Hepg2 cells support viral replication and gene expression of hepatitis C virus genotype 4 in vitro

AIM： TO establish a cell culture system with longterm replication of hepatitis C virus （HCV） genome and expression of viral antigens in vitro. METHODS： Hepg2 cell line was tested for its susceptibility to HCV by incubation with a serum from a patient with chronic hepatitis C. Cells and supernatant were harvested at various time points during the culture. Culture supernatant was tested for its ability to infect na＇ive cells. The presence of minus （antisense） RNA strand, and the detection of core and E1 antigens in cells were examined by RT-PCR and immunological techniques （flow cytometry and Western blot） respectively. RESULTS： The intracellular HCV RNA was first detected on d 3 after infection and then could be consistently detected in both cells and supernatant over a period of at least three months. The fresh cells could be infected with supernatant from cultured infected cells. Flow cytometric analysis showed surface and intracellular HCV antigen expression using in house made polyclonal antibodies （anti-core, and anti-E1）. Western blot analysis showed the expression of a cluster of immunogenic peptides at molecular weights extended between 31 and 45 kDa in an one month old culture of infected cells whereas this cluster was undetectable in uninfected Hepg2 cells. CONCLUSION： Hepg2 cell line is not only susceptible to HCV infection but also supports its replication in vitro. Expression of HCV structural proteins can be detected in infected Hepg2 cells. These cells are also capable of shedding viral particles into culture media which in turn become infectious to uninfected cells.

Effect of Pb Ions on the Ionic Conductivity of Some Silicate glass Systems

Conductivity of glasses in the systems ***, (where R=Ca, Sr or Ba) was investigated between room temperature and 530 K. The dependence of log resistivity as well as the activation energy on the PbO content has been studied. Based on the present experimental results, the possible different conduction mechanisms in such glasses are discussed. It was postulated that Pb2+ ions may represent the major charge carrying species in these glasses. This assumption was confirmed by the calculations of the mean distance between the interstitial Pb2+ ions and the effective center of the O2- ions in the glass networks. The variation in the values of the density and the molar volume with PbO content is also discussed in view of the obtained activation energies for the studied glass-systems.

Renewable energy is one of the essential elements of the social and economic development in any civilized *** use of fossil fuels and the non-renewable form of energy has many adverse effects on the most of *** the high potential of renewable energy sources in Yemen and the absence of similar studies in the region,this study aimed to examine the wind energy potential of Hodeidah-Yemen Republic by analyzing wind characteristics and assessment,determining the available power density,and calculate the wind energy extracted at different *** average wind speed of Hodeidah was obtained only for the data currently available for the five years 2005–2009(due to the current economic and the political situation in Yemen).The results show that the average wind speed in the five years is(25.2 W/m2 at 10 m,93.9 W/m2 at 30 m,and 173.5 W/m2 at 50 m).The average yearly wind power density(25.2 W/m2 at 10 m,93.9 W/m2 at 30 m and 173.5 W/m2 at 50m),and the average yearly energy density(220.8 KWh/m2/year at 10 m,822.6 KWh/m2/year at 30 m and 1519.9 KWh/m2/year at 50 m).This research is a preliminary assessment of the potential of wind energy in Hodeidah,which provides useful information for developing wind energy and an efficient wind *** to the International Wind Energy Rating criteria,the region of Hodeidah falls under‘Class 2’and is classified as‘Marginal’for most of the year.

Dust Acoustic Solitary Waves in Saturn F-ring's Region

Effect of hot and cold dust charge on the propagation of dust-acoustic waves （DAWs） in unmagnetized plasma having electrons, singly charged ions, hot and cold dust grains has been investigated. The reductive perturbation method is employed to reduce the basic set of fluid equations to the Kortewege-de Vries （KdV） equation. At the critical hot dusty plasma density Nho, the KdV equation is not appropriate for describing the system. Hence, a set of stretched coordinates is considered to derive the modified KdV equation. It is found that the presence of hot and cold dust charge grains not only significantly modifies the basic properties of solitary structure, but also changes the polarity of the solitary profiles. In the vicinity of the critical hot dusty plasma density Nho, neither KdV nor mKdV equation is appropriate for describing the DAWs. Therefore, a further modified KdV （fmKdV） equation is derived, which admits both soliton and double layer solutions.

Radioactivity and mineralogy of the altered granites of the Wadi ghadir shear zone, South Eastern Desert, Egypt

Quartz-diorite, gneissose granodiorites, two-mica granite and perthite leucogranie are the main rock units cropping out in the Wadi ghadir area, South Eastern Desert of Egypt. Along the NNE-SSW mega-faults, a broad brittle shear zone is developed in the ghadir two-mica granite. Brittle deformation is manifested by severe myloniti-zation and alteration of these granites. These sheared altered granites are characterized by the presence of radioactive mineralization, associated with alteration features such as silicification, hematization and kaolinitization. Radioelement measurements revealed that the unaltered and altered two-mica granites are considered as uraniferous granites. The average uranium and thorium contents in the unaltered two-mica granites are 12.29×10-6 and 19.81×10-6, respectively, and the average Th/U ratio is 1.62. The altered granites exhibit higher concentrations of U (averaging 97.949), but have lower Th and Th/U ratios (13.83 and 0.16, respectively), which indicates uranium enrichment in the granites. Binary relations of eTh/eU against either eU or eTh and eU with eTh in the studied gran-ites suggest that the distribution of radioactive elements not only magmatic (positive correlation between eU and eTh), but also due to hydrothermal redistribution of radioelements (weak correlation between eU and eTh/eU). The magmatic U and Th are indicated by the presence of uraninite, thorite, zircon and monazite whereas the evidence of hydrothermal mineralization is the alteration of rock-forming minerals such as feldspar and the forma-tion of secondary minerals such as uranophane and pyrite. Microscopic, XRD and scanning electron microscopic studies revealed the presence of uraninite, uranophane, thorite, Ce-monazite and zircon, in addition to phlogopite-fluor mica in the studied altered granites of the Wadi ghadir shear zone.

gastric cancer remains a global killer with a shifting burden from the developed to the developing world. The cancer develops along a multistage process that is defined by distinct histological and pathophysiological phases. Several genetic and epigenetic alterations mediate the transition from one stage to another and these include mutations in oncogenes, tumour suppressor genes and cell cycle and mismatch repair genes. The most significant advance in the fight against gastric caner came with the recognition of the role of Helicobacter pylori （H pylori） as the most important acquired aetiological agent for this cancer. Recent work has focussed on elucidating the complex host/microbial interactions that underlie the neoplastic process. There is now considerable insight into the pathogenesis of this cancer and the prospect of preventing and eradicating the disease has become a reality. Perhaps more importantly, the study of H pylori-induced gastric carcinogenesis offers a paradigm for understanding more complex human cancers. In this review, we examine the molecular and cellular events that underlie Hpyloriinduced gastric cancer.

gamma-ray measurements of natural radioactivity in sedimentary rocks from Egypt

The aim of this study was to measure concentrations and distributions of natural radionuclides occurring in rocks. The activity concentrations (Bq·kg-1) of the naturally occurring radionuclides 226Ra, 232Th, and 40K in sedi- mentary rock samples from Eastern Desert (Um el-Huetat), Nile Valley (gebel Owina) and from southwest Sinai (Wadi ghweiba) were measured using a high-purity germanium detector. The samples under investigation (clay, shale and sandstone) were used as raw materials in the construction industry (bricks, ceramics, cement, fillers, etc.). Though the sediments of Egypt have already been investigated in the geological and mineralogical aspects, it is necessary to investigate the natural radioactivity in order to complete their classification. The average concentration values of 226Ra, 232Th, 40K in the surveyed samples were 47 ± 7 , 21± 5, 393±19 ***-1 (clay); 23 ± 5, 30 ± 6, 563 ± 24 ***-1 (shale); and 17 ± 4, 14 ± 4, 299 ± 17 ***-1 (sandstone), respectively. All sediment samples have radium equivalent activities ranging from 55 to 115 Bq·kg-1, lower than the limit set in the OECD Report (370 Bq·kg-1). The overall mean outdoor terrestrial gamma dose rates fluctuate from 28 to 55 ngy·h-1. The external gamma radiation dose due to natural radionuclides present in the samples have been computed and compared with the global averages. In terms of the radiation safety, the natural radioactivity of the sediment in Egypt is below the recommended limits of the gamma dose rate. Therefore, they can be used for all kinds of public buildings.

Computational fluid dynamics(CFD)simulations of multi-lobe progressive cavity(PC)pumps are limited in the literature due to the geometric complexity of the pump,which places numerous restrictions on the grid generation *** present study attempts to alleviate such restrictions by developing a detailed numerical procedure for the numerical simulations of multi-lobe progressive cavity *** profile equations for the multi-lobe configuration at any section at each instant of rotation are presented.A structured grid generation method is developed to generate mesh files required for CFD simulations of multi-lobe PC *** from the present procedure are validated against single-lobe PC pump numerical results available in the ***,a numerical parametric study is carried out to investigate the effect of the number of lobes,the stator pitch,the circular fillet and clearance on the volumetric efficiency of PC pumps with viscous oils.

Lifestyles of Older Adults in Castilla-La Mancha(Spain):Influence of Sex,Age,and Habitat

This paper presents the quantitative research results of a comprehensive study titled“Current Profle of Older Adults in Castilla-La Mancha(Spain).”The study aims to understand and describe the characteristics of individuals aged 65 and above within this autonomous region,with a focus on the variables of sex,age,and habitat.A cross-sectional approach was employed,utilizing quantitative methods to collect *** gather information,a customized questionnaire was developed to address the study objectives,covering various aspects such as sociodemographic factors,living conditions,leisure activities,social participation,and cultural *** survey was conducted via telephone,ensuring representation by employing gender quotas,age-group stratifcation,and considerations for habitat size.A representative sample of 1,065 participants was included in the *** fndings reveal a degree of homogeneity in the lifestyles of older adults,with discernible variations infuenced by factors such as sex,habitat,and *** results shed light on the diverse habits and experiences of older adults,emphasizing the signifcance of habitat conditions,social participation,and the aging process.
