Commercial Vehicle Cab Fatigue Life Study Based on the FEM and Tests

The study of coach welded structures durability based on the tests and FEM Numerical simulation was carried out.First,the tension-compression fatigue tests with welded joints were carried out.The steel S-N curves which will be used in the FEA fatigue analysis were gained;The finite element models of weld structure has been built and the fatigue analysis of welded joints were taken out,the comparison between fatigue test results and the FEA simulation results was performed,the results of both methods showed very closed and the verification of FEA weld seam model which was built in the FEA;Finally,the FEA model of coach with welded joints has been established,the number of seam welds reached at 3870,the fatigue analysis of the whole coach has been carried out.The research can be used for coach or bus fatigue analysis of high cycle in welded joints of profiles with thin wall tubes.

Innovations and stepwise evolution of CBFs/DREB1s and their regulatory networks in angiosperms

In this paper,commercial vehicle fatigue lift including static torsion fatigue and random fatigue was carried out using the finite element method and test method.The static cab torsion fatigue analysis based on the finite element model was carried with the MSC.Nastran software.Comparing the finite element fatigue calculation results and testing fatigue results,they are very closed and the finite element fatigue analysis can be verified by the Test We established the process of cab random fatigue lift analysis.The spot-welds built in the Hypermesh software must be converted in the FEMFAT software before the inertia relief static compulation.The static and multi-body dynamic simulation were carried with MSC.Nastran and MSC.Adams software.We took the cab random fatigue simulation with the static analysis result and the B grade,C grade,D grade road load history which was obtained from multi-body computation using the testing ground data as input The combination of finite element simulation and test can be used for predicting the cab durability and analyzing the fatigue damage of the cab.The study indicate that spot weld fatigue damage play an important role of the cab fatigue which need to be estimated accurately.

The C-repeat binding factors/dehydrationresponsive element binding protein 1 s(CBFs/DREB1 s)have been identified as major regulators of cold acclimation in many angiosperm plants.However,their origin and evolutionary process associated to cold responsiveness are still lacking.By integrating multi-omics data of genomes,transcriptomes,and CBFs/DREB1 s genome-wide binding profiles,we unveil the origin and evolution of CBFs/DREB1 s and their regulatory network.Gene collinearity and phylogeny analyses show that CBF/DREB1 is an innovation evolved from tandem duplication-derived DREBⅢgene.A subsequent event of e-whole genome duplication led to two CBF/DREB1 archetypes(CladesⅠandⅡ)in ancient angiosperms.In contrast to cold-insensitivity of Clade I and their parent DREBⅢgenes,CladeⅡevolved a further innovation in cold-sensitive response and was stepwise expanded in eudicots and monocots by independent duplications.In geological time,the duplication events were mainly enriched around the Cretaceous-Paleogene(K-Pg)boundary and/or in the Late Cenozoic Ice Age,when the global average temperature significantly decreased.Consequently,the duplicated CBF/DREB1 genes contributed to the rewiring of CBFs/DREB1 s-regulatory network for cold tolerance.Altogether,our results highlight an origin and convergent evolution of CBFs/DREB1 s and their regulatory network probably for angiosperms adaptation to global cooling.

C-repeat binding factors(CBFs)are well-known transcription factors(TFs)that regulate plant cold acclimation.RNA sequencing(RNA-seq)data from diverse plant species provide opportunities to identify other TFs involved in the cold response.However,this task is challenging because gene gain and loss has led to an intertwined community of co-orthologs and in-paralogs between and within species.Using orthogroup(closely related homologs)analysis,we identified 10,549 orthogroups in five representative eudicots.A phylotranscriptomic analysis of cold-treated seedlings from eudicots identified 35 high-confidence conserved cold-responsive transcription factor orthogroups(CoCoFos).These 35 CoCoFos included the well-known cold-responsive regulators CBFs,HSFC1,ZAT6/10,and CZF1 among others.We used Arabidopsis BBX29 for experimental validation.Expression and genetic analyses showed that cold-induction of BBX29 is CBF-and abscisic acid-independent,and BBX29 is a negative regulator of cold tolerance.Integrative RNA-seq and Cleavage Under Targets and Tagmentation followed by sequencing analyses revealed that BBX29 represses a set of cold-induced TFs(ZAT12,PRR9,RVE1,MYB96,etc.).Altogether,our analysis yielded a library of eudicot CoCoFos and demonstrated that BBX29 is a negative regulator of cold tolerance in Arabidopsis.

Convergent evolution of AP2/ERF Ⅲ and IX subfamilies through recurrent polyploidization and tandem duplication during eudicot adaptation to paleoenvironmental changes

Whole-genome duplication(WGD or polyploidization)has been suggested as a genetic contributor to angiosperm adaptation to environmental changes.However,many eudicot lineages did not undergo recent WGD(R-WGD)around and/or after the Cretaceous-Paleogene(K-Pg)boundary,times of severe environmental changes;how those plants survived has been largely ignored.Here,we collected 22 plants from major branches of the eudicot phylogeny and classified them into two groups according to the occurrence or absence of R-WGD:12 R-WGD-containing plants(R-WGD-Y)and 10 R-WGD-lacking plants(R-WGD-N).Subsequently,we identified 496 gene-rich families in R-WGD-Y and revealed that members of the AP2/ERF transcription factor family were convergently over-retained after R-WGDs and showed exceptional cold stimulation.The evolutionary trajectories of the AP2/ERF family were then compared between R-WGD-Y and R-WGD-N to reveal convergent expansions of the AP2/ERF Ⅲ and IX subfamilies through recurrent independent WGDs and tandem duplications(TDs)after the radiation of the plants.The expansions showed coincident enrichments in-times around and/or after the K-Pg boundary,when global cooling was a major environmental stressor.Consequently,convergent expansions and co-retentions of AP2/ERF Ⅲ C-repeat binding factor(CBF)duplicates and their regulons in different eudicot lineages contributed to the rewiring of cold-specific regulatory networks.Moreover,promoter analysis of cold-responsive AP2/ERF genes revealed an underlying cis-regulatory code(G-box:CACGTG).We propose a seesaw model of WGDs and TDs in the convergent expansion of AP2/ERF Ⅲ and IX genes that has contributed to eudicot adaptation during paleoenvironmental changes,and we suggest that TD may be a reciprocal/alternative mechanism for genetic innovation in plants that lack WGD.

针对客车推力杆附近车身骨架断裂问题,结合有限元分析的方法,运用Hypermesh、Nastran有限元分析计算软件,对客车的车身骨架建立有限元模型;结合设计的要求及分析的经验,分3种工况分析,研究车身骨架断裂问题的原因,并基于有限元分析法对结构进行优化研究。研究结果表明,客车紧急制动时,车身骨架受到纵向载荷和垂向载荷综合作用,车身骨架的断裂处正好位于高应力区域;在断裂处延展加强板,增加矩形管支撑,断裂处的应力降幅50%;采用优化后的结构,在路试中未发生推力杆附近车身骨架断裂问题。

目的探讨食管癌患者静脉血栓栓塞症(VTE)的发生情况。方法采用回顾性队列研究方法。收集2017年1月至2021年12月四川省肿瘤医院收治的8458例食管癌患者的临床病理资料;男6923例,女1535例;年龄为(64±9)岁;行手术治疗3187例,非手术治疗5271例。观察指标:(1)食管癌患者VTE的发生情况。(2)VTE患者治疗和转归情况。正态分布的计量资料以x±s表示,组间比较采用t检验。偏态分布的计量资料以M(范围)表示,组间比较采用非参数秩和检验。计数资料以绝对数或百分比表示,组间比较采用χ2检验或Fisher确切概率法。等级资料比较采用非参数秩和检验。结果(1)食管癌患者VTE的发生情况。8458例食管癌患者中,175例发生VTE,发生率为2.069%(175/8458);其中深静脉血栓形成164例,肺栓塞4例,深静脉血栓形成合并肺栓塞7例;手术患者59例,非手术患者116例。手术与非手术食管癌合并VTE患者血栓类型比较,差异无统计学意义(χ2=1.95,P>0.05)。3187例手术患者VTE发生率为1.851%(59/3187),其中肺栓塞发生率为0.157%(5/3187),占手术患者VTE的比例为8.475%(5/59)。5271例非手术治疗患者VTE发生率为2.201%(116/5271),其中肺栓塞发生率为0.114%(6/5271),占非手术患者VTE的比例为5.172%(6/116)。手术与非手术患者VTE及肺栓塞发生率比较,差异均无统计学意义(χ2=1.20、0.05,P>0.05)。(2)VTE患者治疗和转归情况。175例食管癌合并VTE患者中,163例行药物治疗,12例未治疗。163例行药物治疗患者中,158例行抗凝治疗,5例行溶栓治疗,均好转出院。结论食管癌患者VTE的发生率较低,为2.069%;手术与非手术患者VTE的发生情况、合并VTE的血栓类型比较,差异均无统计学意义。
