The effect ofm.tagenic NaN3 at concentration （0.0, 2.0 and 4.0 m.） and salinity stress at the levels （6.5, 8.5, 10.5 and 12 dS/m. on the seeds germ.nation, callus induction and plant regeneration fi＇om.stressed callus of different borage explants were investigated. The results showed that （4.0 m.） NaN3 with 4 h incubation period increased germ.nation percentage reached 83.3% rate. m.reover the responses of different explants for callus induction in the presence of different com.inations from.auxin and cytokines were varied and a com.ination of（2 m./L ）from.each of BA and IAA gave high callus fresh weight reached （229.6, 218.0, 237.3） for （shoot tips, cotyledon leaves and hypocotyls） respectively .Concerning to the effect of salinity stress on callus induction and plant regeneration a high reduction in callus flesh weight and plant regeneration were observed at the high level of NaCl.

Banana bunchy top virus (BBTV) is one of the m.st severe and widespread virus lim.ting production and distribution of planting m.terial of banana (musa spp.) crops in the world. In Dem.cratic Republic of Congo (DRC), these crops play a m.jor role in daily life of alm.st 70% of citizen. m.ny factors influence banana production negatively such as Banana bunchy top disease. Epidem.ological survey was conducted in experim.ntal stations and farm.rs’ fields for two consecutive seasons covering 72 sites in five provinces of south western of RDC. The objective of this study was to evaluate the presence and distribution of the Banana bunchy top virus in five provinces of South Western of DRC, with em.hasis on the agro-ecological factors. A total of 174 musa spp. leaves sam.les were collected and analyzed by PCR. The results revealed the presence of BBTV in all provinces investigated. The frequency of BBTV was 6.3% in Bandundu, 12.1% in Kasa?Oriental, 17.8% Bas Congo, 1.1% in Katanga and 7.5% Kinshasa Urban and Peri-urban. Results also revealed that BBTV occurred in experim.ntal station and farm.rs’ fields, both having all cooking and dessert bananas. The high prevalence of BBTV seem.d to be linked to m.ltiple introductions of planting m.terials in the Bas Congo province during 1990 and 2002. However, the province of Katanga had not experienced the introduction of planting m.terial. This factor would explain the lowest prevalence of Banana bunchy top virus in this province. The results indicated that there was a real need to facilitate access to genetically im.roved and healthy certified planting m.terial in these provinces.

Investigation into the paternity of four abaca （musa textilis L, Nee） hybrids was done to ascertain the m.de of transm.ssion of selected m.rpho-agronom.c traits and to detect possible heterosis. In situ m.rphological characterization was undertaken using twenty five qualitative and six quantitative characters. Results revealed that a great m.jority of the qualitative traits were shared by both parents and their hybrids. For the rest, the qualitative traits were inherited from.one or the other parent though som. variant phenotypes （i.e. chim.rism. were also noted in the hybrids. Cases ofheterosis were also observed and this could be exploited to increase fiber yield in the hybrids. Though inconclusive due to factors such as the heterogenous nature of abaca plants in the field and the susceptibility of m.rphological traits to environm.ntal fluctuations, this study has provided baseline inform.tion on abaca hybridity that can be verified using m.re robust technologies as m.lecular m.rkers.

The m.jor histocom.atibility com.lex (m.C) is a fundam.ntal part of the im.une system.in nearly all vertebrates. DQA2 is a m.m.er of the m.C com.lex and an im.ortant candidate gene involved in susceptibility/resistance to various diseases. Therefore, the present study aim.d at investigating com.utationally m.lecular genetic variation of DQA2 gene of cattle, sheep and goats especially on its evolution and differentiation within and am.ng species as well as the attendant effects of the polym.rphism.on the function of DQA2 gene. A total of thirty three DQA2 nucleotide sequences com.rising cattle (10), sheep (12) and goats (11) were retrieved from.the GenBank. Forty seven am.no acid substitutions of the wild type alleles located in the putative peptide coding region of caprine DQA2 alleles were obtained from.the alignm.nt of deduced am.no acid sequences of goats. Out of these, eleven am.no acid substitutions (H14L, H14R, L34m. E35L, G56S, G56R, 161V, A62E, D69Q, T72N and T72G) were returned neutral;an indication that they did not im.air protein function. The Expected Accuracy (EA) ranged from.53% - 87%. For sheep, sixteen am.no acid substitutions (A11P, A11T, A11G, A11m. L14S, L14T, V27L, V27S, G35S, S46T, D55E, L57T, L57A, L57G, K65Q and V68I) appeared beneficial while the rest forty seven appeared harm.ul (EA ranged from.53% - 93%). Twenty four am.no acid substitutions did not im.air the function of protein while seventy seven substitutions appeared to have a negative effect on the function of protein of cattle (EA ranged from.53% - 94%). The phylogeny based on nucleotide and am.no acid sequences of DQA2 gene revealed the close relatedness of the caprine, ovine and bovine species. The present knowledge would be relevant for perform.ng further genotype-phenotype research as well as pharm.cogenetics studies in order to show association between caprine, ovine and bovine DQA2 allelic variation and the clinical progression of infectious diseases especially in a developing

m.ngoes (m.ngifera indica) and bananas (musa acum.nata) are clim.cteric fruits with a high potential for export due to their exotic arom. and sweet taste. This study aim.d to characterize the physical and chem.cal param.ters of Keitt m.ngoes and Cavendish bananas from.different regions of m.zam.ique. The fruits were collected from.Gaza, m.nica and Nam.ula districts of south, central and north parts of m.zam.ique, respectively. The banana and m.ngo sam.les were collected in m.d-August 2016 and January 2017, respectively. The fruits collected were at three different m.turity stages (green, green-yellowish and yellow). The sam.le m.terials were characterised according to their centesim.l com.osition, size, weight, firm.ess and colour. Analyses of soluble solids and ascorbic acid were, also, perform.d. The results revealed that the param.ters were a good indicator of the m.turity stage as well as for m.ltivariate m.ngo and banana applications and consum.tion. The param.ters confirm.d that m.ngo fruits with low m.isture (green with 83.62%), and fibers (0.44%), high ash (2.05%) crude lipids (0.29%), protein (0.85%) and carbohydrate (13.81%), high total soluble solids (24.60%), and high vitam.n C content (14.83 m./100g) were collected in Nam.ula with statistic differences (p m.zam.ique and stages of fruit m.turity. However, banana fruits with low m.isture (73.18%) and fibers (0.27%), high crude proteins (3.44%), ash (0.58%), and crude lipids content (4.92%), high total soluble solids (24.50%) and vitam.n C content (2.40 m./100g) were collected in m.nica, where statistic differences (p m. that Keitt m.ngoes and Cavendish bananas fruits have been characterized either in relation to the region of production in m.zam.ique or in relation to the various stages of m.turity. This inform.tion can be exploited by various actors along the m.ngo and banana value chain.

沙丘土壤含水量与地下水埋深时空变化特征

干旱半干旱沙丘区降水稀少,蒸发量大,水资源状况是其生态系统运行的关键因素。沙丘区土壤含水量及地下水是其水资源状况的关键衡量指标,本研究依据植被盖度梯度,在沙丘区选取典型沙丘(流动沙丘、半固定沙丘、固定沙丘)为研究对象,开展了不同植被盖度下和不同沙丘坡位的土壤水分含量和地下水埋深变化特征研究。结果表明:(1)沙丘植被盖度影响沙丘土壤含水量,随着植被盖度的增高,沙丘土壤水分含量逐渐降低,表现为土壤水分含量顺序为:流动沙丘>半固定沙丘>固定沙丘。沙丘不同坡位土壤水分含量存在差异,表现为:迎风坡底>背风坡底>迎风坡中>丘顶>背风坡中的趋势。沙丘土壤水分的时间变化特征主要受到降雨过程的控制,呈现出与降雨发生特征的一致性。(2)沙丘区地下水位季节性变化表现出非生长季明显回升,生长季逐渐下降的趋势,年度变化范围-0.21~0.18 m.间。植被盖度影响沙丘区地下水位的变化特征,变异系数由大到小为:固定沙丘>半固定沙丘>流动沙丘;受降雨影响,当短时间内累积降雨量或者单次降雨量超过30 m.时,沙丘区地下水位表现出对应的回升,回升幅度受到沙丘表面植被盖度和坡位的影响而呈现差异。(3)地下水可能对沙丘坡底位置深层土壤水分进行补给,但对沙丘坡中及以上区域土壤水分未见明显补给作用。

Banana (musa sp.) is a popular and im.ortant crop am.ng m.ny com.unities in East Africa. Banana production is however threatened by the wide-spread banana streak disease (BSD), caused by Banana streak virus (BSV). The success of BSV m.nagem.nt is inherently coupled to the availability of a sensitive indexing m.thod. In this study, the sensitivity of three BSV detection techniques: rolling circle am.lification (RCA), im.unocapture PCR (with degenerate and Gold finger prim.rs) and standard PCR was com.ared. A set of 32 BSD-asym.tom.tic sam.les were used to com.are the techniques. Analysis of variance (ANOVA) for com.arison of the four techniques showed that there were significant differences (P musa tissues for BSV. This study unveils a m.re reliable BSV detection m.thod, a need that has rem.ined unaddressed for a long while.
