Impact of poly chlorinated biphenyl (Aroclor 1254) and vitamin C on antioxidant system of rat ventral prostate

Aim: To evaluate the effect of polychlorinated biphenyls (PCB) and vitamin C on ventral prostatic antioxidant system in adult male rats. Methods: A group of 20 adult male rats were administered ip Aroclor 1254 in corn oil at a dose of 2 mg·kg-1·day-1 for 30 days. Ten control rats were administered only the vehicle. After 30 days the treated rats were divided at random into 2 sub-groups of 10 animals each. One sub-group received vitamin C at a dose of 500 mg·kg-1·day-1 for 10 days. The other group was maintained as Aroclor 1254 control. Twenty-four hours after the last treatment the rats were killed by decapitation. Ventral prostatic homogenate was prepared and used for the estimation of enzymatic antioxidants, including superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST) and hydrogen peroxide (H2O2), lipid peroxidation (LPO) and prostatic acid phosphatase. The serum levels of total T3, total T4, TSH, testosterone and estradiol were also assayed. Results: The body weight and ventral prostatic weight were reduced in PCB treated rats. The activities of SOD, CAT, GST and acid phosphatase were decreased while the levels of H2O2 and lipid peroxidation were increased in the ventral prostate of PCB treated rats. Administration of vitamin C restored these parameters. Serum levels of thyroid hormones, estradiol and testosterone were decreased in PCB treated animals. Administration of vitamin C restored the thyroid hormone levels. Conclusion: PCB induces oxidative stress and decreases the antioxidant enzymes in the ventral prostate of adult male rats; the effects could be reversed by the administration of vitamin C.

Genetic epidemiology of irritable bowel syndrome

Irritable bowel syndrome(IBS) is the most common functional gastrointestinal disorder characterized by presence of abdominal pain or discomfort associated with altered bowel habits. It has three main subtypes- constipation predominant IBS(C-IBS),diarrhea predominant IBS(D-IBS) and IBS with mixed featuresof both diarrhea as well as constipation(M-IBS). Its pathophysiology and underlying mechanisms remain elusive. It is traditionally believed that IBS is a result of multiple factors including hypersensitivity of the bowel,altered bowel motility,inflammation and stress. Initial studies have shown familial aggregation of IBS suggesting shared genetic or environmental factors. Twin studies of IBS from different parts of world have shown higher concordance rates among monozygotic twins than dizygotic twins,and thus suggesting a genetic component to this disorder. Multiple studies have tried to link single-nucleotide polymorphisms(SnPs) to IBS but there is little evidence that these SnPs are functional. Various molecules have been studied and investigated by the researchers. Serotonin,a known neurotransmitter and a local hormone in the enteric nervous system,has been most extensively explored. At this time,the underlying gene pathways,genes and functional variants linked with IBS remain unknown and the promise of genetically-determined risk prediction and personalize medicine remain unfulfilled. However,molecular biological technologies continue to evolve rapidly and genetic investigations offer much promise in the intervention,treatment and prevention of IBS.

Glass and Mineral Chemistry of northern Central Indian Ridge Basalts:Compositional Diversity and Petrogenetic Significance

The glass and mineral chemistry of basaits examined from the northern central Indian ridge （nCIR） provides an insight into magma genesis around the vicinity of two transform faults： Vityaz （VT） and Vema （VM）. The studied mid-ocean ridge basalts （MORBs） from the outer ridge flank （VT area） and a near-ridge seamount （VM area） reveal that they are moderately phyric plagioclase basalts composed of plagioclase （phenocryst [An60-90] and groundmass [An35-79]）, olivine （Fo81-88）, diopside （Wo45-51, En25-37, Fs14-24）, and titanomagnetite （FeOt -63.75 wt% and TiO2 -22.69 wt%）. The wholerock composition of these basalts has similar Mg# [mole Mg/mole（Mg＋Fe2＋）] （VT basalt： -0.56-0.58; VM basalt： -0.57）, but differ in their total alkali content （VT basalt： -2.65; VM basalt： -3.24）. The bulk composition of the magma was gradually depleted in MgO and enriched in FeOt, TiO2, P2O5, and na2O with progressive fractionation, the basalts were gradually enriched in Y and Zr and depleted in ni and Cr. In addition, the ∑REE of magma also increased with fractionation, without any change in the （La/ Yb）n value. Glass from the VM seamount shows more fractionated characters （Mg#： 0.56-0.57） compared to the outer ridge flank lava of the VT area （Mg#： 0.63-0.65）. This study concludes that present basalts experienced low-pressure crystallization at a relatively shallow depth. The geochemical changes in the nCIR magmas resulted from fractional crystallization at a shallow depth. As a consequence, spinel was the first mineral to crystallize at a pressure 〉10 kbar, followed by Fe-rich olivine at 〈10 kbar pressure.

A Petrogenetic Model of Basalts from the northern Central Indian Ridge:3-11°S

Mid-Ocean Ridge Basalts （MORB） from the northern Central Indian Ridge （nCIR） were recovered between latitudes 3° and 11° S and are olivine tholeiite with higher abundances of K and Rb. They are of typical transitional MORB （T-MORB） variety and appear to have been generated from an enriched-mantle peridotite source. The primitive nCIR MORBs having Mg^# 〉 0.68 are the product of partial melting at an estimated pressure of - 1 GPa. It is inferred that the magma was subsequently modified at a pressure 〉 1 GPa by crystal fractionation and spinel was the first mineral to crystallize followed by separation of relatively Fe-rich olivine with subsequent decrease in pressure. During progressive fractionation at lower pressure （between 1-0.5 GPa）, the bulk composition of the magma became systematically depleted in MgO, and enriched in ∑FeO, TiO2, P2Os and na20. There was, however, limited gradual depletion in Al2O3 and CaO and concomitant enrichment in K20. With the progressive fractionation these basalts became gradually enriched in V, Co, Y, Zr and to some extent in Sr, and depleted in ni and Cro In addition, the T_JtEE of the magma also increased with fractionation, without any change in （La/Yb）n value.

An efficient synthesis of 3,4-dihydropyrimidine 2 （1H）-ones and thiones （3,4-DHPMs） core was prepared by one-pot threecomponent Biginelli condensation and which was catalyzed by trifluoromethane sulfonic acid. The classical BigneUi reaction has been extended by the use of n-substituted benzoxazolyl semicarbazides and thiosemicarbazides and this method has the advantage of excellent yields and short reaction times. ？2009 M. PM<sub>10</sub>Emissions from Cooking Fuels in nigerian Households and Their Impact on Women and Children

A study was carried out on the type of fuels being used by households in Ibadan, a highly populated city in south-west nigeria. Ekotedo, Agbeni, Bodija and Agbowo communities were selected through stratified random sampling, keeping in view of their socioeconomic background and population density. The study monitored 186 households for the type of fuel used for cooking and measured PM10 and the lung function of the respondents. The study population involved 130 women and 130 children. In addition, a community nurse carried out a physical and clinical examination of both the mothers and the children for respiratory and non-respiratory symptoms. The results showed that 37.7% used firewood, 33.1% used kerosene, 17.7% used charcoal and 11.5% used liquefied natural gas. Firewood use followed by charcoal emitted high PM10 at a level of 1640 μg/m3 and 1159 μg/m3, respectively. Charcoal users showed the lowest lung function values and gas users the highest value. A majority (78.5%) of the respondents complained of having cough during and after cooking. Some of them reported breathing problems, eyes and skin irritations. It is recommended that the communities be advised to switch over from the use of biomass fuels to natural gas so as to sustained the natural resources and again, reduced the health impacts of these cooking fuels on the population.

Cloning, Expression, and Homology Modeling of GroEL Protein from Leptospira interrogans Serovar Autumnalis Strain n2

Leptospirosis is an infectious bacterial disease caused by Leptospira species. In this study, we cloned and se- quenced the gene encoding the immunodominant protein GroEL from L. interrogans serovar Autumnalis strain n2, which was isolated from the urine of a patient during an outbreak of leptospirosis in Chennai, India. This groEL gene encodes a protein of 60 kDa with a high degree of homology （99% similarity） to those of other leptospiral serovars. Recombinant GroEL was overexpressed in Escherichia coli. lmmunoblot analysis indi- cated that the sera from confirmed leptospirosis patients showed strong reactivity with the recombinant GroEL while no reactivity was observed with the sera from seronegative control patient. In addition, the 3D structure of GroEL was constructed using chaperonin complex cpn60 from Thermus thermophilus as template and vali- dated. The results indicated a Z-score of -8.35, which is in good agreement with the expected value for a pro- tein. The superposition of the Ca traces of cpn60 structure and predicted structure of leptospiral GroEL indicates good agreement of secondary structure elements with an RMSD value of 1.5A. Further study is necessary to evaluate GroEL for serological diagnosis of leptospirosis and for its potential as a vaccine component.

The Sodium Montmorillonite(na-MMT)is not susceptible to polymer due to its organophilic character and low basal *** primary objective of this study was to improve na-MMT platelets separation by organically modifying it with cation and anion exchanges using Cetyl trimethyl ammonium bromide(CTAB)and Sodium dodecyl sulfate(SDS)*** spacing,presence of functional groups,Zetapotential with particle size analysis and thermal stability of the Organomodified na-MMT(OMMT)were characterized using XRD,FTIR,zeta-potential analyzer and TGA *** basal spacing of CTAB modified OMMTincreased to 19.5Åfrom 11.0Åwhich corresponds to the basal spacing of *** SDS modified OMMT did not show any increase in the basal *** spectra of CTAB modified na-MMT illustrated the attachment of CTAB functional groups to na-MMT,while the same was absent in the case of SDS modified *** zeta-potential of na-MMT shifted from–24.88 mV to 15.66 mV in the case of CTAB modified na-MMTand 12.49 mV for SDS modified na-MMT,indicating a greater surface potential of the modified *** TGA showed greater weight loss for CTAB modified na-MMT than that for na-MMT,indicating the effective na+ion exchange with alkyl amines.
