Adaptation to a novel host and performance trade-off in host-generalist and host-specific insect ectoparasites

We investigated the performance trade-offs of fleas(Siphonaptera)while adapting to a novel host using two host generalists(Xenopsylla conformis and Xenopsylla ramesis)and one host specialist(Parapulex chephrenis)maintained on their principal hosts(Meriones crassus for Xenopsylla and Acomys cahirinus for P.chephrenis).We asked whether,over generations,(i)a host generalist may become a specialist by evolving the ability to exploit a novel host and losing the ability to exploit an original host and(ii)a host specialist can become a generalist by evolving the ability to exploit a novel host without losing the ability to exploit an original host.We established an experimental line of each species on a novel host(Acomys russatus for Xenopsylla and M.crassus for P.chephrenis)and maintained this line on this host during 23 generations.We compared reproductive performance of progenitors of each line and their descendants when they exploited either original or novel host in terms of egg number and size,hatching success,offspring production,and offspring size.We found changes in performance over generations in female offspring size only.Xenopsylla conformis demonstrated a tendency to become a host specialist(increased performance on the novel host with a concomitant decreased performance on the original host),whereas P.chephrenis demonstrated a tendency to become a host generalist(increased performance on the novel host without a concomitant decreased performance on the original host).We conclude that the probability of generalist to specialist transition,and vice versa,is context-dependent and varies between species.

Identification of Proteins and Genes Associated with Hedgehog Signaling Pathway Involved in Neoplasm Formation Using Text-Mining Approach

Analysis of molecular mechanisms that lead to the development of various types of tumors is essential for biology and medicine,because it may help to find new therapeutic opportunities for cancer treatment and cure including personalized treatment approaches.One of the pathways known to be important for the development of neoplastic diseases and pathological processes is the Hedgehog signaling pathway that normally controls human embryonic development.Systematic accumulation of various types of biological data,including interactions between proteins,regulation of genes transcription,proteomics,and metabolomics experiments results,allows the application of computational analysis of these big data for identification of key molecular mechanisms of certain diseases and pathologies and promising therapeutic targets.The aim of this study is to develop a computational approach for revealing associations between human proteins and genes interacting with the Hedgehog pathway components,as well as for identifying their roles in the development of various types of tumors.We automatically collect sets of abstract texts from the NCBI PubMed bibliographic database.For recognition of the Hedgehog pathway proteins and genes and neoplastic diseases we use a dictionary-based named entity recognition approach,while for all other proteins and genes machine learning method is used.For association extraction,we develop a set of semantic rules.We complete the results of the text analysis with the gene set enrichment analysis.The identified key pathways that may influence the Hedgehog pathway and their roles in tumor development are then verified using the information in the literature.

Early Neoproterozoic Granite-Gneisses of the Junggar Alataw(Southeastern Kazakhstan):Age,Petrogenesis and Tectonic Implications

The combined petrographic,petrological,geochemical and geochronological study of the Neoproterozoic gneisses of the Sarychabyn and Baskan complexes of the Junggar Alataw of South Kazakhstan elucidate the Precambrian tectonic evolution of the Aktau–Yili terrane.It is one of the largest Precambrian crustal blocks in the western Central Asian orogenic belt.The U-Pb single-grain zircon ages indicate that granite-gneisses formed from the same source and crystallised in the early Neoproterozoic ca.930–920 Ma.The chemical composition of gneisses corresponds to A2-type granites.The whole-rock Nd isotopic characteristics(εNd(t)=−4.9 to−1.0 and TNd(DM-2st)=1.9 to 1.7 Ga)indicate the involvement of Paleoproterozoic crustal rocks in magma generation.Early Neoproterozoic ca.930–920 Ma A-type granitoids in the Aktau–Yili terrane of South and Central Kazakhstan might reflect within-plate magmatism adjacent to the collisional belt or a local extension setting in back-arc areas of the continental arc.

BACKGROUND Lung cancer(LC)is the leading cause of morbidity and mortality among malignant neoplasms.Improving the diagnosis and treatment of LC remains an urgent task of modern oncology.Previously,we established that in gastric,breast and cervical cancer,tumor microvessels(MVs)differ in morphology and have different prognostic significance.The connection between different types of tumor MVs and the progression of LC is not well understood.AIM To evaluate the morphological features and clinical significance of tumor MVs in lung squamous cell carcinoma(LUSC).METHODS A single-center retrospective cohort study examined medical records and archival paraffin blocks of 62 and 180 patients with stage I-IIIA LUSC in the training and main cohorts,respectively.All patients underwent radical surgery(R0)at the Orenburg Regional Cancer Clinic from May/20/2009 to December/14/2021.Tumor sections were routinely processed,and routine Mayer's hematoxylin and eosin staining and immunohistochemical staining for cluster of differentiation 34(CD34),podoplanin,Snail and hypoxia-inducible factor-1 alpha were performed.The morphological features of different types of tumor MVs,tumor parenchyma and stroma were studied according to clinicopathological characteristics and LUSC prognosis.Statistical analysis was performed using Statistica 10.0 software.Univariate and multivariate logistic regression analyses were performed to identify potential risk factors for LUSC metastasis to regional lymph nodes(RLNs)and disease recurrence.Receiver operating characteristic curves were constructed to discriminate between patients with and without metastases in RLNs and those with and without disease recurrence.The effectiveness of the predictive models was assessed by the area under the curve.Survival was analyzed using the Kaplan-Meier method.The log-rank test was used to compare survival curves between patient subgroups.A value of P<0.05 was considered to indicate statistical significance.RESULTS Depending on the morphology,we classified tumor vessels i

Gastric cancer(GC) remains one of the most common lethal diseases. Every year, about 1 million people fall ill and almost 800 thousand people die worldwide~1. Surgical treatment has reached its limit. The current advances in immunology and detection of epidermal growth factor molecules(EGFR) in gastric cancer cells allowed to improve the treatment results;however, the metastatic form of this disease is still incurable.

Modeling of Leachate Propagation in a Municipal Solid Waste Landfill Foundation

The study deals with the numerical modeling of leachate distribution in the porous medium located under a municipal solid waste disposal landfill(MSWLF).The considered three-layer system is based on geological data obtained from field measurements.For simplicity,the problem is investigated by assuming a two-component approach.Nevertheless,the heat produced by landfills due to biological and chemical processes and the thermal diffusion mechanism contributing to pollution transport are taken into account.The numerical modeling of the propagation of leachate in the considered layered porous medium is implemented for parameters corresponding to natural soil and for the case where disruption of the porous medium structure is due to cracks formation or erosion.The latter is accompanied by an increase in rock permeability.In this case,the emergence of multi-vortex flow in the layer of high permeability is observed to substantially influence the bottom layers and cause intensification of the leachate propagation.These effects should be taken into account when estimating the parameters and properties of materials required for the construction of solid waste disposal facilities.

Predictors of disease recurrence after radical resection and adjuvant chemotherapy in patients with stage IIb-IIIa squamous cell lung cancer:A retrospective analysis

BACKGROUND Lung cancer(LC)is a global medical,social and economic problem and is one of the most common cancers and the leading cause of mortality from malignant neoplasms.LC is characterized by an aggressive course,and in the presence of disease recurrence risk factors,patients,even at an early stage,may be indicated for adjuvant therapy to improve survival.However,combined treatment does not always guarantee a favorable prognosis.In this regard,establishing predictors of LC recurrence is highly important both for determining the optimal treatment plan for the patients and for evaluating its effectiveness.AIM To establish predictors of disease recurrence after radical resection and adjuvant chemotherapy in patients with stage IIb-IIIa lung squamous cell carcinoma(LSCC).METHODS A retrospective case-control cohort study included 69 patients with LSCC who underwent radical surgery at the Orenburg Regional Clinical Oncology Center from 2009 to 2018.Postoperatively,all patients received adjuvant chemotherapy.Histological samples of the resected lung were stained with Mayer's hematoxylin and eosin and examined under a light microscope.Univariate and multivariate analyses were used to identify predictors associated with the risk of disease recurrence.Receiver operating characteristic curves were constructed to discriminate between patients with a high risk of disease recurrence and those with a low risk of disease recurrence.Survival was analyzed using the Kaplan-Meier method.The log-rank test was used to compare survival curves between patient subgroups.Differences were considered to be significant at P<0.05.RESULTS The following predictors of a high risk of disease recurrence in patients with stage IIb-IIa LSCC were established:a low degree of tumor differentiation[odds ratio(OR)=7.94,95%CI=1.08-135.81,P=0.049];metastases in regional lymph nodes(OR=5.67,95%CI=1.09-36.54,P=0.048);the presence of loose,fine-fiber connective tissue in the tumor stroma(OR=21.70,95%CI=4.27-110.38,P=0.0002);and fragmentation of the tumor s

Acute pericarditis is usually a self-limiting disorder that commonly occurs due to viral etiology or unexplained causes.Meningococcal pericarditis(MP)is a remarkably uncommon clinical entity that manifests as a complication during the acute or convalescent phase of meningococcal meningitis.However,its occurrence within the context of meningococcal septicemia has also been documented.We report a rare case of a previously healthy male patient who developed both acute pericarditis and uveitis as a presentation of meningococcal septicemia,with no characteristic skin rashes or meningeal signs.

According to the classification presented by Lehmann BD(2016),triple-negative breast cancer(TNBC)is a heterogeneous group of malignant tumors with four specific subtypes:basal-like(subtype 1 and subtype 2),mesenchymal,and luminal androgen receptor(LAR)subtypes.The basal-like subtypes of carcinomas predominate in this group,accounting for up to 80%of all cases.Despite the significantly lower proportions of mesenchymal and LAR variants in the group of breast carcinomas with a TNBC profile,such tumors are characterized by aggressive biological behavior.To this end,the LAR subtype is of particular interest,since the literature on such tumors presents different and even contradictory data concerning the disease course and prognosis.This review is devoted to the analysis of the relevant literature,reflecting the main results of studies on the molecular properties and clinical features of the disease course of LAR-type TNBC carcinomas.

Recent advances in nanoparticle generation in liquids by lasers:Revealing formation mechanisms and tailoring properties

When Fojtik and Henglein [1] submitted in 1992 the first scientific paper on generation of nanoparticles(NPs) by laser ablation in liquids, the authors probably could not realize how it will revolutionize the nanoparticle science and technology. In 2017, marking a quarter century of this pioneering study, Amendola and Barcikowski [2] pointed out that the simple and bright idea of Henglein and Fojtik was an inspiration to hundreds of scientists worldwide over 25 years and opened a way to use lasers in synthesis of colloids.
