Disease X Testing:The results of an international external quality assessment exercise

The United nations Secretary-General Mechanism(UNSGM)for investigation of the alleged use of chemical and biological weapons is the only established international mechanism of this type under the *** UNGSM may launch an international investigation,relying on a roster of expert consultants,qualified experts,and analytical laboratories nominated by the member *** the framework of the UNSGM,we organized an external quality assurance exercise for nominated laboratories,named the Disease X Test,to improve the ability to discover and identify new pathogens that may cause possible epidemics and to determine their animal ***“what-if”scenario was to identify the etiological agent responsible for an outbreak that has tested negative for many known pathogens,including viruses and *** microbes were added to the samples,Dabie bandavirus,Mammarenavirus,and Gemella spp.,of which the last two have not been taxonomically named or *** animal samples were from Rattus norvegicus,Marmota himalayana,New Zealand white rabbit,and the tick Haemaphysalis *** the 11 international laboratories that participated in this activity,six accurately identified pathogen X as a new Mammarenavirus,and five correctly identified the animal origin as *** results showed that many laboratories under the UNSGM have the capacity and ability to identify a new virus during a possible international investigation of a suspected biological *** technical details are discussed in this report.

The p38/MAPK pathway as a therapeutic target to prevent therapeutic escape of breast cancer stem cells

Cancer stem cells(CSCs) play an important role in metastasis development, tumor recurrence, and treatment resistance, and are essential for the eradication of cancer. Currently, therapies fail to eradicate CSCs due to their therapeutic stress-induced cellular escape, which leads to enhanced aggressive behaviors compared with CSCs that have never been treated. However, the underlying mechanisms regulating the therapeutic escape remain unknown. To this end, we established a model to isolate the therapeutic escaped CSCs(TSCSCs) from breast CSCs and performed the transcription profile to reveal the mechanism. Mechanistically, we demonstrated that the behavior of therapeutic escape was regulated through the p38/MAPK signaling pathway, resulting in TSCSCs exhibiting enhanced motility and metastasis. Notably,blocking the p38/MAPK signaling pathway effectively reduced motility and metastasis ability both in vitro and in vivo, which were further supported by downregulated motility-related genes and epithelial-mesenchymal transition(EMT)-related proteins vimentin and *** obtained findings reveal the p38/MAPK pathway as a potential therapeutic target for TSCSCs and would provide profound implications for cancer therapy.

Two New Inorganic-organic Hybrid Compounds Constructed from Different Polymolybdates and Transition Metal-amine Subunits

Two new polyoxometalate（POM）-based hybrid compounds, [Cu（en）][H4Mo4O16]0.5（1）（en=ethylenediamine） and [Ag（3-CsH6N2）2][H2PMo12O40]（2）（3-C5H6N2=3-aminopyridine）, containing different transition metal- amine subunits were hydrothermally synthesized and characterized by elemental analyses, infrared spectroscopy and single-crystal X-ray diffraction. For compound 1, each [H4Mo4O16]4-（M04O16） cluster was linked to four neighboring Mo4O16 clusters through four [Cu（en）]2＋ subunits to yield a （2,4）-connected 2D layer, which was further extended to a 3D supramolecular network via hydrogen bonding interactions. For compound 2, the adjacent [H2PMo12O40]- clusters were bridged by [Ag（3-C5H6N2）2]＋ subunits to generate a 1D chain. The electrochemical behaviors and the photoca- talytic activities of compounds 1 and 2 were studied in detail.

Radiofrequency ablation,heat shock protein 70 and potential anti-tumor immunity in hepatic and pancreatic cancers:a minireview

BACKGROUND: Radiofrequency ablation (RFA) is a minimally invasive surgical procedure which has widespread popularity in the treatment of hepatic and pancreatic cancers. Increased evidence indicates that RFA stimulates anti-tumor immunity, possibly through the induction heat shock protein 70 (HSP70) expression. HSP70 has the capacity to affect the immunogenicity of tumor cells, to chaperone antigenic peptides and deliver these into antigen presentation pathways within antigen-presenting cells, and to activate and regulate innate and adaptive immunity, which makes it useful in immunotherapeutic strategies for the treatment of cancers. DATA SOURCES: An English-language literature search was conducted using MEDLINE (1991-2010) on anti-tumor immunity, heat shock protein 70, radiofrequency ablation, hepatic cancer, pancreatic cancer, and other related subjects. RESULTS: RFA has an increasing application in the surgical treatment of hepatic and pancreatic cancers. Increased evidence indicates that RFA can induce the expression of HSP70 which possesses properties that enable it to influence a variety of immunological processes. Tumor-derived HSP70 is regarded as a potent adjuvant facilitating presentation of tumor antigens and induction of anti-tumor immunity. CONCLUSIONS: This review addresses the potential association of RFA, HSP70, and anti-tumor immunity in treatment of hepatic and pancreatic cancers. To establish direct evidence of a potential association of RFA, HSP70, and anti-tumor immunity in hepatic and pancreatic cancers, further investigations should be conducted.

Analgesic effects and structural elucidation of two new flavone C-glycosides from Artemisa sacrorum

The present study was designed to isolate and characterize the analgesic compounds of Artemisa sacrorum Ledeb. The Et OAc crude extracts from the aerial parts of Artemisa sacrorum Ledeb were separated by chromatography and the structures of new compounds were elucidated based on spectral analyses. Analgesic activities of the isolated compounds were assessed in rats with hot plate test and paw pressure assay. Two new flavone C-glycosides, named as Sacroroside A and B(Compounds 1 and 2) were isolated from the Et OAc crude extract of the aerial parts of Artemisa sacrorum Ledeb. They showed significant analgesic effects. In conclusion, Compounds 1 and 2 are new natural products, which show significant analgesic effects in a dose-dependent manner.
