Differences and identification on multi-time hydrocarbon generation of carboniferous-permian coaly source rocks in the Huanghua Depression,Bohai Bay Basin

Coal is a solid combustible mineral,and coal-bearing strata have important hydrocarbon generation potential and contribute to more than 12%of the global hydrocarbon resources.However,the deposition and hydrocarbon evolution process of ancient coal-bearing strata is characterized by multiple geological times,leading to obvious distinctions in their hydrocarbon generation potential,geological processes,and production,which affect the evaluation and exploration of hydrocarbon resources derived from coaly source rocks worldwide.This study aimed to identify the differences on oil-generated parent macerals and the production of oil generated from different coaly source rocks and through different oil generation processes.Integrating with the analysis of previous tectonic burial history and hydrocarbon generation history,high-temperature and high-pressure thermal simulation experiments,organic geochemistry,and organic petrology were performed on the Carboniferous-Permian(C-P)coaly source rocks in the Huanghua Depression,Bohai Bay Basin.The oil-generated parent macerals of coal's secondary oil generation process(SOGP)were mainly hydrogen-rich collotelinite,collodetrinite,sporinite,and cutinite,while the oil-generated parent macerals of tertiary oil generation process(TOGP)were the remaining small amount of hydrogen-rich collotelinite,sporinite,and cutinite,as well as dispersed soluble organic matter and unexhausted residual hydrocarbons.Compared with coal,the oil-generated parent macerals of coaly shale SOGP were mostly sporinite and cutinite.And part of hydrogen-poor vitrinite,lacking hydrocarbon-rich macerals,and macerals of the TOGP,in addition to some remaining cutinite and a small amount of crude oil and bitumen from SOGP contributed to the oil yield.The results indicated that the changes in oil yield had a good junction between SOGP and TOGP,both coal and coaly shale had higher SOGP aborted oil yield than TOGP starting yield,and coaly shale TOGP peak oil yield was lower than SOGP peak oil yield.There were significa

Is there a great unconformity between Xiamaling and Longshan formations in the North China Craton?

The existence of a 0.3 Ga"great unconformity"between the Mesoproterozoic"Undefined System"Xiamaling Formation and the"Neoproterozoic"Longshan Formation has long been controversial.In this study,stratigraphy,sedimentology,detrital zircon dating,lithium isotope,and major and trace elements were applied to systematically analyze their relationship.Results demonstrate that coarse to fine sandstone-mudstone normal cycles with different grades and thicknesses exist from the Xiamaling to the Longshan formations.Sedimentary succession formed from a restricted platform with lagoon-shallow marine shelf-tidal flat-beach and tidal shoal developing in ascending order.The pebbly sandstone at the bottom of the Long shan Formation does not have the properties of basal conglomerate with tectonic significance.The youngest detrital zircon is older than 1.6 Ga for the Long shan Formation and overlying Jing'eryu Formation sandstones.In addition,no(zircon)provenance has been found from the large igneous province of 1.32-1.2 Ga in Xiamaling Formation.Theδ^(7)Li isotope values exhibit a gradually increasing trend.The Li content and CIA(Chemical index of alteration)gradually decreases from the top of the Xiamaling Formation to the lower part of the Long shan Formation.Simultaneously,as K_(2)O/Al_(2)O_(3)and FeO/Fe_(2)O_(3)decreases,Na_(2)O/Al_(2)O_(3)increases,and Li/Al,V/Cr,and V/(V+Ni)fluctuates slightly.At the boundary,the Ce/Ce^(*)and Eu/Eu^(*)show positive and negative anomalies,respectively.Further,the Rb/Sr and Al/Si values decrease,and U/Th values increase.This indicates that the geochemistry of the weathering crust at the interface of the Xiamaling Formation-Long shan Formation is not characteristic.However,it indicates a weakening of continental weathering intensity from the bottom to the top of the interface.These results do not support the existence of a large unconformity between the Xiamaling and the Longshan formations.Therefore,it is suggested that the Long shan Formation-Jing'eryu Formation should be moved to the lower l

Genetic technologies for extremely thermophilic microorganisms of Sulfolobus, the only genetically tractable genus of crenarchaea

Archaea represents the third domain of life, with the information-processing machineries more closely resembling those of eukaryotes than the machineries of the bacterial counterparts but sharing metabolic pathways with organisms of Bacteria, the sister prokaryotic phylum. Archaeal organisms also possess unique features as revealed by genomics and genome comparisons and by biochemical characterization of prominent enzymes. Nevertheless, diverse genetic tools are required for in vivo experiments to verify these interesting discoveries. Considerable efforts have been devoted to the development of genetic tools for archaea ever since their discovery, and great progress has been made in the creation of archaeal genetic tools in the past decade. Versatile genetic toolboxes are now available for several archaeal models, among which Sulfolobus microorganisms are the only genus representing Crenarchaeota because all the remaining genera are from Euryarchaeota. Nevertheless, genetic tools developed for Sulfolobus are probably the most versatile among all archaeal models, and these include viral and plasmid shuttle vectors, conventional and novel genetic manipulation methods, CRISPR-based gene deletion and mutagenesis, and gene silencing, among which CRISPR tools have been reported only for Sulfolobus thus far. In this review, we summarize recent developments in all these useful genetic tools and discuss their possible application to research into archaeal biology by means of Sulfolobus models.

Neonatal Brain Abscess:Case Report of 12 Cases and Literature Review

Objective:To investigate the etiological characteristics,clinical manifestations,and early identification methods of neonatal brain abscess.Methods:The baseline characteristics,clinical manifestations,and laboratory results of 12 neonatal brain abscess cases were retrospectively analyzed.Results:The clinical manifestations were fever,convulsion,and lethargy.A small number of them had respiratory and circulatory failure.The diagnosis made was based on imaging examination.All 12 cases were confirmed by cranial enhanced computed tomography(CT)or magnetic resonance imaging(MRI).Blood cultures of 9 cases were positive,with Escherichia coli in 6 cases,β-hemolytic Streptococcus in 1 case,methicillin-resistant Staphylococcus aureus in 1 case,and Enterococcus faecium in 1 case.However,only 3 of them had positive cerebrospinal fluid(CSF)cultures.All the 12 neonates were treated with antibiotic therapy upon admission,with only 3 cases treated with surgery.Among them,4 recovered and were discharged,while the remaining 8 discontinued their therapy.Conclusion:Escherichia coli is the most common pathogen of neonatal brain abscess in our study.The clinical manifestations of neonatal brain abscess are atypical,and the prognosis is poor.Respiratory and circulatory failure in children with intracranial infection may indicate the presence of brain abscess.For children with suspected brain abscess,cranial enhanced CT or MRI should be performed as soon as possible to make an early diagnosis.The prevention of brain abscess should be prioritized;neonates with sepsis or meningitis should receive prompt and strong antibiotic therapy in an effort to prevent the development of brain abscess.

基于PCA-SVM结合共聚焦拉曼光谱的特级初榨橄榄油掺伪压榨菜籽油定量分析

为了促进国内橄榄油市场的健康发展,对掺伪同样存在天然类胡萝卜素的低温压榨菜籽油的特级初榨橄榄油进行了定量鉴别研究。采用共聚焦拉曼光谱技术对不同掺伪浓度油样进行测试,基于密度泛函理论对油样的拉曼光谱峰的归属进行了理论分析,并对拉曼光谱数据进行主成分分析(PCA),然后利用支持向量机(SVM)构建PCA-SVM模型。另外,对PCA-SVM模型的检出限进行了研究。结果表明:特级初榨橄榄油与低温压榨菜籽油的拉曼光谱存在一定差异,最明显的光谱差异主要集中在谱峰1008、1161、1528 cm^(-1)和谱段2800~3000 cm^(-1)内,与密度泛函理论对不同油样拉曼光谱峰的分析一致;不考虑类胡萝卜素特征信号建立的PCA-SVM模型决定系数大于0.989,均方根误差小于2.990%,检出限为2%(低温压榨菜籽油体积分数);在特级初榨橄榄油掺伪定量分析中,考虑类胡萝卜素的特征信号有助于提高模型预测精度,但仅限于掺伪低价植物油中无类胡萝卜素存在的情况;PCA-SVM模型在不考虑类胡萝卜素特征信号的情况下依然具有良好的定量预测效果。综上,所建立的PCA-SVM模型可以用于掺伪2%以上低温压榨菜籽油的特级初榨橄榄油的定量鉴别。

Reprogramming Mycobacterium tuberculosis CRISPR System for Gene Editing and Genomewide RNA Interference Screening

Mycobacterium tuberculosis is the causative agent of tuberculosis(TB), which is still the leading cause of mortality from a single infectious disease worldwide. The development of novel anti-TB drugs and vaccines is severely hampered by the complicated and time-consuming genetic manipulation techniques for M. tuberculosis. Here, we harnessed an endogenous type Ⅲ-A CRISPR/Cas10 system of M. tuberculosis for efficient gene editing and RNA interference(RNAi).This simple and easy method only needs to transform a single mini-CRISPR array plasmid, thus avoiding the introduction of exogenous protein and minimizing proteotoxicity. We demonstrated that M. tuberculosis genes can be efficiently and specifically knocked in/out by this system as confirmed by DNA high-throughput sequencing. This system was further applied to single-and multiple-gene RNAi. Moreover, we successfully performed genome-wide RNAi screening to identify M. tuberculosis genes regulating in vitro and intracellular growth. This system can be extensively used for exploring the functional genomics of M. tuberculosis and facilitate the development of novel anti-TB drugs and vaccines.

Mesozoic basins and associated palaeogeographic evolution in North China

In North China, the Mesozoic terrestrial basins, sedimentary palaeogeography and tectonic settings involved five evolutionary stages：（1） the Early-Middle Triassic, （2） the Late Triassic to Early-Middle Jurassic, （3） the Late Jurassic to early Early Cretaceous, （4） the middle-late Early Cretaceous and （5） the Late Cretaceous. The regional punctuated tectonic events occurred during these evolutionary stages. During the Early-Middle Triassic （stage 1）, the Xingmeng Orogenic Belt （XMOB, i.e., east- ern part of Central Asia Orogenic Belt, CAOB） of the northern North China was settled in the transition of tectonic environment from syn-orogenic compression to post-orogenic extension with intensive uplifting. It is a main provenance in the unified Ordos-North China Basin.The united continental plate of China and the Qinling-Dabie-Sulu Orogenic Belt formed due to convergence and collision between the North China Plate and the Yangtze Plate along two suture zones of the Mianlue and the Shangdan in the terminal Middle Triassic. During the Late Triassic to the Early-Middle Jurassic （stage 2）, the Late Triassic mafic or alkaline rocks and intrusions occurred on the northern and southern margins of North China Craton （NCC） and XMOB, implying that intensified extension happened all over the North China （early phase of stage 2）. Additionally, in the late phase of stage 2, the basic volcanic- filing faulted basins were widely distributed in the northeastern North China during the Early- Middle Jurassic, including a series of smalt- to medium-sized basins with coal-bearing strata and some volcanic rocks in other areas of North China, which was the result of subduction of the Palaeo-Pacific Plate during the Early-Middle Jurassic. An active continental margin with accretionary complex developed in the eastern Heilongjiang of China, Japan and the Far East of Russia at that time. Prediction of potential antimalarial targets of artemisinin based on protein information from whole genome of Plasmodium falciparum

On the basis of the genomic data and protein pathway information about Plasmodium falciparum clone 3D7 from the NCBI taxonomy database and the KEGG database, eight key protein enzymes in the signal pathways were selected to perform molecular docking with artemisinin. The binding modes obtained from the molecular docking suggested that purine nucleoside phosphorylase (pfPNP), peptide deformylase (pfPDF), and ribose 5-phosphate isomerase (pfRpiA) may be involved in the antimalarial mode of action of artemisinin. Artemisinin exhibited its antimalarial activity probably by interfering with the metabolic pathways of purine, pyrimidine, methionine, glyoxylate and dicarboxylate, or pentose phosphate.

Detecting putative recombination events of hepatitis B virus: An updated comparative genome analysis

An updated collection of 791 human hepatitis B virus (HBV) genomes and 38 non-human primate HBV genomes was analyzed for identifying putative recombination events and their recombinants by using two bioinformatics software tools: Simplot and RDP3 with five algorithms (RDP, GENECONV, MaxChi, Chimaera, and SiScan). A total of 61 recombinants from nine putative recombination events were detected with RDP3, especially the breakpoints of six events which have both two parental sequences that can be determined precisely with Simplot. To our knowledge, 53 recombinants were found for the first time. Our study also suggests that a relatively high recombination frequency occurs in the PreC/C gene region and the position near gene boundaries.
