Background:Persons with mental disorders are at a higher risk than the general population for the subsequent development of certain medical conditions.Methods:We used a population-based cohort from Danish national registries that included data on more than 5.9 million persons born in Denmark from 1900 through 2015 and followed them from 2000 through 2016,for a total of 83.9 million person-years.

Amyotrophic lateral sclerosis(ALs)is associated with proteostasis collapse:ALs is an unrelenting neurodegenerative disease that is characterized by the loss of motor neurons in the brain and spinal cord,resulting in the progressive atrophy,and eventual paralysis,of skeletal muscles.Death due to respiratory failure usually occurs within 2-5 years from symptom onset.Approximately 90%of ALs cases are of unknown etiology and are termed sporadic ALs(sALs).The remaining 10%of ALs cases present with a family history(familial ALs;fALs)and are associated with genetic mutations in a range of over 20 functionally heterogeneous genes.Regardless of disease origin,the pathological hallmark of ALs is the accumulation of ubiquitylated protein inclusions in motor neurons and surrounding glial cells.The presence of these inclusions,compromised largely of misfolded and aggregated proteins,implies a collapse in proteostasis.Proteostasis refers to the maintenance of the proteome in a state of balance or equilibrium so that a cell can perform its proper function.

Background Greater physical activity is associated with improved outcomes in people living with and beyond cancer.However,most studies in exercise oncology use self-reported measures of physical activity.Few have explored agreement between self-reported and device-based measures of physical activity in people living with and beyond cancer.This study aimed to describe physical activity in adults affected by cancer across self-reported and device-assessed activity,to explore levels of agreement between these measures in terms of their utility for categorizing participants as meeting/not meeting physical activity guidelines,and to explore whether meeting guidelines is associated with fatigue,quality of life,and sleep quality.Methods A total of 1348 adults living with and beyond cancer from the Advancing survivorship Cancer Outcomes Trial completed a survey assessing fatigue,quality of life,sleep quality,and physical activity.The Godin-shephard Leisure-Time Physical Activity Questionnaire was used to calculate a Leisure score Index(LsI)and an estimate of moderate-to-vigorous physical activity(MVPA).Average daily steps and weekly aerobic steps were derived from pedometers worn by participants.Results The percentage of individuals meeting physical activity guidelines was 44.3%using LsI,49.5%using MVPA,10.8%using average daily steps,and 28.5%using weekly aerobic steps.Agreement(Cohen'sκ)between self-reported and pedometer measures ranged from 0.13(LsI vs.average daily steps)to 0.60(LsI vs.MVPA).After adjusting for sociodemographic and health-related covariates,meeting activity guidelines using all measures was associated with not experiencing severe fatigue(odds ratios(ORs):1.43–1.97).Meeting guidelines using MVPA was associated with no quality-of-life issues(OR=1.53).Meeting guidelines using both self-reported measures were associated with good sleep quality(ORs:1.33–1.40).Conclusion Less than half of all adults affected by cancer are meeting physical activity guidelines,regardless of measure.Meeting guidelines is asso

Ultraviolet light exposure and its penetrance through the eye in a porcine model

AIM:To determine the amount of ultraviolet(UV)light irradiance that various layers of the eye receive as sunlight passes through the eye,and to investigate the protective benefits of UV light-blocking contact lenses.METHODs:Twenty-four porcine eyes were prepared in one of three ways:isolated cornea,cornea and lens together,or whole eye preparation.UV light irradiance was measured with a UV-A/B light meter before and after the eye preparations were placed over the meter to measure UV light penetration in each eye structure.In the whole eye preparation,a hole was placed in the fovea to measure light as it passed through the vitreous.subsequently,UVprotective contact lenses were placed over the structures,and UV light penetrance was measured.Measurements of UV light exposure were taken outdoors at various locations and times.REsULTs:Cornea absorbed 63.56%of UV light that reached the eye.Cornea and lens absorbed 99.34%of UV light.Whole eye absorbed 99.77%of UV light.When UV-protective contact lenses were placed,absorption was 98.90%,99.55%,and 99.87%,respectively.UV light exposure was dependent on directionality and time of day,and was greatest in areas of high albedo that reflect significant amounts of light,such as a beach.CONCLUsION:Cornea absorbs the majority of UV light that reaches the eye in this model.UV-protective contact lenses reduce UV exposure to the eye.Locations with high albedo expose the eye to higher levels of UV light.

The letter is to respond to the recent publication“Trends in hospitalization for alcoholic hepatitis from 2011 to 2017:A UsA nationwide study”(World j Gastroenterol 2022;28:5036-5046).We noticed a significant difference in the total numbers of reported hospitalized alcohol-associated hepatitis(AH)patients between this publication and our publication on Alcohol Clin Exp Res(2022;46:1472-1481).We believe the number of“AH-related hospitalizations”inflated by the inclusion of patients with non-AH forms of alcohol-associated liver disease.

An experiment was conducted to explore the effects of digestible amino acid(dAA)concentrations and supplemental protease on live performance and energy partitioning in broilers.Ross 308 male broilers(n?288)were distributed into 24 floor pens and offered 1 of 4 dietary treatments with 6 replicates from 1 to 35 d of age.Dietary treatments consisted of a 2×2 factorial arrangement with dAA concentrations(standard and reduced[34 g/kg below standard])and supplemental protease(without or with)as the main factors.At 1,15,28,and 35 d of age,feed and broilerswereweighed to determine live performance.From20 to 23 d of age,a total of 32 birds(2 birds/chamber,4 replicates)were placed in closed-calorimeter chambers to determine respiratory exchange(heat production,HP),apparent metabolisable energy(AME),retained energy(RE),and net energy(NE).From 29 to 35 d of age,supplemental protease in the reduced-dAA diet decreased broiler feed conversion ratio(FCR)by 5.6 points,whereas protease supplementation in the standard-dAA diet increased FCR by 5.8 points.The indirect calorimetry assay revealed that supplemental protease decreased(Pj/kg.Also,from 20 to 23 d of age,broilers offered the reduced-dAA diet with supplemental protease had a higher daily body weight gain(BWG)(t10.4%),N intake(t7.1%),and N retention(t8.2%)than those offered the standard-dAA with supplemental protease.Broilers offered the reduced-dAA without supplemental protease exhibited a 3.6%higher AME-to-crude protein(CP)ratio than those offered other treatments.Protease supplementation in the standard-and reduced-dAA diets resulted in 2.7%and 5.6%lower AME intake-to-N retention ratios,respectively,compared with the unsupplemented controls.Reduced-dAA increased(Pse supplementation increased(Pspiratory quotient(t1.2%)and N retention-to-N intake ratio(t2.2%),NE-to-AME ratio(t1.9%),and reduced HP(3.6%),hea

Bandicoots and bilbies(Order Peramelemorphia)occupy a broad range of habitats across Australia and New Guinea,from open,arid deserts to dense forests.This once diverse group has been particularly vulnerable to habitat loss and introduced eutherian predators,and numerous species extinctions and range retractions have occurred.Understanding reasons for this loss requires greater understanding of their biology.Morphology of the pinnae and tympanic bullae varies markedly among species.As hearing is important for both predator avoidance and prey location,the variability in ear morphology could reflect specialization and adaptation to specific environments,and therefore be of conservation relevance.We measured 798 museum specimens representing 29 species of Peramelemorphia.Controlling for phylogenetic relatedness and head length,pinna surface area was weakly negatively correlated with average precipitation(rainfall being our surrogate measure of vegetation productivity/complexity),and there were no environmental correlates with effective diameter(pinna width).Controlling for phylogenetic relatedness and skull length,tympanic bulla volume was negatively correlated with precipitation.species that inhabited drier habitats,which would be open and allow sound to carry further with less obstruction,had relatively larger pinnae and tympanic bullae.In contrast,species from higher rainfall habitats,where sounds would be attenuated and diffused by dense vegetation,had the smallest pinnae and bullae,suggesting that low-frequency hearing is not as important in these habitats.Associations with temperature did not reach statistical significance.These findings highlight linkages between hearing traits and habitat that can inform conservation and management strategies for threatened species.

Background and aim:Non-alcoholic fatty liver disease(NAFLD)is becoming a leading cause of chronic liver disease worldwide.The molecular events that influence disease progression from non-alcoholic fatty liver(NAFL)to aggressive non-alcoholic steatohepatitis(NAsH)remain incompletely understood,leading to lack of mechanism-based targeted treatment options for NAsH.This study aims to identify early signatures associated with disease progression from NAFL to NAsH in mice and humans.Materials and methods:Male C57BL/6j mice were fed a high-fat,-cholesterol,and-fructose(HFCF)diet for up to 9 months.The extent of steatosis,inflammation,and fibrosis was evaluated in liver tissues.Total RNA sequencing(RNA-seq)was conducted to determine liver transcriptomic changes.Results:After being fed the HFCF diet,mice sequentially developed steatosis,early steatohepatitis,steatohepatitis with fibrosis,and eventually spontaneous liver tumor.Hepatic RNA-seq revealed that the key signatures during steatosis progression to early steatohepatitis were pathways related to extracel-lular matrix organization and immune responses such as T cell migration,arginine biosynthesis,C-type lectin receptor signaling,and cytokine-cytokine receptor interaction.Genes regulated by transcription factors forkhead box M1(FOXM1)and negative elongation factor complex member E(NELFE)were significantly altered during disease progression.This phenomenon was also observed in patients with NAsH.

Introduction The efficacy and safety of local intra-arterial(IA)thrombolytics during endovascular thrombectomy(EVT)for large-vessel occlusions is uncertain.We analysed how often IA thrombolytics were administered in the Multicenter Randomized Clinical Trial of Endovascular Treatment for Acute Ischemic stroke in the Netherlands(MR CLEAN)Registry,whether it was associated with improved functional outcome and assessed technical and safety outcomes compared with EVT without IA thrombolytics.Methods In this observational study,we included patients undergoing EVT for an acute ischaemic stroke in the anterior circulation from the MR CLEAN Registry(March 2014-November 2017).The primary endpoint was favourable functional outcome,defined as an modified Rankin scale score≤2 at 90 days.secondary endpoints were reperfusion status,early neurological recovery and symptomatic intracranial haemorrhage(sICH).subgroup analyses for IA thrombolytics as primary versus adjuvant revascularisation attempt were performed.Results Of the 2263 included patients,95(4.2%)received IA thrombolytics during EVT.The IA thrombolytics administered were urokinase(median dose,250000 IU(IQR,193750-250000))or alteplase(median dose,20 mg(IQR,12-20)).No association was found between IA thrombolytics and favourable functional outcome(adjusted OR(aOR),1.16;95%CI 0.71 to 1.90).successful reperfusion was less often observed in those patients treated with IA thrombolytics(aOR,0.57;95%CI 0.36 to 0.90).The odds of sICH(aOR,0.82;95%CI 0.32 to 2.10)and early neurological recovery were comparable between patients treated with and without IA thrombolytics.For primary and adjuvant revascularisation attempts,IA thrombolytics were more often administered for proximal than for distal occlusions.Functional outcomes were comparable for patients receiving IA thrombolytics as a primary versus adjuvant revascularisation attempt.Conclusion Local IA thrombolytics were rarely used in the MR CLEAN Registry.In the relatively small study sample,no statistical difference was observe
