium ion concentration

Objective: To study the calcium channel reaction of human Leydig cells induced by hCG/ATP at different extracellular calcium ion concentrations. Methods: The Leydig cell calcium ion concentration was examined with laser confocal microscope, when the cells were stimulated with hCG/ATP at different extracellular calcium contrations. Results: With calcium-containing extracellular fluid, the Leydig cells were sensitive to hCG stimulation and when the extracellular fluid was calcium-free, the Leydig cells did not respond to the stimulation. However, the Leydig cells did respond to ATP stimulation no matter the extracellular fluid contained calcium or not. Conclusion: In human Leydig cells, there are calcium channels sensitive to hCG and ATP. The extracellular calcium ion concentration plays an important role in the regulation of Leydig cell metabolism by hCG/ATP.
