The isolation of high-grade （i.e. high-pluripotency） human induced pluripotent stem cells （hiPSCs） is a decisive factor for enhancing the purity of hiPSC populations or differentiation efficiency. A non-invasive imaging system that can monitor microRna （miRna） expression provides a useful tool to identify and analyze specific cell populations. However, previous studies on the monitoring/isolation of hiPSCs by miRna expression have limited hiPSCs＇ differentiation system owing to long-term incubation with miRna imaging probe-nanocarriers. Therefore, we focused on monitoring high-grade hiPSCs without influencing the pluripotency of hiPSCs. We reduced nanoparticle transfection time, because hiPSCs are prone to spontaneous differentiation under external factors during incubation. The fluorescent nanoswitch （＂ON＂ with target miRna）, which can be applied for either imaging or sorting specific cells by fluorescence signals, contains an miRna imaging probe （miP） and a PEI-PEG nanoparticle （miP-P）. Consequently, this nanoswitch can sense various endogenous target miRnas within 30 min in vitro, and demonstrates strong potential for not only imaging but also sorting pluripotent hiPSCs without affecting pluripotency. Moreover, miP-P-treated hiPSCs differentiate well into endothelial cells, indicating that miP-P does not alter the pluripotency of hiPSCs. We envisage that this miRna imaging system could be valuable for identifying and sorting high-grade hiPSCs for improved practical applications.

Anoxic gas recirculation system for fouling control in anoxic membrane reactor

Anoxic gas recirculation system was applied to control the membrane fouling in pilot-scale 4- stage anoxic membrane bioreactor （MBR）. In the anaerobic-anoxic-anoxic-aerobic flow scheme, hydrophilic polytetrafluoroethylene （PTFE） membrane （0.2 μm, 7.2 m2/module） was submerged in the second anoxic zone. During 8 months operation, the average flux of the membrane was 21.3 L/（***）. Chemical cleaning of the membrane was conducted only once with sodium hydroxide and sodium hypochlorite. Dissolved oxygen （DO） concentration in the second anoxic zone was maintained with an average of 0.19 ± 0.05 mg/L. Gas chromatography analysis showed that the headspace gas in the second anoxic reactor was mainly consisted of N2 （93.0% ± 2.5%）, O2 （3.8% ± 0.6%）, and CO2 （3.0% ± 0.5%）, where the saturation DO concentration in liquid phase was 1.57 mg/L. Atmospheric 02 content （20.5% ± 0.8%） was significantly reduced in the anoxic gas. The average pH in the reactor was 7.2 ± 0.4. As a result, the recirculation of the anoxic gas was successfully applied to control the membrane fouling in the anoxic MBR.

Metal nanoparticles(NPs) supported on porous materials have shown great advantages in many catalytic application fields. Supported metal NPs are receiving extensive attention due to their significant contribution in a wide range of current and future applications, and this is arguably one of the fastest growing research fields. In this review, we highlight various types of metal catalysts that possess great potential in several catalytic reactions. The major focus has been on metal oxides, nanoporous metals and metal NPs supported on metal-organic frameworks(MOFs) and zeolites. Special attention has been given to the synthesis strategies and application of the NPs supported on MOFs and zeolites, which are considered highly interesting and rapidly expanding areas in heterogeneous catalysis. Finally, the prospects of these catalysts have been included in the concluding remarks.

Effect of hyperthermia on calbindin-D 28k immunoreactivity in the hippocampal formation following transient global cerebral ischemia in gerbils

Calbindin D-28K （CB）, a Ca2＋-binding protein, maintains Ca2＋ homeostasis and protects neurons against various insults. Hyperthermia can exacerbate brain damage produced by ischemic insults. However, little is reported about the role of CB in the brain under hyperthermic condition during ischemic insults. We inves- tigated the effects of transient global cerebral ischemia on CB immunoreactivity as well as neuronal damage in the hippocampal formation under hyperthermic condition using immunohistochemistry for neuronal nuclei （NeuN） and CB, and Fluoro-Jade B histofluorescence staining in gerbils. Hyperthermia （39.5 ＋ 0.2~C） was induced for 30 minutes before and during transient ischemia. Hyperthermic ischemia resulted in neu- ronal damage/death in the pyramidal layer of CA1-3 area and in the polymorphic layer of the dentate gyrus at 1, 2, 5 days after ischemia. In addition, hyperthermic ischemia significantly decreaced CB immunoreac- tivity in damaged or dying neurons at 1, 2, 5 days after ischemia. In brief, hyperthermic condition produced more extensive and severer neuronal damage/death, and reduced CB immunoreactivity in the hippocampus following transient global cerebral ischemia. Present findings indicate that the degree of reduced CB immu- noreactivity might be related with various neuronal damage/death overtime and corresponding areas after ischemic insults.

Potentiating activity of rhein in targeting of resistance genes in methicillin-resistant Staphylococcus aureus

Objective: To investigate the synergistic effect between rhein(RHE) and oxacillin against Staphylococcus aureus(MRSA) at the gene level. Method: A minimum inhibitory concentration and checkerboard dilution test were conducted to evaluate antibacterial activity. Reverse transcriptase polymerase chain reaction was conducted to investigate the gene expressions. Results: RHE exhibited a minimum inhibitory concentration of 62.5-250.0 μg/mL against various MRSA strains and the reference strain, respectively. As revealed by the checkerboard assay, a combination of RHE and oxacillin exhibited synergistic or partially synergistic effects against MRSA strains. RHE decreased the expressions of mecA/blaZ in a dose-dependent manner. RHE also decreased the expressions of the regulator genes mecI/blaI and mecR1/blaR1. Conclusions: We suggest that RHE affects the activity of mecR1/blaR1, which is located in the cell membrane of MRSA and results in the suppression of mecA/mecI/mecR1 and blaZ/blaI/blaR1 gene expressions.

Nusinersen for spinal muscular atrophy types Ⅱ and Ⅲ:a retrospective single-center study in South Korea

Background This study investigated the efficacy and safety of nusinersen,an antisense oligonucleotide,in patients with spinal muscular atrophy(SMA)types II(OMIM:253,550)or III(OMIM:253,400),including those with severe scoliosis or requiring respiratory support via mechanical *** Data from 40 patients with genetically confirmed SMA who were treated with nusinersen at our institute from March 2019 to April 2022 were retrospectively *** these,30 patients with an age of onsetnal Motor Scale-Expanded(HFMSE)*** The mean age of symptom onset was 1.2 *** patients were diagnosed with SMA type II(27/30,90%).Nusinersen was administered via computed tomography-guided or direct intrathecal injection in 87%(26/30)and 13%(4/30)of the patients,*** the 6-,14-,22-,and 26-month follow-ups,72%,71%,88%,and 86%of patients showed motor improvement,respectively,with mean changes in HFMSE scores of 2.10,2.88,4.21,and 5.29,*** analysis showed that the use of noninvasive ventilation was associated with poorer outcomes of motor *** Patients with SMA type II or III who received nusinersen treatment showed significant improvement in motor function.A longer treatment duration led to a higher number of patients with improved motor *** significant side effects of nusinersen were *** with SMA,even those with severe scoliosis or on respiratory support,can be safely treated using nusinersen.

Effects of bisphenol compounds on the growth and epithelial mesenchymal transition of MCF-7 CV human breast cancer cells

Bisphenol-A（BPA） has been considered as an endocrine disrupting chemical（EDC） because it can exert estrogenic *** bisphenol-S（BPS） and bisphenol-F（BPF） that are BPA analogs and substitutes,their risk to estrogendependent cancer has been reported rarely compared with the numerous cases of *** this study,we examined whether BPA,BPS,and BPF can lead to the proliferation,migration,and epithelial mesenchymal transition（EMT） of MCF-7 clonal variant（MCF-7 CV） breast cancer cells expressing estrogen receptors（ERs）.In a cell viability assay,BPA,BPS,and BPF significantly increased proliferation of MCF-7 CV cells compared to control（DMSO） as did17β-estradiol（E2）.In Western blotting assay,BPA,BPS,and BPF enhanced the protein expression of cell cycle progression genes such as cyclin D1 and *** addition,MCF-7 CV cells lost cell to cell contacts and acquired fibroblast-like morphology by the treatment of BPA,BPS,or BPF for 24 *** cell migration assay,BPA,BPS,and BPF accelerated the migration capability of MCF-7 CV cells as did *** relation with the EMT process,BPA,BPS,and BPF increased the protein expression of N-cadherin,while they decreased the protein expression of *** BPA,BPS,and BPF were co-treated with ICI 182,780,an ER antagonist,proliferation effects were reversed,the expression of cyclin D1 and cyclin E1 was downregulated,and the altered cell migration and expression of N-cadherin and E-cadherin by BPA,BPS,and BPF were restored to the control ***,these results imply that BPS and BPF also have the risk of breast cancer progression as much as BPA in the induction of proliferation and migration of MCF-7 CV cells by regulating the protein expression of cell cycle-related genes and EMT markers via the ER-dependent pathway.

Panax ginseng(***.)has been traditionally employed in Korea and China to alleviate fatigue and digestive *** particular,Korean red ginseng(KRG),derived from streamed and dried ***,is known for its anti-aging and anti-inflammatory ***,its effects on benign prostatic hyperplasia(BPH),a representative aging-related disease,and the underlying mechanisms remain *** study aims to elucidate the therapeutic effects of KRG on BPH,with a particular focus on mitochondrial dynamics,including fission and fusion *** effects of KRG on cell proliferation,apoptosis,and mitochondrial dynamics and morphology were evaluated in a rat model of testosterone propionate(TP)-induced BPH and TP-treated LNCaP cells,with mdivi-1 as a *** results revealed that KRG treatment reduced the levels of androgen receptors(AR)and prostate-specific antigens in the BPH *** inhibited cell proliferation by downregulating cyclin D and proliferating cell nuclear antigen(PCna)levels,and it promoted apoptosis by increasing the ratio of B-cell lymphoma protein 2(Bcl-2)-associated X protein(Bax)to Bcl-2 ***,KRG treatment enhanced the phosphorylation of dynamin-related protein 1(DRP-1,serine 637)compared with that in the BPH group,which inhibited mitochondrial fission and led to mitochondrial *** modulation of mitochondrial dynamics was associated with decreased cell proliferation and increased *** dysregulating AR signaling and inhibiting mitochondrial fission through enhanced DRP-1(ser637)phosphorylation,KRG effectively reduced cell proliferation and induced *** findings suggest that KRG’s regulation of mitochondrial dynamics offers a promising clinical approach for the treatment of BPH.

A BPR-CNN Based Hand Motion Classifier Using Electric Field Sensors

In this paper,we propose a BPR-CNN(Biometric Pattern Recognition-Convolution Neural Network)classifier for hand motion classification as well as a dynamic threshold algorithm for motion signal detection and extraction by EF(Electric Field)***,an EF sensor or EPS(Electric Potential Sensor)system is attracting attention as a next-generationmotion sensing technology due to low computation and price,high sensitivity and recognition speed compared to other sensor ***,it remains as a challenging problem to accurately detect and locate the authentic motion signal frame automatically in real-time when sensing body-motions such as hand motion,due to the variance of the electric-charge state by heterogeneous surroundings and operational *** hinders the further utilization of the EF sensing;thus,it is critical to design the robust and credible methodology for detecting and extracting signals derived from the motion movement in order to make use and apply the EF sensor technology to electric consumer products such as mobile *** this study,we propose a motion detection algorithm using a dynamic offset-threshold method to overcome uncertainty in the initial electrostatic charge state of the sensor affected by a user and the surrounding environment of the *** method is designed to detect hand motions and extract its genuine motion signal frame successfully with high *** setting motion frames,we normalize the signals and then apply them to our proposed BPR-CNN motion classifier to recognize their motion *** experiment and analysis show that our proposed dynamic threshold method combined with a BPR-CNN classifier can detect the hand motions and extract the actual frames effectively with 97.1%accuracy,99.25%detection rate,98.4%motion frame matching rate and 97.7%detection&extraction success rate.
