Themis regulates metabolic signaling and effector functions in CD4^(+) T cells by controlling NFAT nuclear translocation

CD8^(+)T cells play a central role in antiviral immune *** infection,naive CD8^(+)T cells differentiate into effector cells to eliminate virus-infected cells,and some of these effector cells further differentiate into memory cells to provide long-term protection after infection is *** extensively investigated,the underlying mechanisms of CD+T-cell differentiation remain incompletely *** is a T-cell-specific protein that plays critical roles in T-cell *** studies using Themis T-cell conditional knockout mice also demonstrated that Themis is required to promote mature CD8^(+)T-cell homeostasis,cytokine responsiveness,and antibacterial *** this study,we used LCMV Armstrong infection as a probe to explore the role of Themis in viral *** found that preexisting CD8^(+)T-cell homeostasis defects and cytokine hyporesponsiveness do not impair viral clearance in Themis T-cell conditional knockout *** analyses showed that in the primary immune response,Themis deficiency promoted the differentiation of CD8^(+)effector cells and increased their TNF and IFNy ***,Themis deficiency impaired memory precursor cell(MPEC)differentiation but promoted short-lived effector cell(SLEC)*** deficiency also enhanced effector cytokine production in memory CD8^(+)T cells while impairing central memory CD8^(+)T-cell ***,we found that Themis mediates PD-1 expression and its signaling in effector CD8^(+)T cells,which explains the elevated cytokine production in these cells when Themis is disrupted.

Characterization and Propagation of Historical and Projected Droughts in the Umatilla River Basin, Oregon, USA

Climate change is expected to have long-term impacts on drought and wildfire risks in Oregon as summers continue to become warmer and drier. This paper investigates the projected changes in drought characteristics and drought propagation in the Umatilla River Basin in northeastern Oregon for mid-century(2030–2059) and late-century(2070–2099) climate scenarios. Drought characteristics for projected climates were determined using downscaled CMIP5 climate datasets from ten climate models and Soil and Water Assessment Tool to simulate effects on hydrologic processes. Short-term(three months) drought characteristics(frequency, duration, and severity) were analyzed using four drought indices, including the Standardized Precipitation Index(SPI-3), Standardized Precipitation-Evapotranspiration Index(SPEI-3), Standardized Streamflow Index(SSI-3), and the Standardized Soil Moisture Index(SSMI-3). Results indicate that short-term meteorological droughts are projected to become more prevalent, with up to a 20% increase in the frequency of SPI-3drought events. Short-term hydrological droughts are projected to become more frequent(average increase of 11% in frequency of SSI-3 drought events), more severe, and longer in duration(average increase of 8% for short-term droughts).Similarly, short-term agricultural droughts are projected to become more frequent(average increase of 28% in frequency of SSMI-3 drought events) but slightly shorter in duration(average decrease of 4%) in the future. Historically, drought propagation time from meteorological to hydrological drought is shorter than from meteorological to agricultural drought in most sub-basins. For the projected climate scenarios, the decrease in drought propagation time will likely stress the timing and capacity of water supply in the basin for irrigation and other uses.

The heterogeneous loss of HO_(2)radicals onto ambient aerosols plays an important role in tropospheric ***,sparse investigation of the dominating parameters controlling the HO_(2)uptake coefficients onto ambient aerosols(γHO_(2))has largely hindered the application of the measuredγHO_(2)to the global spatial *** we induced an offine method using LFP-LIF technique to measure the kinetics of HO_(2)uptake onto ambient aerosols collected in summertime and wintertime in Yokohama city,a regional urban site near Tokyo,*** controlling the dominating parameters which infuenceγHO_(2),we were able to investigate the detailed HO_(2)uptake *** characterized the chemical composition of the collected ambient aerosols,including organics,inorganics,transition metals ions,*** modeledγHO_(2)using different *** show thatγHO_(2)increased with the increase in RH,and the aerosol states(“dry”or wet/aqueous)have large effects onγHO_(2).With fixed RH and aerosol chemical composition,γHO_(2)was highly dependent on pH and inversely correlated with[HO_(2)]_*** combing the measuredγHO_(2)values with the modeled ones,we found that both the HO_(2)self-reaction and transition metal-catalyzed reactions should be accounted for to yield a single parameterization to predictγHO_(2),and different chemical compositions may have collective effects onγHO_(2).Results may serve for extending theγHO_(2)values measured at one observation site to different environmental conditions,which will help us to achieve more accurate modeling results concerning secondary pollutant formation(i.e.,ozone).

Outcome of patients with traumatic cranial nerve palsy admitted to a university hospital in Nepal

Background Cranial nerve palsy (CNP) is a common complication of traumatic brain injury (TBI). Despite a high incidence of TBI in Nepal (382 per 100,000), literature on the specific management and outcome of CNP is lacking. This study aimed to examine the outcomes of TBI patients involving single versus multiple *** A retrospective chart review of 170 consecutive TBI patients admitted to the tertiary neurosurgical center in Nepal between April 2020 and April 2022 was conducted. Demographic, clinical, and etiological characteristics; imaging findings; and management strategies were recorded, compared, and analyzed using descriptive statistics. The Glasgow Outcome Scale Extended (GOSE) was used to measure the outcomes in two groups of patients (single and multiple CNP) at 3 *** Out of 250 eligible patients, 80 were excluded and CNP was noted in 29 (17.1%) of the remaining 170. The median age was 34.9 years, and falls (60.6%) were the most common cause of trauma. TBI severity was categorized based on GCS: mild (82.4%), moderate (15.9%), and severe (1.8%). Cranial nerve involvement was seen in 29 (17.05%) patients: single cranial nerve involvement in 26 (89.65%) and multiple nerve involvement in 3 (10.34%). The most common isolated cranial nerve involved was the oculomotor nerve (37.9%). CT findings revealed a maximum of skull fractures with no significant association between CNP and CT *** CNP is a common consequence of TBI with the most common etiology being falls followed by RTA. Single CNP was more common than multiple CNP with no significant difference in the outcome in the 3-month GOSE score. Further research is needed to determine the burden of traumatic CNP and establish specific management guidelines for different types of CNP.
