Halomonas bluephagenesis TD serves as an exceptional chassis for next generation industrial biotechnology to produce various ***,the simultaneous editing of multiple loci in *** TD remains a significant ***,we report the development of a multiple loci genome editing system,named CRISPR-deaminase-assisted base editor(CRISPR-BE)in *** *** system comprises two components:a cytidine(CRISPR-cBE)and an adenosine(CRISPR-aBE)deaminase-based base ***-cBE can introduce a cytidine to thymidine mutation with an efficiency of up to 100%within a 7-nt editing window in *** ***,CRISPR-aBE demonstrates an efficiency of up to 100%in converting adenosine to guanosine mutation within a 7-nt editing ***-cBE has been further validated and successfully employed for simultaneous multiplexed editing in *** *** findings reveal that CRISPR-cBE efficiently inactivated all six copies of the IS1086 gene simultaneously by introducing stop *** system achieved an editing efficiency of 100%and 41.67%in inactivating two genes and three genes,*** substituting the Pcas promoter with the inducible promoter PMmp1,we optimized CRISPR-cBE system and ultimately achieved 100%editing efficiency in inactivating three *** conclusion,our research offers a robust and efficient method for concurrently modifying multiple loci in *** TD,opening up vast possibilities for industrial applications in the future.

Seed viability is an essential feature for genetic resource conservation as well as sustainable crop ***-term storage induces seed viability deterioration or seed aging,accompanied by the accumulation of toxic reactive oxygen species(ROS)to suppress seed *** deterioration treatment(CDT)is a gen-eral approach for mimicking seed *** transcription factor ANAC089 was previously reported to modulate seed primary *** this study,we evaluated the ability of ANAC089 to control seed viability during *** with that in the wild-type line,the mutation of ANAC089 signiﬁcantly increased H_(2)O_(2),thereby reducing seed viability after CDT,while the overexpression of ANAC089 reduced H_(2)O_(2) and improved seed long-evity,indicating a critical role for ANAC089 in maintaining seed viability through H_(2)O_(2) signaling.A series of stu-dies have shown that ANAC089 targets and negatively regulates the level of ABI5,an important transmitter of abscisic acid(ABA)signals,to affect seed viability after ***,ABI5 negatively regulated the expres-sion of VTC2,which is involved in the biosynthesis of the antioxidant ascorbic acid and H_(2)O_(2) *** a result,ANAC089 attenuates the generation of H_(2)O_(2),thereby enhancing seed viability through the ABI5-VTC2 module during the seed aging *** together,our results reveal a novel mechanism by which ANAC089 enhances seed viability by coordinating ABI5 and VTC2 expression,ultimately preventing the overac-cumulation of H_(2)O_(2),which would have led to reduced seed viability.

Superluminous supernovae(SLSNe)are a class of intense celestial events that can be standardized for measuring cosmological parameters,bridging the gap between type Ia supernovae and the cosmic microwave *** this work,we discuss the cosmological applications of SLSNe from the Chinese Space Station Telescope(CSST).Our estimation suggests that SLSNe rate is biased tracing the cosmic star formation rate,exhibiting a factor of(1+z)^(1.2).We futher predict that CSST is poised to observe~360 SLSNe in the 10 square degrees ultra-deep field survey within a span of 2.5 years.A stringent constraint on cosmological parameters can be derived from their peak-color *** is anticipated to uncover a substantial number of SLSNe,contributing to a deeper understanding of their central engines and shedding light on the nature of dark energy at high redshifts.

The Zhongjiang gas field is a typical large gas field in terrigenous strata of the Western Sichuan *** remains debatable which member of the Upper Triassic Xujiahe Formation served as the source rocks and how significant the member contributed to the gas accumulations in the Zhongjiang gas *** this study,we analyzed the essential characteristics of the Lower Jurassic source rocks and the geochemical features of light hydrocarbons in natural gas from the 2nd(T_(3)χ^(2))and 4th members(T_(3)χ^(4))of the Upper Triassic Xujiahe Formation(T_(3)χ),as well as the Middle Jurassic Shaximiao(J_(2)s)and Qianfoya(J_(2)q)*** on this,we explored the sources of the natural gas in the Zhongjiang gas field and determined the natural gas migration patterns and their effects on the properties of light hydrocarbons in the natural *** results indicate that the Lower Jurassic lacustrine source rocks of the Zhongjiang gas field contain humic organic matter,with vitrinite reflectance(R_(0))values ranging from 0.86%to 0.98%.Samples meeting the criterion for effective source rocks[total organic carbon(TOC)content≥0.75%]exhibited an average TOC content of merely 1.02%,suggesting significantly lower hydrocarbon generation potential than source rocks in the underlying T3x,which show higher thermal maturity and TOC *** natural gas samples from T_(3)χ^(2),T_(3)χ^(4),J_(2)s,and J_(2)q reservoirs,their C_(5-7)iso-alkane content was significantly higher than their n-alkane content,and their methylcyclohexane(MCH)index ranged from 59.0%to 77.3%,indicating the predominance of methylcyclohexane in C_(7)light *** indicated by the origin identification and gas-source correlation based on the geochemical features of light hydrocarbons,the natural gas in the Zhongjiang gas field is typical coal-derived *** gas from the primary pay zone of the Shaximiao Formation,with significantly high K_(1),(P_(2)+N_(2))/C_(7),and P_(3)/C_(7)values,predominantly ori

Thienoacenes is one of most important groups of semiconducting materials due to the high stability and superior ***,there are scarce studies on the emission properties of thienoacenes to ***,we synthesized fluorinated and chlorinated dibenzo[d,d’]thieno[3,2-b;4,5-b’]dithiophenes(DBTDTs)derivatives F6-DBTDT and Cl6-DBTDT by sulfoxide cyclization,significantly lowering the energy levels relative to the parent compound *** to single crystal structure analysis,F6-DBTDT molecules adopt one-dimensional slipped stacking with closeπ-πinteractions of 3.43Å(1Å=0.1 nm),which is different from the parent compound DBTDT with herringbone stacking ***,the halogenated DBTDT derivatives exhibit enhanced emission properties both in solution and in the solid state,opening up possiblities to improve photoluminescence of thienoacences by halogenation.

The paper focuses on the ergodicity of a φ-irreducible Markov chain {X_n, n ≥ 0} that is generated iteratively through the expression Xn+1= f（X_n） + ?n+1. Here, {?_n, n ≥ 1} is a sequence of independent identically distributed centralized random variables, f（·） is an R-valued continuous function, and X0is arbitrary but independent of {?_n, n ≥ 1}. Our main contribution is to provide necessary and sufficient conditions for ergodicity of this special class of Markov chains. We also present a generalized approach for f（·） in the end.

AIM:To elucidate whether differences exist in the impact on retarding the elongation of axial length(AL)among children with myopia when utilizing orthokeratology(ortho-k)lenses employing the corneal refractive therapy(CRT)design versus those employing the vision shaping treatment(VST)***:This retrospective clinical trial aimed to collect and analyze AL data from individuals who wore ortho-k lenses for three years.A total of 654 subjects were enrolled and prescribed one of the three specific brands of ortho-k lenses:CRT,Euclid,and *** study’s primary focus was to compare the rates of AL elongation and myopic progression across these three brands of ortho-k ***:In the 3-year follow-up,the AL elongation exhibited variations of 0.73±0.36 mm in the CRT lens group,0.59±0.37 mm in the Euclid lens group,and 0.63±0.38 mm in the Mouldway lens group.A noteworthy disparity emerged between the CRT and Mouldway groups(Pnificant distinction between the CRT and Euclid groups(Pnificant difference when compared to the Mouldway group(P<0.05).CONCLUSION:Ortho-k lenses represent a pragmatic strategy for mitigating the advancement of *** contradistinction to ortho-k lenses utilizing the CRT design,those employing the VST design exhibited a more favorable impact regarding retarding AL elongation.

A gas sensor can convert external gas concentration or species into electric voltage or current signals by physical adsorption or chemical changes. As a result, a gas sensor in a nonlinear circuit can be used as a sensitive sensor for detecting external gas signals from the olfactory system. In this paper, a gas sensor and a field-effect transistor are incorporated into a simple Fithzhugh–Nagumo neural circuit for capturing and encoding external gas signals. An improved functional neural circuit is obtained, and the effect of gas concentration, gas species and neuronal activity can be discerned as the gate voltage, threshold voltage and activation coefficient of the field-effect transistor, respectively. The gas concentration can affect the neural activities from quiescent to normal working and, finally, to saturation state in bursting, spiking, periodic and chaotic firings with different frequencies. The effects of gas species and neuronal activity on the firing state can also be achieved in this functional neural circuit. In addition, variations in the gate voltage, threshold voltage and activation coefficient can cause switching between different firing modes. These results can be helpful in designing artificial olfactory devices for bionic gas recognition and other coupled systems arising in applied sciences.
