Application of a concrete thermal pile in cooling the warming permafrost under climate change

Permafrost degradation caused by climate warming is posing a serious threat to the stability of cast-in-place pile foundations in warm permafrost regions.Ambient cold energy can be effectively utilized by two-phase closed thermosyphons(TPCTs)to cool the permafrost.Therefore,we installed TPCTs in a cast-in-place pile foundation to create a unique structure called a thermal pile,which effectively utilizes the TPCTs to regulate ground temperature.And we conducted a case study and numerical simulation to exhibit the cooling performance,and optimize the structure of the thermal pile.The purpose of this study is to promote the application of thermal piles in unstable permafrost regions.Based on the findings,the thermal pile operated for approximately 53%of the entire year and effectively reduced the deep ground temperature at a rate of at least-0.1℃per year.Additionally,it successfully raised the permafrost table that is 0.35 m shallower than the natural ground level.These characteristics prove highly beneficial in mitigating the adverse effects of permafrost degradation and enhancing infrastructure safety.Expanding the length of the condenser section and the diameter of the TPCT in a suitable manner can effectively enhance the cooling capability of the thermal pile and ensure the long-term mechanical stability of the pile foundation even under climate warming.

RNA interference in insects:the link between antiviral defense and pest control

RNA interference(RNAi)is a form of gene silencing triggered by double-stranded RNA(dsRNA)that operates in all eukaryotic cells.RNAi has been widely investigated in insects to determine the underlying molecular mechanism,to investigate its role in systemic antiviral defense,and to develop strategies for pest control.When insect cells are infected by viruses,viral dsRNA signatures trigger a local RNAi response to block viral replication and generate virus-derived DNA that confers systemic immunity.RNAi-based insect pest control involves the application of exogenous dsRNA targeting genes essential for insect development or survival,but the efficacy of this approach has limited potency in many pests through a combination of rapid dsRNA degradation,inefficient dsRNA uptake/processing,and ineffective RNAi machinery.This could be addressed by dsRNA screening and evaluation,focusing on dsRNA design and off-target management,as well as dsRNA production and delivery.This review summarizes recent progress to determine the role of RNAi in antiviral defense and as a pest control strategy in insects,addressing gaps between our fundamental understanding of the RNAi mechanism and the exploitation of RNAi-based pest control strategies.

Machine learning-based predictions of current and future susceptibility to retrogressive thaw slumps across the Northern Hemisphere

Retrogressive thaw slumps(RTSs)caused by the thawing of ground ice on permafrost slopes have dramatically increased and become a common permafrost hazard across the Northern Hemisphere during previous decades.However,a gap remains in our comprehensive understanding of the spatial controlling factors,including the climate and terrain,that are conducive to these RTSs at a global scale.Using machine learning methodologies,we mapped the current and future RTSs susceptibility distributions by incorporating a range of environmental factors and RTSs inventories.We identified freezing-degree days and maximum summer rainfall as the primary environmental factors affecting RTSs susceptibility.The final ensemble susceptibility map suggests that regions with high to very high susceptibility could constitute(11.6±0.78)%of the Northern Hemisphere's permafrost region.When juxtaposed with the current(2000-2020)RTSs susceptibility map,the total area with high to very high susceptibility could witness an increase ranging from(31.7±0.65)%(SSP585)to(51.9±0.73)%(SSP126)by the 2041-2060.The insights gleaned from this study not only offer valuable implications for engineering applications across the Northern Hemisphere,but also provide a long-term insight into the potential change of RTSs in permafrost regions in response to climate change.

The Qinghai-Tibet Plateau(QTP)has experienced rapid environmental changes,including climate warming and wetting,since the 1980s.These environmental changes significantly impact the shallow soil hydrothermal conditions,which have key roles in land-atmosphere feedback and ecosystem functions.However,the spatial variations and responses of soil hydrothermal conditions to environmental changes over the QTP with permafrost(PF)and seasonal frost(SF)remain unclear.In this study,we investigated the spatial variations in soil temperature(ST)and soil moisture(SM)changes over the QTP from 2000 to 2020 using 99 in-situ sites with observations at 4 depths(i.e.10,40,100 and 200 cm).The main environmental controlling factors were further identified using a calibrated statistical model.Results showed that significant(pfuture environmental changes over the QTP.

Tissue adhesive,ROS scavenging and injectable PRP-based‘plasticine’for promoting cartilage repair

Platelet-rich plasma(PRP)that has various growth factors has been used clinically in cartilage repair.However,the short residence time and release time at the injury site limit its therapeutic effect.The present study fabricated a granular hydrogel that was assembled from gelatin microspheres and tannic acid through their abundant hydrogen bonding.Gelatin microspheres with the gelatin concentration of 10wt%and the diameter distribution of 1-10 lm were used to assemble by tannic acid to form the granular hydrogel,which exhibited elasticity under low shear strain,but flowability under higher shear strain.The viscosity decreased with the increase in shear rate.Meanwhile,the granular hydrogel exhibited self-healing feature during rheology test.Thus,granular hydrogel carrying PRP not only exhibited well-performed injectability but also performed like a‘plasticine’that possessed good plasticity.The granular hydrogel showed tissue adhesion ability and reactive oxygen species scavenging ability.Granular hydrogel carrying PRP transplanted to full-thickness articular cartilage defects could integrate well with native cartilage,resulting in newly formed cartilage articular fully filled in defects and well-integrated with the native cartilage and subchondral bone.The unique features of the present granular hydrogel,including injectability,plasticity,porous structure,tissue adhesion and reactive oxygen species scavenging provided an ideal PRP carrier toward cartilage tissue engineering.

A HEVC Video Steganalysis Method Using the Optimality of Motion Vector Prediction

Among steganalysis techniques,detection against MV(motion vector)domain-based video steganography in the HEVC(High Efficiency Video Coding)standard remains a challenging issue.For the purpose of improving the detection performance,this paper proposes a steganalysis method that can perfectly detectMV-based steganography in HEVC.Firstly,we define the local optimality of MVP(Motion Vector Prediction)based on the technology of AMVP(Advanced Motion Vector Prediction).Secondly,we analyze that in HEVC video,message embedding either usingMVP index orMVD(Motion Vector Difference)may destroy the above optimality of MVP.And then,we define the optimal rate of MVP as a steganalysis feature.Finally,we conduct steganalysis detection experiments on two general datasets for three popular steganographymethods and compare the performance with four state-ofthe-art steganalysis methods.The experimental results demonstrate the effectiveness of the proposed feature set.furthermore,our method stands out for its practical applicability,requiring no model training and exhibiting low computational complexity,making it a viable solution for real-world scenarios.

Insect-specific RNA virus affects the stylet penetration activity of brown citrus aphid(Aphis citricidus)to facilitate its transmission

Sap-sucking insects often transmit plant viruses but also carry insect viruses,which infect insects but not plants.The impact of such insect viruses on insect host biology and ecology is largely unknown.Here,we identified a novel insect-specific virus carried by brown citrus aphid(Aphis citricidus),which we tentatively named Aphis citricidus picornavirus(AcPV).Phylogenetic analysis discovered a monophyletic cluster with AcPV and other unassigned viruses,suggesting that these viruses represent a new family in order Picornavirales.Systemic infection with AcPV triggered aphid antiviral immunity mediated by RNA interference,resulting in asymptomatic tolerance.Importantly,we found that AcPV was transmitted horizontally by secretion of the salivary gland into the feeding sites of plants.AcPV influenced aphid stylet behavior during feeding and increased the time required for intercellular penetration,thus promoting its transmission among aphids with plants as an intermediate site.The gene expression results suggested that this mechanism was linked with transcription of salivary protein genes and plant defense hormone signaling.Together,our results show that the horizontal transmission of AcPV in brown citrus aphids evolved in a manner similar to that of the circulative transmission of plant viruses by insect vectors,thus providing a new ecological perspective on the activity of insect-specific viruses found in aphids and improving the understanding of insect virus ecology.

A nanoagent for concurrent therapy of breast cancer bone metastasis and cancer-induced bone pain through SLC7A11 interruption and photodynamic therapy

Bone metastasis,a life-threatening complication of advanced breast cancer,is often accompanied by debilitating pain(cancer-induced bone pain,CIBP)that severely impairs life quality and survival.The concurrent treatment of bone metastases and CIBP remains a clinical challenge because the therapeutic options are limited.In this study,we construct a near-infrared light-activated nano-therapeutic system to meet this conundrum.In detail,sorafenib(SRF)and photosensitizer(chlorin e6,Ce6)are encapsulated into mesoporous hydroxyapatite nanoparticles(HANPs),which are further functionalized with hyaluronic acid(HA)to obtain HA-SRF/Ce6@HANPs system.The designed nanoplatform destroys tumor cells in vitro and in vivo via the synergism of SRF(interrupting the exchange of cystine/glutamate by inhibiting SLC7A11)and photodynamic therapy(PDT,inducing reactive oxygen species generation).The decrease in tumor burden and reduction of extracellular glutamate significantly attenuate CIBP in mice model with developing bone cancer.Moreover,the combination of HA-SRF/Ce6@HANPs and PDT inhibit osteoclasts activation,promote osteoblast differentiation and accelerate bone repair.Overall,the nanoagent with good biocompatibility may provide an effective therapy method for the concurrent treatment of breast cancer bone metastasis and CIBP.

Fabrication of Molecularly Imprinted Electrospun Nanofibers with Mono-amidoxime functional Ligand for Efficient Decontamination of Toxic Organophosphates

There is an urgent imperative to discover practical and efficacious artificial catalysts for the expeditious decontamination of toxic organophosphates. Herein, we propose a novel molecularly imprinted approach utilizing electrospun fiber scaffolds. Specifically, an amidoxime-based functional polymer (PMAOX) has been synthesized, which contains amidoxime groups that can act as nucleophiles and ligands for the formation of catalytic active sites. This polymer was blended with polyacrylonitrile (PAN), a well-processable material, to prepare a nanofiber mat using electrospinning techniques. Then, the amidoxime side groups on fiber surface were further cross-linked after the molecular coordination of templates to complete the molecularly imprinting process. This approach can not only enhance the content of functional molecularly imprinted polymers without affecting structural stability, but also combines surface MI technology to fully expose and leverage the advantages of active sites. The as prepared molecularly imprinted electrospun nanofibers MIF-Zn-PMAOX/PAN-6/4 catalyzes the degradation of paraoxon with a half-life of 32 min, and MIF-Ag-PMAOX/PAN catalyzes the degradation of parathion with a half-life of 18 min. The maximum catalytic rate evidence rate enhancements nearly 3700-fold of the self-hydrolysis. Thus, the mono-amidoxime based molecularly imprinted fibers demonstrate the versatility and superiority as self-detoxifying for organophosphates.

A review of physicochemical properties of dissolved organic carbon and its impact over mountain glaciers

Investigating the characteristics and transformation of water-soluble carbonaceous matter in the cryosphere regions is important for understanding biogeochemical process in the earth system.Water-soluble carbonaceous matter is a heterogeneous mixture of organic compounds that is soluble in aquatic environments.Despite its importance,we still lack systematic understanding for dissolved organic carbon(DOC)in several aspects including exact chemical composition and physical interactions with microorganisms,glacier meltwater.This review presents the chemical composition and physical properties of glacier DOC deposited through anthropogenic emission,terrestrial,and biogenic sources.We present the molecular composition of DOC and its effect over snow albedo and associated radiative forcings.Results indicate that DOC in snow/ice is made up of aromatic protein-like species,fulvic acid-like materials,and humic acid-like materials.Light-absorbing impurities in surface snow and glacier ice cause considerable albedo reduction and the associated radiative forcing is definitely positive.Water-soluble carbonaceous matter dominated the carbon transport in the high-altitude glacial area.Owing to prevailing global warming and projected increase in carbon emission,the glacial DOC is expected to release,which will have strong underlying impacts on cryosphere ecosystem.The results of this work have profound implications for better understanding the carbon cycle in high altitude cryosphere regions.A new compilation of globally distributed work is required,including large-scale measurements of glacial DOC over high-altitude cryosphere regions,to overcome and address the scientific challenges to constrain climate impacts of light-absorbing impurities related processes in Earth system and climate models.
