Ethanol Fermentation by High-Stress-Tolerance Aquatic Yeasts and Their Mutants

Bioethanol is thought to be a renewable source of energy, because the biomasses used to make ethanol, such as sugar cane and its residual substance, molasses, are resources that can be continuously produced. But the practical use of ethanol to replace fossil fuels or atomic energy has been limited, because the production efficiencies of ethanol in relation to its substrates are not so high. Thus, for industrial production of the bioethanol, yeast fermentation would ideally be carried out in biomasses containing more highly concentrated carbohydrates. However, the environmental stresses in highly concentrated cultures might weaken the yeast’s physiological activities. From various kinds of aquatic yeast with stress tolerance, Torulaspora derbrueckii F2-11 and Wicherhamomyces anomalus AN2-64 were selected as candidates for high-sugar-tolerance yeasts as they showed remarkable growth in the YPD + sorbitol (600 g/L) medium at 25°C for 120 hrs. When the amounts and kinds of sugar alcohols in the cells of the two strains were measured in cultures containing 20 g/L or 400 g/L of D-glucose, maltose, or sucrose, the main two sugar alcohols that accumulated as the sugar concentration increased were glycerol and arabitol. Mutation by ethyl methanesulfonate of the parent strains T. derbrueckii F2-11 and W. anomalus AN2-64 induced mutants F2-11M or AN2-64M, which showed higher sugar, heat, and ethanol tolerances than their respective parents. Ethanol productivities and sugar assimilation activities of the mutants were also higher than those of the parents in the 25% (v/v) molasses.

Effect of zinc supplementation in patients with type C liver cirrhosis

Zinc is often deficient in patients with liver cirrhosis, and treatment with zinc provides short-term improvement in protein metabolism. However, the long-term effects of zinc have not been fully clarified. The present study aimed to analyze the effect of zinc on the long-term clinical course, especially hepatocarcinogenesis, in type C liver cirrhosis. Am- ong patients with type C liver cirrhosis visiting our hospital between June 1998 and January 2009, th- ose with a serum albumin level ≤3.5 g/dL and a serum zinc level ≤70 μg (1.07 μmol)/dL were selected. Thirty-seven patients were randomly divided into 2 groups: group B (12 g/day branched-chain amino acid granules) and group BZ (same as group B plus 100 mg/day - 600 mg/day zinc sulfate or 150 mg/day - 225 mg/day polaprezinc). Multivariate analysis revealed that the administration of zinc was not a significant determinant, but pretreatment serum zinc levels (hazard ratio [HR], 0.921;95% confidence interval [CI], 0.853–0.994) and serum zinc levels less than 80 μg (1.22 μmol)/dL 12 months after beginning this study (HR, 6.866;95% CI, 1.399 - 33.707) were significant determinants of carcinogenesis and death. Serum albumin levels in patients whose serum zinc levels had not increased up to 80 μg/dL by the third year of this study were significantly lower (p = 0.023) than those of patients that had increased up to 80 μg/dL. Conclusions: In type C liver cirrhosis with zinc deficiency, administration of zinc does not improve cancer-free survival. However, serum zinc levels can predict outcomes in patients with type C liver cirrhosis. However, although zinc may play a role in hepatocarcinogenesis, the precise implications remain to be clarified.

We examined the effect of a combination of astaxanthin （AX） supplementation, repeated heat stress, and intermittent reloading （IR） on satellite cells in unloaded rat soleus muscles. Forty-nine male Wistar rats （8-week-old） were divided into control, hind-limb unweighting （HU）, IR during HU, IR with AX supplementation, IR with repeated heat stress （41.0-41.5 ℃ for 30 min）, and IR with AX supplementation and repeated heat stress groups. After the experimental period, the antigravitational soleus muscle was analyzed using an immunohistochemical technique. Our results revealed that the combination of dietary AX supplementation and heat stress resulted in protection against disuse muscle atrophy in the soteus muscle. This protective effect may be partially due to a higher satellite cell number in the atrophied soleus muscle in the IR/AX/heat stress group compared with the numbers found in the other groups. We concluded that the combination treatment with dietary AX supplementation and repeated heat stress attenuates soleus muscle atrophy, in part by increasing the number of satellite cells.

Influence of Sex Differences on Temporal Sequence of Sensations after Ingesting Fruit-Flavored Tea—A Preliminary Study

The goal of this preliminary study was to examine the effect of participants’ sexes on the temporal sequence of dominant sensations elicited by fruit-flavored teas. Twelve healthy young adults were assigned to male (n = 6) and female (n = 6) groups. Both groups were evaluated for four sensations using four fruit-flavored teas with 0.05 M sucrose using the temporal dominance of sensations (TDS) method. Only two sensations, sweetness and fruitiness, were consistently reported in the two groups. The male group first reported fruitiness and then sweetness as the dominant sensation following ingestion of the samples. Conversely, the female group reported these two dominant sensations in the reverse order. Significant dominant durations between the two sensations largely varied among the samples in both groups. These results suggest that there are sex-based differences in the temporal sequence of dominant sensations elicited by fruit-flavored teas as evaluated by the TDS method.

Simplifying Laparoscopic Surgery for Left Side Colon and Rectal Cancer Using Linear Stapler for Vascular Ligation: A Prospective Cohort Study

Introduction: Systematic lymphadenectomy and ligation of the feeding artery is extremely important when performing radical resection in colorectal cancer. However, vascular surgery via laparoscopy requires advanced skills and techniques;thus, this procedure needs to be simplified while maintaining quality of the surgery to make it a preferred technique for the surgeons. Methods: There were 49 patients who underwent laparoscopic sigmoidectomy or anterior resection till T2 level for sigmoid colon cancer and recto-sigmoid colon cancer. We analyzed short-term and long-term outcomes between stapling ligation and clipping ligation techniques used in these surgeries. Results: The mean volume of blood loss in the stapling ligation group was 12.8 ± 12.3 ml, which was significantly lower than 41.9 ± 71.2 ml of mean volume of blood loss in the clipping ligation group. There was no significant difference in the mean duration of surgery, the mean number of harvested lymph nodes, morbidity, recurrence, and 5-year relapse free survival rates between the 2 groups. Conclusions: This study demonstrates a surgical technique using staplers for vascular treatment of tumor-feeding arteries as a new technical improvement in laparoscopic colectomy for the treatment of early-stage colon cancer. We found that the described procedure was technically safe, simple, convenient, and oncologically valid.

Association between high cystatin C levels and carotid atherosclerosis

We developed new laser desorption ionization method(DIUTHAME: Desorption Ionization Using Through Hole Alumina MEmbrane) as matrix-free ionization method. DIUTHAME is alumina membrane which has many regularly aligned through holes. The diameter of through hole is submicron meter. The ionization principle of DITHAME is that place the DIUTHAM on the sample, wait until the molecules in the sample rise to the surface by capillary action, and then irradiate a laser beam onto molecules that have risen to the surface, the target molecules are desorbed and ionized due to the effect from the time convexo-concave structure. And the new DITHAME method has none background noise, high reproducibility and high resolution. For MALDI Imaging MS, recommended thickness of cutting sample is under 10 micro meter. However, there are some samples which is impossible to cut with the thin thickness. This time, took MS imaging of strawberry which is difficult to cut thinner because it contains high amount of water. And the result shows that: 1. Possible to measure saccharide derived from strawberry by using DIUTHAME 2. Measured MS distribution of all strawberry surface by negative ion mode.(Succeed in taking MS image by transfer method.) 3. Will investigate the mass peaks gotten by negative ion mode as next step.

AIM To investigate the association between carotid atherosclerosis and cystatin C(CysC) and to determine the optimal CysC cut-off *** One hundred twenty-eight subjects were included in this study. Atherosclerosis was defined as a maximum carotid plaque thickness(MCPT) of greater than 2 mm. A receiver operating characteristic curve analysis was used to determine the diagnostic value of serum CysC for atherosclerosis. The subjects were divided into two groups according to the CysC cut-off value. We screenedfor diabetes, hypertension, dyslipidemia, smoking status, alcohol consumption, and exercise behavior. The association between atherosclerosis and CysC levels was assessed using multivariate *** The subjects were then divided into two groups according to the CysC cut-off value(0.73 mg/L). The median age of the high CysC group was 72 years(85% males), whereas that of the low CysC group was 61 years(63% males). The CysC levels were significantly correlated with Cr and estimated glomerular filtration rate(eGFR) values. Bodymass index, visceral fat area, hypertension, diabetes mellitus, and MCPT were significantly higher in the high CysC group than in the low CysC group. Furthermore, the eG FR was significantly lower in the high CysC group. Regarding lifestyle habits, only the exercise level was lower in the high CysC group than in the low CysC group. Multivariate analysis, adjusted for age and sex, revealed that high CysC levels were significantly associated with an MCPT of ≥ 2 mm(odds ratio: 2.92; 95%CI: 1.13-7.99).CONCLUSION Higher CysC levels were associated with an MCPT of ≥ 2 mm. The CysC cut-off value of 0.73 mg/L appears to aid in the diagnosis of atherosclerosis.
