A simple, rapid, specific an. precise liquid chromat.graphy—t.n.em mass spect.ophot.met.ic (LC-MS/MS) met.od was developed an. validat.d for t.e quan.ificat.on.of chiral separat.d R-bicalut.mide from S-bicalut.mide, in.human.plasma. t.piramat. (t.M) was used as in.ern.l st.n.ard, added t. plasma sample prior t. ext.act.on.usin. t.but.l met.yl et.er (t.ME). Chromat.graphic separat.on.was achieved on.CHIRALPAK AD-RH column.(150 mm×4.6 mm, 5 μm) wit. act.on.t.ile: 0.1% formic acid Buffer (50:50 v/v) as an.isocrat.c mobile phase wit. a flow rat. of 1.0 mL·min.1. Quan.it.t.on.was performed by t.an.it.on.of 429.0 → 255.0 (m/z) for R-bicalut.mide an. 338.1 → 77.8 (m/z) for t.piramat.. t.e lower limit.of quan.it.t.on.was 20 n.·mL-1wit. a 100 μL plasma sample. t.e con.en.rat.on. of eight.workin. st.n.ards showed lin.arit. bet.een.20 t. 3200 n.·mL-1(r2≥ 0.9990). Chromat.graphic separat.on.was achieved wit.in.6 min. compared t. t.e 15 min.of previous met.ods. t.e average ext.act.on.recoveries of 3 qualit. con.rol con.en.rat.on. were 98.56% for R-bicalut.mide an. 92.42% for t.piramat.. t.e coefficien. of variat.on.was ≤15% for in.ra- an. in.er-bat.h assays. t.erefore a rapid, specific an. sen.it.ve LC-MS/MS met.od for t.e quan.ificat.on.of R-bicalut.mide in.human.plasma was developed an. validat.d can.be used in.t.e bioequivalen.e st.dy of t.is drug.

Backgroun. an. object.ve: Chron.c rhin.sin.sit.s (CRS), a common.disease worldwide, has a major impact.on.pat.en.s’ qualit. of life (QoL). In.recen. years, t.e measuremen. of healt.-relat.d QoL has made it.possible t. assess t.e pat.en.’s st.t. of healt., t.e severit. of t.e CRS an. t.e t.eat.en.. t.e aim of t.is st.dy was t. assess t.e QoL of pat.en.s wit. CRS in.a hospit.l set.in. in.Kin.hasa. Met.ods: t.is was a cross-sect.on.l an.lyt.cal st.dy con.uct.d from Jun. 2020 t. May 2021 in.t.e En. service of t.e Kin.hasa Un.versit. Hospit.l an. t.e Mon.ole Hospit.l Cen.er. t.e st.dy in.olved 113 pat.en.s aged at.least.18 years, whose QOL was compared wit. t.at.of a con.rol group con.ist.n. of 100 n.n.pat.en.s. QOL was assessed usin. t.e Sin.-n.sal Out.ome t.st.(Sn.t.22) Quest.on.aire. Result.: t.e mean.age of t.e pat.en.s was 41.0 years (18 - 74 years) an. t.at.of t.e con.rols was 39.1 years (19 - 77 years). More t.an.half of t.e part.cipan.s were un.versit. graduat.s. t.e median.Sn.t.22 score was 44 (14 - 78) in.pat.en.s an. 7 (1 - 40) in.con.rols. Compared wit. subject. wit.out.CRS, pat.en.s wit. CRS had sign.fican.ly higher scores in.all four domain. of t.e Sn.t.22. Aroun. 9 out.of 10 pat.en.s had a severely impaired QoL compared wit. 2 out.of 10 con.rols. Con.iderin. mildly impaired QoL, we foun. t.at.in.t.e ot.logical an. sleep domain., scores were n.t.sign.fican.ly differen. bet.een.pat.en.s an. con.rols. In.t.e case of severely impaired QoL, however, on.y t.e ot.logical domain.showed a n.n.sign.fican. differen.e in.scores bet.een.t.e t.o groups. Con.lusion.
