Association of acute glycemic parameters at admission with cardiovascular mortality in the oldest old with acute myocardial infarction

OBJECTIVES Stress-related glycemic indicators,including admission blood glucose(ABG),stress-hyperglycemia ratio(SHR),and glycemic gap(GG),have been associated with worse outcomes after acute myocardial infarction(AMI).However,data regarding their prognostic value in the oldest old with AMI are ***,this study aimed to investigate the association of stress-related glycemic indicators with short-and long-term cardiovascular mortality(CVM)in the oldest old(≥80 years)with *** In this prospective study,a total of 933 consecutive old patients with AMI admitted to FuWai hospital(Beijing,China)were *** admission,ABG,SHR,and GG were assessed and all participants were classified according to their ***-Meier,restricted cubic splines(RCS),and multivariate Cox regression analyses were performed to evaluate the association between these glycemic indicators and CVM within 30 days and long-term *** During an average of 1954 patient-years of follow-up,a total of 250 cardiovascular deaths were ***-Meier analyses showed the lowest CVM in quartile 1 of ABG and in quartile 2 of SHR and *** adjusting for potential covariates,patients in quartile 4 of ABG,SHR,and GG had a respective 1.67-fold(95%CI:1.03-2.69;P=0.036),1.80-fold(95%CI:1.16-2.79;P=0.009),and 1.78-fold(95%CI:1.14-2.79;P=0.011)higher risk of long-term CVM risk compared to those in the reference groups(quartile 1 of ABG and quartile 2 of SHR and GG).Furthermore,RCS suggested a J-shaped relationship of ABG and a Ushaped association of SHR and GG with long-term ***,we observed similar associations of these acute glycemic parameters with 30-day *** Our data first indicated that SHR and GG consistently had a U-shaped association with both 30-day and longterm CVM among the oldest old with AMI,suggesting that they may be useful for risk stratification in this special population.

miR-125b reverses cisplatin resistance by regulating autophagy via targeting RORA/BNIP3L axis in lung adenocarcinoma

The platinum-based chemotherapy is one of the most frequently used treatment protocols for lung adenocarcinoma(LUAD),and chemoresistance,however,usually results in treatment failure and limits its application in the *** has been shown that microRnas(miRnas)play a significant role in tumor *** this study,miR-125b was identified as a specific cisplatin(DDP)-resistant gene in LUAD,as indicated by the bioinformatics analysis and the real-time quantitative PCR *** decreased serum level of miR-125b in LUAD patients was correlated with the poor treatment response rate and short survival ***-125b decreased the A549/DDP proliferation,and the multiple drug resistance-and autophagy-related protein expression levels,which were all reversed by the inhibition of *** addition,xenografts of human tumors in nude mice were suppressed by miR-125b,demonstrating that through autophagy regulation,miR-125b could reverse the DDP resistance in LUAD cells,both in vitro and in *** mechanistic studies indicated that miR-125b directly repressed the expression levels of RORA and its downstream BNIP3L,which in turn inhibited autophagy and reversed *** on these findings,miR-125b in combination with DDP might be an effective treatment option to overcome DDP resistance in LUAD.

NLRP3-mediated autophagy dysfunction links gut microbiota dysbiosis to tau pathology in chronic sleep deprivation

Emerging evidence indicates that sleep deprivation(SD)can lead to Alzheimer’s disease(AD)-related pathological changes and cognitive ***,the underlying mechanisms remain *** the present study,we identified the existence of a microbiota-gut-brain axis in cognitive deficits resulting from chronic SD and revealed a potential pathway by which gut microbiota affects cognitive functioning in chronic *** findings demonstrated that chronic SD in mice not only led to cognitive decline but also induced gut microbiota dysbiosis,elevated NLRP3 inflammasome expression,GSK-3βactivation,autophagy dysfunction,and tau hyperphosphorylation in the *** with the“SD microbiota”replicated the pathological and behavioral abnormalities observed in chronic sleep-deprived ***,both the deletion of NLRP3 in NLRP3-/-mice and specific knockdown of NLRP3 in the hippocampus restored autophagic flux,suppressed tau hyperphosphorylation,and ameliorated cognitive deficits induced by chronic SD,while GSK-3βactivity was not regulated by the NLRP3 inflammasome in chronic ***,deletion of NLRP3 reversed NLRP3 inflammasome activation,autophagy deficits,and tau hyperphosphorylation induced by GSK-3βactivation in primary hippocampal neurons,suggesting that GSK-3β,as a regulator of NLRP3-mediated autophagy dysfunction,plays a significant role in promoting tau ***,gut microbiota dysbiosis was identified as a contributor to chronic SD-induced tau pathology via NLRP3-mediated autophagy dysfunction,ultimately leading to cognitive ***,these findings highlight GSK-3βas a regulator of NLRP3-mediated autophagy dysfunction,playing a critical role in promoting tau hyperphosphorylation.

Gestational diabetes mellitus combined with fulminant type 1 diabetes mellitus, four cases of double diabetes: A case report

BACKGROUND Fulminant type 1 diabetes mellitus(FT1DM)that occurs during pregnancy or the perinatal period is known as pregnancy-related FT1DM(PF),always without history of abnormal glucose ***,we present four patients who developed FT1DM during treatment but were first diagnosed with gestational diabetes mellitus(GDM).CASE SUMMARY The clinical data of four patients with GDM combined with FT1DM admitted to our hospital between July 2018 and April 2021 were collected,and the patients and their infants were followed *** patients were diagnosed with GDM during the second trimester and were *** blood glucose level elevated suddenly during the third trimester and then were diagnosed with *** patients had an insulin allergy,and two had symptoms of upper respiratory tract infection before *** patient developed ketoacidosis,and three developed *** patients had cesarean section deliveries,and two had vaginal *** growth and development of the infants were normal.C-peptide levels were lower than those at onset,suggesting progressive impairment of islet *** frequencies of the DRB109:01,DQB103:03,DQA103:02,DPA101:03,DPA102:02,DPB105:01,DRB401:03,G 01:01,and G 01:04 human leukocyte antigen(HLA)-G alleles were high in the present *** In comparison with pregnancy-associated FT1DM(PF),patients with GDM combined with FT1DM had an older age of onset,higher body mass index,slower onset,fewer prodromal symptoms,and less *** pathogenesis may be due to various factors affecting the already fragileβ-cells of GDM patients with genetically susceptible class II HLA *** speculate that GDM combined with FT1DM during pregnancy,referred to as“double diabetes,”is a subtype of PF with its own unique characteristics that should be investigated further.

淮河生态经济带极端天气变化对粮食生产影响研究

随着全球气候变化速度加快,高温、暴雨、干旱、低温等极端天气频发增加了农业气象灾害的发生频率,给粮食产量稳定供给带来了巨大的挑战。基于淮河生态经济带28个地市2001~2020年日气温、降雨气象数据和农业生产相关数据,利用广义最小二... 详细信息

随着全球气候变化速度加快,高温、暴雨、干旱、低温等极端天气频发增加了农业气象灾害的发生频率,给粮食产量稳定供给带来了巨大的挑战。基于淮河生态经济带28个地市2001~2020年日气温、降雨气象数据和农业生产相关数据,利用广义最小二乘法(FGLS)研究淮河生态经济带极端天气变化及其对粮食生产的影响。结果表明:(1)从时间趋势上看,极高温日数(SU35)呈现增长趋势、极低温日数(FD5)呈现下降趋势,年际倾向率分别为1.85、-2.24 d/10a,暴雨日数(R50)以及最长连续无降水日数(CDD)年际变化趋势不明显;从空间区域分布上看,淮河生态经济带中西部地区极高温日数较多、北部地区极低温日数较多,暴雨日数呈现由东部向中西部再到北部地区减少的趋势、最长连续无降水日数呈现相反变动趋势。(2)极端天气对粮食生产具有显著不利影响。极低温日数、极高温日数、最长连续无降水日数、暴雨日数每增加1%,粮食单产分别下降0.069%、0.021%、0.018%、0.008%。(3)极端天气对淮河生态经济带粮食生产影响具有区域异质性。极端天气主要影响中西部地区和北部地区的粮食生产,对东部地区粮食生产影响不显著。提出通过培育抗旱抗涝等作物新品种、提高极端天气变化监测能力、加强区域内政府间防灾减灾等协同联动、继续加强高标准农田建设等措施,维护淮河生态经济带粮食产量稳定。
