Functionality of a bicistronic construction containing HEXA and HEXB genes encoding β-hexosaminidase A for cell-mediated therapy of GM2 gangliosidoses

Tay-sachs disease and sandhoff disease are severe hereditary neurodegenerative disorders caused by a deficiency ofβ-hexosaminidase A(HexA)enzyme,which results in the accumulation of GM2 gangliosides in the nervous system *** this work,we analyzed the efficacy and safety of cell-mediated gene therapy for sandhoff disease and sandhoff disease using a bicistronic lentiviral vector encoding cDNA of HexAα-andβ-subunit genes separated by the nucleotide sequence of a P2A peptide(HEXA-HEXB).The functionality of the bicistronic construct containing the HEXA-HEXB genetic cassette was analyzed in a culture of HEK293T cells and human umbilical cord blood mononuclear cells(hUCBMCs).Our results showed that the enzymatic activity of HexA in the conditioned medium harvested from genetically modified HEK293T-HEXA-HEXB and hUCBMCs-HEXA-HEXB was increased by 23 and 8 times,respectively,compared with the conditioned medium of native *** blot analysis showed that hUCBMCs-HEXA-HEXB secreted both completely separated HEXA and HEXB proteins,and an uncleaved protein containing HEXA+HEXB linked by the P2A *** injection of genetically modified hUCBMCs-HEXA-HEXB to laboratory Wistar rats was carried out,and the HexA enzymatic activity in the blood plasma of experimental animals,as well as the number of live cells of immune system organs(spleen,thymus,bone marrow,lymph nodes)were determined.A significant increase in the enzymatic activity of HexA in the blood plasma of laboratory rats on days 6 and 9(by 2.5 and 3 times,respectively)after the administration of hUCBMCsHEXA-HEXB was *** the same time,the number of live cells in the studied organs remained ***,the functionality of the bicistronic genetic construct encoding cDNA of the HEXA and HEXB genes separated by the nucleotide sequence of the P2A peptide was shown in vitro and in *** hypothesize that due to the natural ability of hUCBMCs to overcome biological barriers,such a strategy can rest

Influence of tributaries on downstream bed sediment grain sizes under flysch conditions

Tributaries are one of the most important factors contributing to variability in the downstream evolution of bed sediment grain *** primary aim of this work is to evaluate the response of the bed sediment texture in the recipient channel induced by ten tributaries of the?ernáOstravice stream and find reach-scale and catchment-scale parameters that would be able to predict this *** research was based on collecting information on the grain size distributions at sites adjacent to confluence zones.A significant change in sediment texture occurred in the vicinity of five *** the other factors contributing to grain size variability(e.g.,local channel geometry,lithology,and lateral sediment sources),it was assumed that only four of them were associated with a sufficient bedload influx to alter the sediment calibre below the ***,a significant morphological effect in the form of a large confluence bar was observed in one *** tributaries had several common features:(i)they had a larger relative catchment area than that of nonsignificant tributaries;(ii)they were characterized by different bed grain sizes,with some exceptions;and(iii)they had a higher unit stream power close to the confluence in relation to that of the *** characteristics were represented by the proposed relative parameters,including the relative unit stream power and bed material texture,which allowed the best classification of significant and nonsignificant *** their simplified form,the parameters described the transport capacity and grain size distribution,which were generally considered to be primary factors responsible for a redefinition of the sediment texture in the recipient ***,it should be noted that these results are subject to some degree of uncertainty due to the relatively small sample size of only 10 tributaries.

This study was an attempt made to explore the possibility of increasing the surface properties of the AZ91D magnesium alloy by applying ZrO_(2)coating using Physical Vapour Deposition(PVD-RF)sputtering *** order to improve the quality of the coating,the PVD process parameter with multiple performance characteristics was optimized by using the Taguchi grey approach.L 18 orthogonal array was selected for conducting the *** proposed Taguchi grey method was find out the optimal process parameter for multiple performance *** optimal combination was attained at chamber pressure of 0.003 bar,argon gas in millibar and power input of 200 *** validation experiment result shows an improvement in the micro hardness and surface ***,the performance characterizations such as sEM,EDX,XRD,coating thickness,surface roughness and micro hardness were measured at optimal process parameter.

Background： Metabolic syndrome （Mets） is one of the high cardiovascular （CV） situations. Endothelial dysfunction, which is a common finding in patients with Mets, is related with increased CV risk. In patients with Mets, the effect of the major CV risk factors, not included in the Mets definition, on endothelial dysfunction is not well known. The aim of this study was to determine the effect of major CV risk factors such as gender, smoking, family history, and biochemical parameters on endothelial dysfunction in patients with Mets. Methods： The study was performed between December 2010 and August 2014. A total of 55 patients （ 15 females and 40 males） with Mets and 81 healthy controls （37 females and 44 males） with a body mass index 〈25 kg/m2 were enrolled in the study. Endothelial dysfunction was measured by flow-mediated dilatation （FMD）, oxidative stress parameters; high-sensitivity C-reactive protein （hs-CRP）, oxidized low-density lipoprotein （ox-LDL）, endothelial nitric oxide synthase （e-NOs）, nitric oxide, and cell adhesion markers; yon Willebrand factor, and e-selectin. Platelet aggregation （endothelial adenosine diphosphate）, total platelet count, and mean platelet volume were additionally analyzed and demographic parameters were explored. student＇s t-test, Mann-Whitney U-test, and Chi-square test were used to analyze the results. Results： The fasting blood glucose （z = 3.52, P = 0.001 ）, hs-CRP （z - 3.23, P = 0.004）, ox-LDL （z = 2.62, P= 0,013）, and e-NOs （z = 2.22, P = 0.026） levels and cardiac risk score （z - 5.23, P 〈 0.001） were significantly higher in patients with Mets compared with the control group. smoking was correlated with decreased FMD （χ2 = 9.26, P = 0.002） in Mets patients but not in the control group. Conclusions： Increased ox-LDL, hs-CRP, and e-NOs are likely to be a result of oxidative stress, a condition in which an imbalance occurs between the production and inactivation of reactive nitrogen and oxygen species. Tailoring nanocomposite membranes of cellulose acetate/silica nanoparticles for desalination

Herein, the fabrication of cellulose acetate (CA) silica-based nanocomposite membranes via the dry-wetphase inversion procedure for water desalination was investigated. The modified and unmodified silicananoparticles (MsNPs and sNPs) were prepared by the sol-gel technique. The effect of the sNPs andMsNPs was investigated on the CA membrane's properties and their performance for water *** CA nanocomposite membranes were characterized to study their structure, hydrophilicity, andmorphology. The fabricated nanocomposite membranes showed hydrophilic surface properties. Theperformance of reverse osmosis (RO) membranes was measured using a crossflow RO unit at 10 bar(1 bar = 0.1 MPa). The membrane with 10 mg of sNPs enhanced permeate water flux compared to thepristine CA membrane by 1.6 L/(m2·h). The effect of MsNPs on the nanocomposites' performance waslower than their counterpart in the case of adding sNPs. The membrane with 30 mg of MsNPs showedthe highest permeate water flux among other nanocomposite membranes with a value oAQsf 35.7 L/(m2·h)at 24 bar.

Corrosion Measurements of Reinforcing steel by Different Electrochemical Techniques

Electrochemical techniques of the corrosion measurements of reinforcing steeI in concrete have been evaluated. These techniques include half-cell potential measurements, impressed voltage method, impressed current method and potentiostatic polarization technique. The results of corrosion behaviour of the steel in both 5%NaCl and 5%MgsO4 show that each electrochemical technique provides some information about the condition of the steel bar or the corrosivity of the environment being evaluated, yet none provides a complete data regarding the corrosion resistance of reinforcing steel in aggressive media

本文通过对高背压(50 bar,1 bar=1.0×105Pa)氩气经长锥型喷嘴(长度L=30 mm)向真空绝热膨胀所形成的超声气体团簇喷流的数值模拟,分析比较了由喷嘴喉口起沿喷流方向在喷流中心轴线上团簇平均尺寸的演化情况.结果表明:沿喷流方向团簇平均尺寸显示先增长后趋于饱和的变化趋势,具有较大尺寸团簇的区域出现在距离喷嘴喉口大约20 mm.据此本文再结合关于喷流中原子密度沿喷流方向变化的模拟结果开展了锥形喷嘴长度的优化研究.针对由常见构型的锥形喷嘴(喉径～0.5 mm,半张角～8.5?)在高背压下形成的团簇喷流,20 mm左右的长度为锥形喷嘴的适宜长度.

Additive friction stir deposition of AZ31B magnesium alloy

The current work explored additive friction stir deposition of AZ31B magnesium alloy with the aid of MELD?***31B magnesium bar stock was fed through a hollow friction stir tool rotating at constant velocity of 400 rpm and translating at linear velocity varied from 4.2 to 6.3 mm/s.A single wall consisting of five layers with each layer of 140×40×1 mm^(3)dimensions was deposited under each processing ***,phase,and crystallographic texture evolutions as a function of additive friction stir deposition parameters were studied with the aid of scanning electron microscopy including electron back scatter diffraction and X-ray *** feed material and additively produced samples consisted of theα-Mg *** additively produced samples exhibited a refined grain structure compared to the feed *** feed material appeared to have a weak basal texture,while the additively produced samples experienced a strengthening of this basal *** additively produced samples showed a marginally higher hardness compared to the feed *** current work provided a pathway for solid state additive manufacturing of Mg suitable for structural applications such as automotive components and consumable biomedical implants.

Background: Corneal astigmatism is a well-documented finding after cataract surgery. Postoperative astigmatism is of greater importance in children than in adults, because of its adverse effect on vision development and the risk of amblyopia. The purpose of this study was to evaluate refractive outcomes in eyes-which had high early postoperative astigmatism after congenital cataract surgery. Methods: We retrospectively reviewed the charts of 74 children (112 eyes), who underwent congenital cataract extraction with intraocular lens implantation, using limbal incision, scleral tunnel, or clear corneal incision. Thirty-two eyes of 28 children, aged 2 months to 11 years, had astigmatism of 3.0 diopters (D) or more when assessed 1 week after surgery. Changes in cylinder vector, spherical equivalent (sEQ) and defocus equivalent (DEQ) between 1 week and 5 months postoperatively were calculated for each group. Results: Mean sEQ and DEQ were 0.7± 3.5 D and 5.1± 2.8 D, respectively, at 1 week postoperatively-and -0.2± 3.4 D and 3.1± 2.3 D, respectively, at 5 months postoperatively in the three groups. DEQ levels showed a significant reduction during the 5-month follow-up period (P stigmatism vector was 2.9± 3.3 D× 93° in all the three groups 1 week postoperatively-and 0.2± 1.1 D× 153° at 5 months after surgery. A significant change during 5-month follow-up in cylinder vector (P sEQ (P=0.01) was found after operations using scleral tunnel technique. Conclusion: Children-who underwent congenital cataract surgery by different surgical techniques showed a significant spontaneous reduction in DEQ values. A significant change in cylinder vector and sEQ was found after operations using scleral tunnel technique.

Breakup of the Neoarchean supercontinent Kenorland:Evidence from zircon and baddeleyite U-Pb ages of LIP-related mafic dykes in the Coorg Block,southern India

The Coorg Block in southern Peninsular India is one of the oldest crustal blocks on Earth that preserves the evidence for continental crust formation during the Paleo-Mesoarchean through subductionrelated arc magmatism,followed by granulite facies metamorphism in the *** this study,we report for the first time,the'bar codes'of a major Paleoproterozoic Large Igneous Province in the Coorg Block through the finding of mafic dyke *** gabbroic dykes from the Coorg Block,dom-inantly composed of plagioclase-pyroxene assemblage,show a restricted range in siO_(2)values of 50.04-51.27 wt.%,and exhibit a sub-alkaline tholeiitic *** rocks show relatively flat LREE and constant HREE patterns and lack obvious Eu *** element modeling suggests that the dyke swarm was fed from a melt that originated at a shallow mantle level in the spinel stability *** grains are rare in the gabbro samples and those separated from two samples yielded 207Pb/206Pb weighted mean dates of 2214±12 Ma and 2221±7 *** grains show magmatic features with depleted LREE and enriched HREE and positive Ce and negative Eu *** grains were dated from five gabbro samples which yielded 207Pb/206Pb weighted mean ages ranging between 2217±7 Ma and 2228±10 *** combined data show a clear age peak at ca.2.2 *** mafic dykes in the Coorg Block show geochemical similarities with ca.2.2 Ga mafic dyke swarms in different regions of the Dharwar and other cratons in Peninsular India and elsewhere on the *** data also support the inference that the global mafic magmatism at ca.2.2 Ga was linked with intracontinental rifting of the Archean cratons through mantle upwelling or plume *** correlate the mafic dyke swarms in the Coorg Block with attempted rifting of the Neoarchean supercontinent Kenorland.
