Direct-acting antivirals sofosbuvir and daclatasvir attenuate carbon tetrachloride-induced liver fibrosis in mice

Background and aim:Advanced liver fibrosis is a major risk for developing hepatocellular carcinoma(HCC)in chronic hepatitis C virus(HCV)***-acting antivirals(DAAs)which are used for treating HCV infection,produce more than 90%cure rate but do not seem to diminish the rate of occurrence or recurrence of *** study aimed to investigate the effect of DAAs sofosbuvir(SOF)and daclatasvir(DAC)on carbon tetrachloride(CCl4)-induced fibrotic changes in ***:Eighty adult male Swiss albino mice were randomly allocated into 8 groups(10 mice/group):normal control group,SOF group(receiving SOF 80 mg/kg body weight(BW),oral gavage,daily),DAC group(receiving DAC 30 mg/kg BW,oral gavage,daily),SOF t DAC group(receiving a combination of both,daily),CCl4 model group(receiving CCl42 mL/kg BW,intraperitoneal twice weekly)and three CCl4-intoxicated groups receiving either SOF or DAC or their *** CCl4 groups received CCl4 for 12 weeks followed by DAAs for another 12 ***:CCl4-induced a significant elevation of alanine aminotransferase(ALT)and aspartate aminotransferase(AST),and produced histopathological evidence of fibrosis and liver degeneration along with a significant increase(P0.001)of the proliferation markers(proliferating cell nuclear antigen(PCNA)and Ki-67),hepatic stellate cells(HSCs)activation markers(alpha-smooth muscle actin(a-SMA)and glial fibrillary acidic protein(GFAP)),fibrosis marker(matrix metalloproteinase-9(MMP-9))and proinflammatory cytokine(tumor necrosis factor-alpha(TNF-a)).CCl4-intoxicated mice treated with SOF,DAC,or their combination revealed a significant amelioration(P0.001)of CCl4-induced elevation of liver enzymes,fibrotic changes,and liver degeneration along with a significant attenuation(P0.001)of CCl4-induced upregulation of all tested *** effects of SOF,DAC,and their combination on liver enzymes were comparable while the effect of SOF t DAC combination on mitigating CCl4-induced upregulation of the

Efficacy of Some Antibiotics in Curing Resistant <i>Escherichia coli</i>Infection

There is growing interest in re-evaluation of older antibiotics with the wide spread of pathogen resistance, especially gram negative bacteria, which impair treatment of some infections. In contrast various studies have reported that some antibiotics have efficacy in clearing resistant bacterial infections. On account of that it was interesting to evaluate the efficacy of erythromycin, chloramphenicol and/or tenoxicam in curing and/or relieving wound infection of highly resistant Escherichia coli and investigate the possible mechanisms beyond their antibacterial activity. This was achieved through evaluating highly resistant E. coli strains in vitro using agar dilution and in vivo rat models of E. coli infected wound and acute inflammation by carrageenin, where possible mechanisms were evaluated through measuring immunological mediators and histopathological examination. This study revealed that in vivo, erythromycin alone or in combination with tenoxicam significantly improved the healing of infected skin wounds with E. coli irresspective of resistancy in vitro. In addition to the improvement of immunological mediators involved in inflammatory reaction, oxidative stress and in cytokines expression as response to the bacterial infection in vivo. On the other hand chloramphenicol neither alone nor in combination with tenoxicam, achieved any significant effect. Tenoxicam didn’t show antimicrobial activity alone nor in combination with tested antibiotics in vitro, but it has shown synergestic activity in combination with tested antibiotics in vivo. Thus we concluded that immunomodulatory activity of erythromycin through anti-inflammatory and antioxidant effects was the possible mechanisms by which this antibiotic had healed infection with resistant E. coli in vivo, despite its resistancy to this antibiotic in vitro.

The use of gas diffusion electrode(GDE)based flow cell can realize industrial-scale CO_(2) reduction reactions(CO_(2)RRs).Controlling local CO_(2) and CO intermediate diffusion plays a key role in CO_(2)RR toward multi-carbon(C_(2+))*** this work,local CO_(2) and CO intermediate diffusion through the catalyst layer(CL)was investigated for improving CO_(2)RR toward C_(2+)*** gas permeability tests and finite element simulation results indicated CL can balance the CO_(2) gas diffusion and residence time of the CO intermediate,leading to a sufficient CO concentration with a suitable CO_(2)/H_(2)O supply for high C_(2+)*** a result,an excellent selectivity of C_(2+)products~79%at a high current density of 400 mA·cm^(-2) could be obtained on the optimal 500 nm Cu CL(Cu500).This work provides a new insight into the optimization of CO_(2)/H_(2)O supply and local CO concentration by controlling CL for C_(2+)products in CO_(2)RR flow cell.

Enhanced Heap-Based Optimizer Algorithm for Solving Team Formation Problem

Team Formation(TF)is considered one of the most significant problems in computer science and *** is defined as forming the best team of experts in a social network to complete a task with least *** real-world problems,such as task assignment,vehicle routing,nurse scheduling,resource allocation,and airline crew scheduling,are based on the TF *** has been shown to be a Nondeterministic Polynomial time(NP)problem,and high-dimensional problem with several local optima that can be solved using efficient approximation *** paper proposes two improved swarm-based algorithms for solving team formation *** first algorithm,entitled Hybrid Heap-Based Optimizer with Simulated Annealing Algorithm(HBOSA),uses a single crossover operator to improve the performance of a standard heap-based optimizer(HBO)*** also employs the simulated annealing(SA)approach to improve model convergence and avoid local minima *** second algorithm is the Chaotic Heap-based Optimizer Algorithm(CHBO).CHBO aids in the discovery of new solutions in the search space by directing particles to different regions of the search *** HBO’s optimization process,a logistic chaotic map is *** performance of the two proposed algorithms(HBOSA)and(CHBO)is evaluated using thirteen benchmark functions and tested in solving the TF problem with varying number of experts and ***,the proposed algorithms were compared to well-known optimization algorithms such as the Heap-Based Optimizer(HBO),Developed Simulated Annealing(DSA),Particle SwarmOptimization(PSO),GreyWolfOptimization(GWO),and Genetic Algorithm(GA).Finally,the proposed algorithms were applied to a real-world benchmark dataset known as the Internet Movie Database(IMDB).The simulation results revealed that the proposed algorithms outperformed the compared algorithms in terms of efficiency and performance,with fast convergence to the global minimum.

Unveiling the Optimal Interfacial Synergy of Plasma-Modulated Trimetallic Mn-Ni-Co Phosphides:Tailoring Deposition Ratio for Complementary Water Splitting

Designing highly active,durable,and nonprecious metal-based bifunctional electrocatalysts for overall water electrolysis is of urgent scientific importance to realize the sustainable hydrogen production,which remains a grand ***,an innovative approach is demonstrated to synthesize flower-like 3D homogenous trimetallic Mn,Ni,Co phosphide catalysts directly on nickel foam via electrodeposition followed by plasma *** electrochemical activity of the catalysts with varying Mn:Ni:Co ratios is assessed to identify the optimal composition,demonstrating that the equimolar trimetallic phosphide yields an outstanding HER catalytic performance with a current density of 10 mA cm^(-2) at an ultra-low overpotential of~14 mV,outperforming the best reported *** is asserted by the DFT calculations,revealing strong interaction of the metals and the P atom,resulting in enhanced water activation and optimized G_(H)^(*)values for the HER ***,this optimal composition appreciably catalyzes the OER by exposing more intrinsic active species in-situ formed on the catalyst surface during the ***,the Mn_(1)-Ni_(1)-Co_(1)-P-(O)/NF catalyst exhibits a decreased overpotential of~289 mV at 10 mA cm^(-2).More importantly,the electrocatalyst sustains perfect durability up to 48 h at a current density of 10 mA cm^(-2) and continued 5000 cycling stability for both HER and ***,the assembled MNC-P/NF||MNC-P/NF full water electrolyzer system attains an extremely low cell voltage of 1.48 V at 10 mA cm^(-2).Significantly,the robust stability of the overall system results in a remarkable current retention of~96%after a continuous 50-h ***,this study provides a facile design and a scalable construction of superb bifunctional ternary MNC-phosphide electrocatalysts for efficient electrochemical energy production systems.

Objective: To assess the role of cardiac troponin I(cTnI) in predicting outcome after percutaneous coronary intervention(PCI). Methods and results: cTnI was measured immediately before and at 6, 14, and 24 hours after PCI in 316 consecutive patients with stable symptoms and native coronary artery disease. The study end point was the occurrence of major adverse cardiac events(MACE) at 30 days and at 18 months after PCI: death, Q wave myocardial infarction(MI), or repeat revascularisation in hospital. Postprocedural cTnI increased in 31%of patients. The cumulative MACE rate at 18 months was 25%(17.7%due to repeat PCI procedures). There was a significant association between postprocedural cTnI increase and death, Q wave MI, or both(odds ratio(OR) 3.28, 95%confidence interval(CI) 1.7 to 6.4, p=0.01). Post-PCI cTnI increase had a positive predictive value(PPV) for adverse events at 18 months of 0.47 and a negative predictive value(NPV) of 0.96(OR 18.9, 95%CI 9.7 to 37, pelective PCI and was independently and significantly predictive of an increased risk of adverse events at 18 months, predominantly in the form of repeat PCI.
