碱性果胶酶(Alkaline polygalacturonate lyase,PGL, E.C.4.***.2.2),是一种广泛应用于纺织工业棉织物煮炼中的酶。为了提高发酵法生产PGL的产量,在前期的研究中从一株PGL高产菌株中扩增出编码该酶的基因,并成功表达于毕赤酵母(P. pastoris) GS115中。本论文在此基础上,通过详尽分析诱导阶段培养基不同pH和甲醇浓度对P. pastoris GS115高效表达碱性果胶酶的影响,提出了pH控制策略和甲醇浓度控制策略,并将两者进行结合,最终实现了PGL的高效生产。在考察培养基pH对PGL生产的影响时,首先将菌体生长阶段的pH控制在5.5,诱导阶段采用不同的pH;在甲醇在线控制策略中,培养基的pH值控制在5.5,诱导阶段通过在线流加不同浓度的甲醇研究甲醇浓度对PGL生产的影响。为了进一步提高PGL的产量,在综合考虑甲醇消耗、最优pH、细胞活力和PGL产量的基础上,把诱导阶段分为两个时期(菌体生长阶段结束至诱导后40 h的诱导阶段称为过渡时期),并将pH控制策略结合甲醇在线控制策略应用于诱导阶段,与恒速流加甲醇相比,该策略使PGL的产量提高了100%,达到1631 U/mL。同时,通过检测细胞活力、AOX酶活和胞内能荷水平等参数,证明了该策略在高效生产PGL的过程中能有效地促进菌体生产PGL的能力。前期的研究成果为:从分离筛选得到的一株碱性果胶酶高产菌株Bacillus sp. WSHB04-02中,扩增出编码碱性果胶酶的基因,并成功表达于P. pastoris GS115中;通过优化培养基成分、采用高密度发酵以及考察甲醇浓度和细胞浓度的比率等因素,使PGL的产量达到800 U/mL。本论文在此基础上对PGL生产的关键因素pH和甲醇浓度进行了以下研究:1)为提高重组P. pastoris GS115发酵生产碱性果胶酶的产量和生产强度,在3 L发酵罐中考察了6个不同的pH(6.5、5.5、4.5、3.5、5、4)对重组毕赤酵母生产PGL的影响。结果发现,在pH为4.5时,PGL产量达到1362 U/mL,细胞干重为144 g/L,发酵结束时,细胞活力仍然可以维持在80%,在整个诱导阶段,培养基中甲醇浓度维持在17 mL/L;而其他pH水平对PGL的生产则没有很明显的促进作用。2)在pH控制策略的基础上,为了研究甲醇浓度对PGL发酵生产的影响,在3 L发酵罐上依次考察了5个不同的甲醇水平(8 mL/L,,12mL/L,16 mL/L,20 mL/L和24 mL/L)对发酵产酶的作用。结果表明,培养基中维持16 mL/L的甲醇浓度最有利于PGL的生产,PGL的产量可以达到1260 U/mL,细胞干重为140 g/L,但是PGL产量仍然稍低于以10 mL/L的速度恒速流加甲醇时的产量。胞内ATP、ADP和AMP的水平分别为1.2 g/L,0.04 g/L ,0. 28 g/L,发酵结束时细胞活力为77%。3)在以上研究的基础上,本论文第一次提出了“过渡时期”的概念,在本研究中,根据菌体的生理特性,将菌体生长阶段结束之后至诱导后40 h之间的一段时间称之为过渡时期。在过渡时期采用pH 5.5和较低的甲醇浓度(12 mL/L)诱导菌体生产PGL,之后的诱导阶段pH设为4.5,分别采用16 mL/ L,20 mL/L和24 mL/L三个不同的甲醇浓度,结果发现在整个诱导阶段,采用两阶段pH控制和甲醇浓度控制策略即pH 5.5,12 mL/L(过渡时期)和pH 4.5,16 mL/L (过渡时期之后的诱导阶段)可以进一步促进菌体生长和PGL生产,菌体浓度可达到162 g/L,PGL产量达到1634 U/mL,发酵结束时,细胞活力仍保持在89%,发酵液中蛋白酶浓度降至0.08 U/mL。

The performance test of a CsI(Tl) crystal (70×27×23 mm3) was performed by applying the pulse shape discrimination technique for identification of light charged particles .The crystal is coupled to a photomultiplier tube during an experiment with 6He *** pulse waveform is fully recorded by employing a high precision digital *** fast and slow gates are used for the pulse shape discrimination and the best values for the gate widths were determined to be 0.5 μs and 1.67 μs,*** 6He,4He and 3He are successfully discriminated with this technique.

In this paper,propagation characteristics of electromagnetic electron cyclotron(EMEC)waves based on kappa-Maxwellian distribution have been investigated to invoke the interplay of the electric field parallel to the Earth’s magnetic field and auroral trapped *** dispersion relation for EMEC waves in kappa-Maxwellian distributed plasma has been derived using the contribution of the parallel electric field and trapped electron *** results show that the presence of the electric field has a stimulating effect on growth rate,which is more pronounced at low values of wave *** is also observed that as the threshold value of trapped electron speed is surpassed,it dominates the effect of the parallel electric field and EMEC instability is enhanced *** electric field acts as another source of free energy,and growth can be obtained even in the absence of trapped electron drift speed and for very small values of temperature *** the present study reveals the interplay of the parallel electric field and trapped electron speed on the excitation of EMEC waves in the auroral region.

In recent decades, the migration rates of the large cities of Punjab have been risen up to a considerable level due to the lack of employment opportunities as well as lack of facilities in the rural areas of the province. It has caused an unprecedented and unplanned urbanization across the urban areas of the province. This study has been undertaken to perform fractal analysis about the sprawl in rapidly growing city. GIS and remote sensing data have been used in this study as an emerging technology which is cost effective as well as accurate at the same time. Landsat images have been taken for the study and the sprawl has been calculated with the analysis of the data of each decade for past more than 40 years. It has been observed that the built up area is 47.8 to 141.12 Sq. Km whereas the pattern of urban settlement has been classified as clustered and linear, following the roads network. The temporal population growth also seconded these results. The population growth rate and population density increase, are based on the pixel based extraction of the data from satellite imagery for the period of 2000 to 2014, which is taken as a decision support tool. In 2000, the population of the district increased from 2,071,694 (1981 census) to 2,939,907 and then in 2013, it became 4,384,191 at a rate to 2.93% upturn per annum. Moreover, the study also reveals the extent of the growth of other land uses as well which may be taken as a reference that in an agricultural country like Pakistan, the natural resources are being wasted (by urbanization of the fertile land). There must be some master planning to avoid such things in the other cities as well.

The temporal variations of mesozooplankton abundance and biomass （1-Volumetric method by settling volume and displacement volume and 2- Gravimetric method through wet weight, dry weight and ash-free dry weight） with relation to environmental parameters were studied in the mangrove creek area of Karachi coast, Pakistan. The data of mesozooplankton samples along with environmental parameters （temperature, salinity, pH, etc.） were collected during January 1998 to December 1998 from two creek stations. The abun-dance of mesozooplankton also exhibited seasonal trends at both stations. At Sta. S2, the highest and low-est abundance values were observed during post-monsoon and southwest monsoon respectively whereas, at Sta. S1, a clear trend of high abundance in pre-monsoon to low abundance in southwest monsoon was observed. Mesozooplankton abundance was also positively correlated with settling volume, displacement volume, wet weight and dry weight. The highest biomass value was observed in the northeast monsoon and pre-monsoon periods. The results of the canonical analysis of the output from the discriminate function was tested. Out of fifteen variables, only one was significantly different in single character ratios dry weight/ash free-dry weight （F3,23=4.78,P＆lt;0.005）. The mesozooplankton community was collectively composed of 28 taxa. Among these groups, copepoda （66.3%）, gastropod larvae （9.94%）, evadne （4.60%）, zoea （3.60%）, cypris nauplii （2.56%）, lemellibranch larvae （1.87%）, chaetognaths （1.81%）, ostracods （1.73%）, lucifer （1.15%） and barnacles nauplii （1.35%） contributed the most to the similarities within Sta. S1, while copepoda （74.68%）, cypris nauplii （5.29%）, gastropods （4.87%）, barnacles nauplii （4.81%）, evadne （1.72%）, zoea （1.53%） and ca-ridean larvae （1.18%） at Sta. S2. Sugar factories are supporting national economy as these are playing key role for development of socio-economic sectors of a country. The main product of Sugar industries is white sugar and by products are alcohol, ethanol, liquid carbon dioxide and household textiles. However, industries are generating effluents during cane crushing and distillery operation;which is hazardous, damaging and degrading the environmental eco-system. Sugar industries were established in the catchment area of Left bank outfall drain(LBOD) system. The untreated effluent generated from these Sugar mills is released into drainage system through network of LBOD system in lower part of Sindh province of Pakistan. The effluent generated is mostly organic having small quantum of in-organic material. Estimated wastes are Sulphur dioxide, hydrochloric acid, lead, fly-ash and dust, plant and workforce sewage, process waste which comprised of bagass, molasses, filter mud and grease oil from various sources of plant. The pollution level of effluents was evaluated against the National Environmental quality standard(NEQS) recommended level as well as with World Health Organization(WHO) level.

本文解决了从N个传感器组成的线性阵列中检测完全或部分缺陷传感器的问题。本文首先提出了一种线性阵列的对称结构,其次,基于结合差分进化的文化算法,建立了一种混合技术。对称结构具有两个优点:(1)不需要找到所有损坏的模式,仅需找到(N–1)/2个必需模式;(2)不需要扫描0°到180°区域,仅需扫描0°到90°区域。显然,这样可以减少计算的复杂度。通过Monte Carlo模拟对该方案性能进行了验证,并在计算时间和均方误差方面与现有方法进行了比较。
