Two-dimensional structure of density fluctuations is examined during the current jump phase, indicating a change from the open magnetic fields to the closed ones. During the smooth current ramp-up phase the two-dimensional contour of the LiI intensity shows vertically alignment, consistent with the magnetic surfaces. At the inflection point in Ip ramp-up the LiI intensity contour becomes flat in the observation regime and then suddenly a steep gradient and higher intensity regime are formed in the vertical direction. This higher intensity corresponds to a burst of LiI waveform. According to these changes in the contour, it is found that, within ~1 ms around the burst of LiI, a low frequency coherent wave with a long wavelength rapidly grows. The relations with other signals （magnetic flux and microwave stray power） are discussed with respect to the topological change in the magnetic configuration and mode conversion of the incident electromagnetic waves.

Recent progress in the physics and engineering design study for themodification programme of JT-60 is presented. In order to achieve a steady state high-βplasmaoperation, which is the dominant issue of this programme, physics design for the MHD control and thestability analysis is investigated. Engineering design and the R & D for the superconducting coils,irradiation shield are performed well towards the mission of programme.

The distribution of chitinolytic enzymes in eight organs of the golden cuttlefish Sepia esculenta was determined. Chitinase activity (activity of endo-type chitinolytic enzyme) was measured using pnP-(GlcnAc)n (n = 2, 3) as substrates, with high activity detected in the liver, posterior salivary gland, and stomach. β-n-acetylhexosaminidase (Hex) activity (activity of exo-type chitinolytic enzyme) was determined using pnP-(GlcnAc) as a substrate, and high activity was observed in six organs, including the liver, branchial heart, posterior salivary gland, and stomach. In addition, two chitin-binding proteins (CBP-A, CBP-B) were isolated from the liver using a chitin affinity column. Two full-length cDnAs (SeChi-1: 1484 bp;SeChi-2: 1748 bp) encoding chitinases were obtained from the liver of S. esculenta. SeChi-1 contained a 1377-bp open reading frame (ORF) encoding 459 amino acids, and SeChi-2 contained a 1656-bp ORF encoding 552 amino acids. Domain structures predicted from the deduced amino acid sequences of SeChi-1 and SeChi-2 (SeChi-1, SeChi-2) contained signal peptides, a GH Family 18 catalytic domain, one chitin binding domain (CBD) in SeChi-1, and two CBDs in SeChi-2. Proteome analysis revealed that 125 peptide residues of CBP-A were present in SeChi-1, and 116 peptide residues of CBP-B were present in SeChi-2. Organ expression analysis revealed that SeChi-1 and SeChi-2 were expressed only in the liver of S. esculenta. Phylogenetic analysis of SeChi-1, SeChi-2, and GH family 18 chitinases revealed that SeChi-2 belongs to a group of previously reported squid chitinases, while SeChi-1 does not belong to any previously reported group of mollusk chitinases.

In this study, we report that we successfully cloned and sequenced a chitinase gene from the ovotestis of Kuroda’s sea hare Aplysia kurodai. By using reverse transcription-polymerase chain reaction (RT-PCR) and a system for the 5’ and 3’ rapid amplification of cDnA ends, we obtained a 1352 bp chitinase gene (AkChi) from the ovotestis of A. kurodai. AkChi contains a 1263 bp open reading frame that encodes 421 amino acids. The domain structure predicted from the deduced amino acid sequence was an n-terminal signal peptide and a catalytic domain of glycoside hydrolase (GH) family 18 chitinase. A comparative analysis of the deduced amino acid sequences of AkChi with those of the acidic mammalian chitinase of the California sea hare Aplysia californica revealed the highest homology at 83%. The purified chitinase from the ovotestis was digested by trypsin, and 119 residues of digested peptides were consistent with the deduced amino acid sequence of AkChi. We used RT-PCR to evaluate the expression of AkChi in various tissues of A. kurodai, and we observed that AkChi was expressed only in the ovotestis. A phylogenetic tree analysis, performed using the amino acid sequences of AkChi and known GH family 18 chitinases, showed that AkChi was separated from the molluscan chitinases with a chitin binding domain. To our knowledge, this is the first study demonstrating the cDnA cloning of an ovotestis chitinase from a sea hare.

no systematic investigation considering BDnF and its receptor TrkB has been conducted on the effects of mood-stabilizing drugs. We therefore decided to quantify BDnF production and the expression of TrkB-T1 receptor and PLCγ in astrocytoma. 1321n human astrocytoma cells were grown to a sufficient quantity in 5% fetal calf serum (FCS). The mood-stabilizing drugs Li and VPA were added to the therapeutic concentrations of 1 mM and 600 μM, respectively. The production of BDnF was determined by an enzyme-linked immunosorbent assay. The expressions of TrkB-T1 and PLCγ were determined by Western blot. The production of BDnF was significantly higher on Day 7 in the VPA samples (P nd was significantly suppressed beginning on Day 1 in the Li samples (P n, in contrast to BDnF production, was significantly higher in the VPA samples (P γ expression did not change. Li and VPA seem differently affect BDnF production and TrkB-T1 (BDnF receptor) expression.

BACKGROUnD Current guidelines recommend colonoscopy within 24 h for acute lower gastrointestinal bleeding;however,the evidence in support for colonic diverticular hemorrhage(CDH)indications remains *** To investigate the effectiveness of early colonoscopy on the length of hospital stay for CDH *** We conducted a single-center retrospective cohort *** who underwent colonoscopy within 24 h of presentation(early group)were compared with those who underwent colonoscopy beyond 24 h of presentation(elective group).The primary outcome was the length of hospital stay,and secondary outcomes were the identification of stigmata of recent hemorrhage(SRH),rebleeding,red blood cell transfusion more than 4 units,and interventional radiology and abdominal surgery after *** We identified 574 CDH *** were divided into the early(n=328)and elective(n=226)*** propensity score matching,191 pairs were *** length of hospital stay did not significantly differ between the two groups(early group vs elective group;median,7 vs 8 d;P=0.10).The early group had a significantly high identification of SRH(risk difference,11.6%;95%CI:2.7 to 20.3;P=0.02).no significant differences were found in the rebleeding(risk difference,4.7%;95%CI:-4.1 to 13.5;P=0.35),red blood cell transfusion more than 4 units(risk difference,1.6%;95%CI:-7.5 to 10.6;P=0.82),and interventional radiology and abdominal surgery rate after colonoscopy(risk difference,0.5%;95%CI:-2.2 to 3.2;P=1.00).COnCLUSIOn Early colonoscopy within 24 h,on arrival for CDH,could not improve the length of hospital stay.

Background: Miriplatin is a slow-release, lipophilic platinum complex, developed to produce a superior antitumor effect for hepatocellular carcinoma (HCC). However, the miriplatin suspension is highly viscous and can form an embolism in the hepatic artery, which can result in insufficient antitumor effect. Thus, reducing the viscosity of the suspension compound by combining it with the less-viscous cisplatin suspension might reduce or even prevent vessel embolism, while providing the quick-release effects of cisplatin. Purpose: To compare the outcomes of therapy using miriplatin plus cisplatin and cisplatin monotherapy in transcatheter arterial chemoembolization (TACE) for HCC. Methods: We retrospectively evaluated a total of 87 patients with Barcelona Clinic Liver Cancer (BCLC) stage A or B HCC who received conventional TACE using a combination of platinum agents (cisplatin and miriplatin) (n = 50) or cisplatin alone (n = 37) for the first time from September 2006 to December 2012. Short term therapeutic effect was measured by dynamic computed tomography 1 - 3 months after TACE, in reference to the modified Response Evaluation Criteria in Solid Tumors. Treatment-related adverse effects were graded by the national Cancer Institute Common Terminology Criteria (ver. 4.0). 1-, 3-, and 5-year survival rates were calculated. Subgroup analyses were performed by Child-Pugh classification and BCLC criteria. Results: Median duration of follow-up was 35 months (range 7 - 90). Median overall survival was 38 months. Patients who had combination therapy had better 1-, 3-, and 5-year survival rates: 100%, 56.7%, and 26.2%, respectively, compared to monotherapy: 100%, 42.1%, and 9.0%, respectively (p = 0.034). no serious complication or treatment-related mortality was observed in both groups. Conclusion: TACE using miriplatin plus cisplatin was related to a prolonged survival, with comparable adverse effects of TACE using cisplatin alone.

利用23周太阳峰期期间南极中山站(不变磁纬74.5°S)成像式宇宙噪声接收机的观测结果, 对由太阳质子和X射线耀斑引起的日侧电离层吸收事件进行了分析, 定量地给出电离层吸收和X射线耀斑强度的对应关系, 同时还利用北极ny-Alesund(不变磁纬76.08癗)站的观测数据对此关系进行对比研究, 得出了在理论和观测上都较为一致的结论. 同时, 认为M级以上的X射线耀斑才能引起日侧电离层较为明显的吸收.

为了阐明磨石研磨加工层对高碳铬轴承钢JIS SUJ2超长寿命疲劳行为的影响,分别使用经砂纸研磨和电解研磨的砂漏形试样,在室温空气环境下进行旋转弯曲疲劳试验。砂纸研磨试样被除去部分磨石研磨层,电解研磨试样被除去了全部的磨石研磨层。结果表明,两种试样的S-n曲线由位于短寿命区的表面破坏模式和位于长寿命区的内部破坏模式的两条组成,表面破坏模式的S-n曲线受表面粗糙度和表面压缩残余应力的影响。内部破坏模式的S-n曲线不受表面条件的影响,是材料固有的特性。砂纸研磨试样表面破坏模式的疲劳极限最高,是电解研磨试样1．11倍和磨石研磨的1．20倍。表面压缩残余应力对表面破坏模式疲劳极限的影响可以用修正Goodman图表示。还讨论裂纹的萌生和扩展条件,推定超长寿命的疲劳极限。
