Practice of Barrier Measures against COVId-19 Among diabetic People Followed at departmental University Hospital Center ouémé-Plateau, Benin, in 2021

Introduction: COVId-19 is a pandemic disease caused by the coronavirus SARS-COV2 appeared in China in 2019. The aim of the study was to evaluate the practice of barrier measures against COVId-19 and to identify associated factors among diabetics followed at departmental University Hospital Center ouémé-Plateau, Benin, in 2021. Methods: This was a cross-sectional study conducted from June to August 2021, including diabetics aged 18 and over, hospitalized or received for medical visits in the department of medicine at the target hospital. data were collected during an interview with each participant thanks to a questionnaire. “Good practice” was defined as regular practice of at least two of three barrier measures: hand washing, physical distancing and mask-wearing. Results: A total of 110 diabetics were included with a female predominance (55.5%) and a mean age of 57 ± 11.3 years. Among them, 89.1% washed their hands, 41.8% respected physical distancing and 12.7% wore a mask. The proportion of good practice was 47.3%. Only 5 practiced the three barrier measures. Only age group was associated with good practice. Conclusion: The study showed a low proportion of good practice of barrier measures against COVId-19 among diabetic people surveyed. It is important to continue the action for COVId-19 prevention among diabetic people in the targeted hospital and in other hospitals in Benin.

Epidemiology of Lower Extremity Artery disease in People Living with HIV Followed at the departmental University Hospital Center ouémé-Plateau in 2019

Introduction: Lower extremity artery disease (LEAd) is a serious cardiovascular disease. People living with the human immunodeficiency virus (HIV) are at risk. The aim of the study was to determine the prevalence of LEAd and identify the associated factors among people living with HIV who were followed at the departmental university hospital ouémé-Plateau in Benin. Methods: This was a cross-sectional study. It included all HIV-infected people who were monitored at the department of medicine of the target hospital during the study period and met the inclusion criteria (followed for at least three months, aged at least 25 years, and having given their written consent to participate). data were collected during an individual interview, followed by the measurement of parameters. The ankle brachial index ≤ 0.9 was used for the diagnosis of LEAd. Associated factors were searched through a multivariable logistic regression. Results: The prevalence of LEAd was 34.2% among 222 respondents having a mean age was 42.9 ± 10.8 years and a female predominance (77.5%). No significant association was found between the presence of LEAd and sociodemographic factors. The presence of LEAd was significantly associated with hypertension and antiretroviral therapy. Hypertensive patients had a higher risk of LEAd compared to non-hypertensive patients (OR = 1.98, 95% CI [1.04-3.83], p = 0.037). Those who were receiving second-line therapy also had a higher risk of LEAd compared to those on first-line therapy (OR = 2.95, 95% CI [1.14-7.60], p = 0.025). Conclusion: This study showed a high prevalence of LEAd especially among hypertensive patients and those who were receiving second-line antiretroviral therapy. LEAd diagnosis and management should be included in the routine care of people living with HIV in Benin.

Two medicinal plants, Gardenia erubescens and Fadogia agrestis were selected to evaluate their biological activities. Their total phenolic and flavonoid contents were assessed using folin-ciocalteu and aluminum chloride regents’ methods. The antioxidant activity was estimated using DPPH (1,1-diphényl-2-picrylhydrazyl), ABTS (2,2’-azinobis-[3-ethylenzothiazoline-6-sulfonic acid]) and FRAP (ferric reducing antioxidant power). The antiplasmodial activity of the extracts was determined in vivo on 42 NMRI mice. The results indicate that all the extracts from both two plants contain some polyphenols. The ethanolic extract of the leaves of Gardenia erubescens showed the best antioxidant activity by the method of DPPH. Amiodarone induces autophagy/mitophagy by activating sirtuin-3 in acute myelocytic leukemia

OBJECTIVE To find a new treatment strategy foracute myelocytic leukemia(AML), we tried to find new targets. Sirtuin-3(SIRT3), a nad+-dependent protein deacetylase localized primarily in the mitochondrion,is well-known to regulate autophagy, which is an evolutionarily conserved, multi-step lysosomal degradation process and also implicated in many diseases, such as *** evidence has revealed that SIRT3 may act as a tumor suppressor in AML, and thus activating SIRT3 will represent a potential therapeutic *** Aotophagy related protein expression and SIRT3 were analyzed by The Cancer Genome Atlas(TCGA)analysis. SIRT3 activator was selected by using in silico screening and anti-proliferative activities. The target selectivity of amiodarone was determined by co-crystal structure and deacetylase activity assays. The mechanisms of amiodarone-induced autophagy/mitophagy were investigated by fluorescence microscope, Western blotting, flow cytometry analysis, immunocytochemistry, as well as i TRAQ-based proteomics analysis. The therapeutic effect of amiodarone was assessed by xenograft AML mouse model. RESULTS TCGA dataset, we identified that SIRT3 was a key prognostic factor in AML, which was closely associated with the survival of patients. Subsequently, we computationally and experimentally screened a series of candidate activators of SIRT3 from drugbank,and found the best candidate compound amiodarone,which was further validated by co-crystal structure of SIRT3/amiodarone. Interestingly, amiodarone induced autophagy via SIRT3-mediated AMPK-m TOR-ULK1-Beclin-1 pathway in U937 and HL-60 cells. Moreover,this activator induced mitophagy via SIRT3-deacetylating FOXO3 a and thus triggering PINK1/Parkin pathway in AML cells. The i TRAQ-based proteomics analyses revealed that amiodarone induced mitophagy by downregulating 14-3-3γ, upregulating and deacetylating Beclin-1,as well as activating FOXO3 a-HIF1α-BNIP3 and *** is more, deacetylation proteomics

Obesity and associated metabolic diseases are characterized by a chronic low-grade inflammatory state with the infiltration of many inflammatory cells,especially *** molecules,including some cytokines,have a close relationship with ***(IL)-25 is a member of the IL-17 cytokine family that can regulate macrophages and alleviate some metabolic dysfunction;however,its role and mechanisms in lipid metabolism remain to be extensively *** serum and liver biopsy specimens,high-fat diet-induced obesity mice and dB/dB(Lepr−/−)animal models were used to examine IL-25 expression in obesity and nonalcoholic fatty liver diseases(NAFLd).To observe the role of IL-25 in lipid metabolism,model mice were administered with IL-25 or adoptively transferred with IL-25-educated macrophages in vivo,whereas bone marrow-derived macrophages,the macrophage cell line RAW264.7 and adipocytes differentiated from 3T3-L1 were used in ***-25 was decreased in NAFLd patients and obese *** addition,IL-25 reduced body weight gain and lipid accumulation,enhanced lipid uptake by macrophages and increased the expression of lipolysis andβ-oxidation enzymes via alternatively activating ***-25 also promoted lipolysis and suppressed lipogenesis in adipocytes co-cultured with the IL-25-educated ***,IL-25 improved the mitochondrial respiratory capacity and oxygen consumption rate of macrophages and produced more nad+/nadH and *** conclusion,IL-25 can stimulate M2 macrophage polarization and thereby promote lipolysis and mitochondrial respiratory capacity,highlighting the potential for IL-25 to be used as a therapeutic agent against obesity and associated metabolic syndromes.

通过对五种网络接入延迟(nad)算法dAP-nad、P-nad、R-nad、H-nad和RE-nad,建立仿真模型,分析各种nad的性能以及适用场合,针对以上几种nad的缺点,提出了一种新的基于缓存队列长度的自适应网络接入算法(AQL-nad)。通过仿真表明,AQL-nad与以上五种nad相比,具有较高的吞吐量,且有更低的端到端延时性能,节省了大量的时隙资源。

目的柚皮素对核因子红细胞2相关因子2(Nrf2)/Ⅱ相解毒酶活性的作用及其对血管炎症反应影响的研究。方法Western blot、免疫荧光、逆转录-qPCR检测蛋白的表达。Ⅱ相酶活性检测通过相关试剂盒进行。利用免疫共沉淀技术分析Nrf2与Kelch样环氧氯丙烷相关蛋白1(Keap-1)的相互作用。结果柚皮素促进RAW264.7细胞中Nrf2与Keap-1的解离和核内的聚集,并提高Ⅱ相解毒酶的表达,包括nad(P)H:醌氧化还原酶1(NQO-1)、谷胱苷肽S-转移酶(GST)和谷胺酸半胱氨酸连接酶(GCL)。同时,柚皮素抑制细胞炎症因子的产生;而Nrf2抑制剂ML385使NQO-1、GST和GCL表达降低、炎症因子的含量提高。另外,检测还发现柚皮素同样通过Nrf2的作用,提高动脉粥样硬化模型小鼠肝脏NQO-1、GST和GCL的表达以及酶的活性,降低主动脉血管的炎症反应。结论柚皮素激活Nrf2,促进相关的Ⅱ相解毒酶活性,从而抑制血管炎症。

采用分光光度法测定了酵母细胞中辅酶Ⅰ(nad+/nadH)的含量.结果表明:辅酶I浓度在一定范围内,吸光度随辅酶I浓度呈线性变化.建立了标准曲线和回归方程.方差检验证明该方法简便,快速,准确可靠,精密度高.
